Determination of the empirical formula of peptides by fast atom bombardment mass spectrometry.

A method for calculating the empirical formulae of peptides from mass spectrometric data is described. Exact mass measurement data and isotopic peak ratios are used to generate a list of potential empirical formulae that fit a given compound within experimental error. The formulae are then analysed by a mathematical algorithm and only those corresponding to chain peptides formed from the basic amino acids are retained. Calculations conducted for typical peptides indicate that the approach may be useful for peptide identification if the experimental values are determined within an acceptable range of errors. Experimental measurements of the exact mass and isotopic peak ratios made using typical peptides demonstrate the feasibility of the approach.

[Extracorporeal piezoelectric lithotripsy by ultra-short waves using the EDAP LT01 device].

Three hundred and sixty one extracorporeal lithotripsies for renal, ureteric and bladder stones have been performed by means of a system of ultrasonographic detection and piezoelectric destruction (EDAP LT01). The localisation of the stone is achieved by a 5 MHz real time sectorial transducer situated in the centre of a small dish containing 320 piezoelectric elements concentrated in a source 5 mm wide by 15 mm high. The pressure recorded in vitro is 900 bars. The stone is easily detected in 87.2% of cases, difficult to detect in 10% of cases and impossible to detect in 28% of cases. By using a frequency of 1.25 to 5 per second, extracorporeal lithotripsy can be performed without any local, regional or general anaesthesia and without premedication in the 120 patients with a renal stone, reviewed between 1 and 3 months, 88 (73%) were considered to be complete successes. Ten (8%) were considered to be failures and 19 (21%) were considered to be partial successes. The best results were obtained in stones of the renal pelvis less than 20 mm in diameter. These results relate to a mean series which must take into account the successive improvements made to the lithotripter. Today, stone fragmentation is obtained in 93% of cases. Thirty six stones of the lumber and pelvic ureter were treated with success rates of 93% and 50%, respectively. Six bladder calculi were treated with a 50% success rate. Forty two patients were treated without being admitted to hospital.(ABSTRACT TRUNCATED AT 250 WORDS)

[Prosthesis without puncture, 3d generation of vascular access for hemodialysis: a study of Hemasite in 10 patients].

Hemasite is a new vascular access for hemodialysis. The device has a carbon coated T-shaped body with a resealable silicon septum that obviates the need for skin punctures. It is implanted with attached PTFE grafts. Since August 1984, 10 Hemasite have been placed in 10 patients. After one year, 63% of Hemasite are still permeable: this success rate is at least similar to usual synthetic grafts. There is no infectious complication. The rate of thrombosis is high, but Hemasite offers the interesting facility to perform local infusion of thrombolytic drugs and/or painless angioplasty through its removable septum in case of venous stenosis. Patients and staff acceptance has been excellent.

Study of the local graft versus host reaction in the rat. I. Evidences for a donor-related "rat effect".

The local graft versus host reaction (GVHR) observed in the hybrid F1 LBN rat was chosen to evaluate the immunologic effect induced by allogeneic transfusion of Brown Norway blood to the Lewis rat. Study of the kinetics of this GVHR with immunized as well as with nonimmunized cells shows an "individual rat effect" which is shown by a variability of the popliteal lymph node weight from one recipient to another. When spleen cells from different Lewis donors are pooled this effect disappears which indicates that it is related to the donor's rather than to the F1 recipient's cells. To even out variation between individual donors, cells from a number of donors should be pooled to allow better interpretation of experimental protocols using this GVHR.

Primary nonseminomatous germ cell tumor of the prostate.

We describe a case of a primary nonseminomatous germ cell prostatic tumor and discuss the problem of extragonadal germ cell tumors. Prognosis, staging and management of these tumors are similar to that of metastatic primary testicular germ cell tumors.

[Lithotripsy by ultrashort pulsation. Experimental study in renal lithiasis in the dog].

A lithotriptor delivering ultrashort shockwaves and designed for use under ultrasonographic guidance has recently been developed. Immersion in water is unnecessary. The device was experimented in renal lithiasis in dogs. Twenty-two animals were treated. In seven, who served as controls, the safety of shots delivered in particular to the lungs, liver, bones and kidneys was demonstrated. Furthermore, no cardiac rhythm disorders were recorded. A human stone was implanted in 15 animals. The stone was successfully broken up in 12. Gross and microscopic examinations done 2 to 30 days after the treatment indicate that the shots initially produce lesions similar to a limited contusion. Healing of the contusion, already underway on the 8th day, is complete by the 4th week. Elimination of the fragments is gradual. The quality of fragmentation can be evaluated during the treatment session using mode A ultrasonography. These experimental data are evidence of the effectiveness of this second generation lithotriptor.

Light-Dependent Oxygen Uptake, Glycolate, and Ammonia Release in l-Methionine Sulfoximine-Treated Chlamydomonas.

Glycolate and ammonia excretion plus oxygen exchanges were measured in the light in l-methionine-dl-sulfoximine treated air-grown Chlamydomonas reinhardii. At saturating CO(2) (between 600 and 700 microliters per liter CO(2)) neither glycolate nor ammonia were excreted, whereas at the CO(2) compensation concentration (<10 microliters per liter CO(2)) treated algae excreted both glycolate and ammonia at rates of 37 and 59 nanomoles per minute per milligram chlorophyll, respectively. From the excretion values we calculate the amount of O(2) consumed through the glycolate pathway. The calculated value was not significantly different from the component of O(2) uptake sensitive to CO(2) obtained from the difference between O(2) uptake of the CO(2) compensation point and at saturating CO(2). This component was about 40% of stationary O(2) uptake measured at the CO(2) compensation point. From these data we conclude that glyoxylate decarboxylation in air-grown Chlamydomonas represents a minor pathway of metabolism even in conditions where amino donors are deficient and that processes other than glycolate pathway are responsible for the O(2) uptake insensitive to CO(2).

