The signal recognition particle-targeting pathway does not necessarily deliver proteins to the sec-translocase in Escherichia coli.

ProW is an Escherichia coli inner membrane protein that consists of a 100-residue-long periplasmic N-terminal tail (N-tail) followed by seven closely spaced transmembrane segments. N-tail translocation presumably proceeds in a C-to-N-terminal direction and represents a poorly understood aspect of membrane protein biogenesis. Here, using an in vivo depletion approach, we show that N-tail translocation in a ProW derivative comprising the N-tail and the first transmembrane segment fused to the globular P2 domain of leader peptidase depends both on the bacterial signal recognition particle (SRP) and the Sec-translocase. Surprisingly, however, a deletion construct with only one transmembrane segment downstream of the N-tail can assemble properly even under severe depletion of SecE, a central component of the Sec-translocase, but not under SRP-depletion conditions. To our knowledge, this is the first demonstration that the SRP-targeting pathway does not necessarily deliver SRP-dependent inner membrane proteins to the Sec-translocase. The data further suggest that N-tail translocation can proceed in the absence of a functional Sec-translocase.

The crystal structure of cricket paralysis virus: the first view of a new virus family.

Numerous small, RNA-containing insect viruses are currently classified as picornaviruses, or as 'picorna-like', since they superficially resemble the true picornaviruses. Considerable evidence now suggests that several of these viruses are members of a distinct family. We have determined the gene sequence of the capsid proteins and the 2.4 A resolution crystal structure of the cricket paralysis virus. While the genome sequence indicates that the insect picorna-like viruses represent a distinct lineage compared to true picornaviruses, the capsid structure demonstrates that the two groups are related. These viral genomes are, thus, best viewed as composed of exchangeable modules that have recombined.

A suggested taxonomy and nomenclature for the cricket paralysis and Drosophila C virus complex.

Biophysical, biological, and serological characters are presented which in combination allow isolates of cricket paralysis virus (CrPV) and Drosophila C virus (DCV) to be separated and seven distinct strains of CrPV to be defined. Reference isolates for CrPV and DCV are suggested along with a system of nomenclature that allows the passage history of a particular isolate to be easily described.

Black beetle virus: propagation in Drosophila line 1 cells and an infection-resistant subline carrying endogenous black beetle virus-related particles.

Black beetle virus (BBV), one of a recently discovered class of viruses with a bipartite genome, multiplied readily in Schneider's line 1 of Drosophila cells. Virus yields, on the order of 100 mg per liter of culture, were unusually high and represented some 20% of the total cell protein within 3 days after infection. A derivative subline of these Drosophila cells was found to be resistant to infection by BBV. These resistant cells were also found to carry small amounts of BBV-related particles, possibly a maturation-defective form of BBV.

Functional evaluation of a corticosteroid molecule activity, dexamethasone-isonicotinate, by aerosol dosage in obstructive lung disease.

The authors experimented with a corticosteroid molecule, dexamethasone-isonicotinate, by pressurized aerosol, testing it on 15 subjects suffering from a bronchial obstructive syndrome (7 cases of asthmatic bronchitis, 6 of chronic bronchitis with pulmonary emphysema, 2 of bronchial asthma). In such patients clinical evolution, spirographic patterns, and airway resistance were studied by means of body plethysmograph before and after administration of 0.25 mg dexamethasone-isonicotinate four times a day. In four subjects daily variations of airway resistance before and after 10 days of treatment were evaluated: in three cases the resistance diminished noticeably at all times of the day after therapy. In eleven cases the functional parameters were recorded before and after 7 days therapy, and ten subjects were followed up by a monthly, check during 6 months of therapy. In both methods, the authors obtained good or very good results in more than half of the subjects and fair results in a fifth of the subjects; in the remaining patients poor results were obtained. Side-effects attributed to improper dosage were never observed.

[Lung ductances and the determination of lung diffusion capacity in silicosis].

A study of the behaviour of global and partial lung ductances for CO having for aim to value the lung diffusion function in 97 silicotic workers was undertaken by authors. The workers were divided into three groups according to the Cincinnati classification. The mean values of the global ductance for CO let appear a fall in all the groups when comparison is made with the standard values previously registered in a group of normal subjects. The partial ductances have put in evidence a fall in the mean values below the normal values, only for what concerns the alveolar-capillar ductance for groups 2 and 3. The study of partial ductances in bronchitic and non-bronchitic silicotic subjects lets appear in the first ones a fall in the ventilatory and alveolar-capillar ductances,-- and in the second ones an isolated fall in the alveolar-capillar ductance.