O(2) uptake in the light in chlamydomonas: evidence for persistent mitochondrial respiration.

The nature of the process responsible for the stationary O(2) uptake occurring in the light under saturating CO(2) concentration in Chlamydomonas reinhardii has been investigated. For this purpose, a mass spectrometer with a membrane inlet system was used to measure O(2) uptake and evolution in the algal suspension. First, we observed that the O(2) uptake rate was constant (about 0.5 micromoles of O(2) per milligram chlorophyll per minute) during a light to dark transition and was not affected by 3-(3,4-dichlorophenyl)-1,1-dimethylurea. Salicylhydroxamic acid had no effect on O(2) uptake in the dark or in the light, but was found to have the same inhibitory effect either in the dark or in the light when added to cyanide-treated algae. The stimulation of the O(2) uptake rate due to the uncoupling effect of carbonyl cyanide m-chlorophenylhydrazone was about the same in the dark or in the light. From these results, we conclude that mitochondrial respiration is maintained during illumination and therefore is not inhibited by high ATP levels. Another conclusion is that in conditions where photorespiration is absent, no other light-dependent O(2) uptake process occurs. If Mehler reactions are involved, in Chlamydomonas, under conditions where both photosynthetic carbon oxidation and reduction cycles cannot operate (as in cyanide-treated algae), their occurrence in photosynthesizing algae either under saturating CO(2) concentration or at the CO(2) compensation point appears very unlikely. The comparison with the situation previously reported in Scenedesmus (R. J. Radmer and B. Kok 1976 Plant Physiol 58: 336-340) suggests that different O(2) uptake processes might be present in these two algal species.

Biological tolerance to polydioxanone absorbable clips: a comparison with metallic ligating clips.

An experiment was conducted in 96 rats to determine the tissue tolerance to a hemostatic and absorbable clip made of polydioxanone, as compared to standard metallic clips. Clinical tolerance was similar and no bleeding or death occurred because of the material. Pathological studies showed a mild histiocytic reaction with absorbable clips and a stronger inflammatory reaction with metallic clips. This experiment indicates that absorbable clips are efficient and well tolerated and we strongly recommend the use of this material.

[What is the use of the radioimmunoassay of prostatic acid phosphatase?].

Radioimmunoassay (RIA) of prostatic acid phosphatases (PAP) using the Gammadab (R) PAP pack supplied by Clinical Assays in an effective method for determining extension of prostatic cancer. In patients with lymph node or metastatic invasion serum assay values are almost always elevated (sensitivity = 94%), especially with well differentiated cancer. Elevated levels of this variable are practically pathognomonic of external extension (specificity = 94%). This is of importance in therapy since the degree of invasion determines the choice between palliative and curative treatment. Strategy leading to this therapeutic choice can be greatly simplified and thus modified by RIA of PAP.

[Inferior vena cava thromboses in renal cancers. 20 years' experience].

The authors present a detailed review of a personal series with careful evaluation of the level of the caval thrombus as far as the right auricle, the degree of lymph node involvement, the surgical treatment and the duration of survival. In contrast with earlier rather optimistic papers, this study revealed a generally poor prognosis. The authors consider that when the cancer has invaded the vena cava, distant microscopic metastases are already present at the time of operation. In the absence of an effective chemotherapy, this type of surgery should only be performed in carefully selected cases (good general health, no signs of metastases or lymph node involvement). In cases with supradiaphragmatic thrombus, cardio-pulmonary by-pass, which is now a common procedure, appears to be safest technique.

Paraneoplastic hypercalcemia associated with bladder carcinoma: report of 2 cases.

Hypercalcemia associated with bladder carcinoma is rare. We report on 2 patients with hypercalcemia and bladder tumor without bone metastases. In both patients serum calcium returned to normal after removal of the tumor. Serum immunoreactive parathyroid hormone levels were normal in both patients. In 1 case a high gradient of immunoreactive parathyroid hormone across the tumor was demonstrated but the secretion of nephrogenous cyclic adenosine monophosphate was normal. Urinary excretion of prostaglandins also was normal in this case. These data show that hypercalcemia was caused by the tumoral production of some humoral factor different from parathyroid hormone.

[Experimental study of an absorbable hemostatic clip].

A new absorbable hemostatic clip was investigated by a study in the rat. The reliability and innocuity of the clip were similar to that of the metallic clips from the clinical point of view. Its absorbability and radiotransparency qualities make it the method of choice for ensuring hemostasis and lymphostasis when long-term follow-up investigations involve modern radiological techniques such as computed tomography.

[Bladder bilharzioma and carcinoma in a bilharzial bladder].

Over 7 years, 592 patients infected by Schistosoma haematobium were detected. Among these, 102 had a cystoscopy which found carcinoma in 3 patients, polypi in 1 and vesical "bilharzioma" (so-called pseudo-polyps) in 14. From the results of the biopsies and data from the medical literature the relationship between schistosomiasis and carcinoma in the schistosomial bladder is discussed, as well as the meaning of various lesions, such as malpighian metaplasia, papillomatous hyperplasia and glandular cystitis, seen in biopsy specimens from "bilharziomas". The cystoscopic diagnosis of one (or several) tumours resembling "bilharzioma" requires biopsy, resection and close monitoring.
