Vascular structural changes in the artery wall of unilateral nephrectomized rats.

Wet and dry weight, concentration and total amount of nucleic acids and proteins, and incorporation of tritiated thymidine were measured in the arteries of rats after 7 and 15 days of unilateral nephrectomy, sham operation and in control animals. Arteries of uninephrectomized rats showed increased wet and dry weight, increased DNA and protein content after 7 and 15 days and a higher rate of tritiated thymidine uptake after the first week. These results suggest an early proliferative process in the arteries of unilateral nephrectomized rats.

Angiotensin--forming enzyme active at the physiological Ph in the brain of normal and nephrectomized rats.

Enzymatic activity generating angiotensin at pH 5.5 and 7.2 has been detected in different areas of the central nervous system (CNS) of the rat. Control animals and those subjected to bilateral nephrectomy 48 h before the experiment (Nx) were analyzed. The different areas of the CNS were studied by the incubation of tissue homogenates in the presence (enzyme concentration) or not (enzyme activity) of an excess of added angiotensinogen. Concentration was determined by incubation at pH 7.2 and 5.5 while activity was evaluated only at pH 7.2. The enzymatic renin-like concentration at both pHs did not change after Nx thus showing they do not depend on plasma and vascular renin. On the other hand the activity of the enzyme showed a significant increase in the cerebral cortex and cerebellum after Nx suggesting an increased concentration of renin substrate and/or different concentrations of inhibitors or activators of the enzymatic system in those areas.

Evidence of early structural change in the artery wall of two-kidney one-clip Goldblatt hypertensive rats.

Vascular structural changes were studied during the development of two-kidney one-clip renal hypertension. The weight of the arteries and the concentration and total amount of ribonucleic acid, deoxyribonucleic acid, alkali-soluble proteins, collagen and elastin of the vascular wall were measured. Tritiated thymidine uptake was also determined 15 and 30 days after clipping. Hypertension developed in 58% of the animals while the rest remained normotensive. A significant increase in artery weight and in the total amount of nucleic acids and proteins was found in hypertensive rats. The uptake of 3H thymidine by the arteries of hypertensive rats was significantly increased 15 days after clipping. This increment showed a significant correlation with blood pressure levels. Present data seem to indicate that the increase in vessel wall dimensions observed is partly due to an increase in the number of smooth muscle cells during the acute phase; this alteration appears to be mainly due to the rise in blood pressure.

Role of monocytes, alloantigens, and individual reactivity in the response of human lymphocytes to mitogenic factor.

The activity of mitogenic factors generated in human mixed lymphocyte cultures (MLC-MF) was analyzed with unstimulated mononuclear leukocytes used as responder cells. Unseparated or adherent, but not nonadherent, irradiated, autologous mononuclear leukocytes were found to significantly enhance the mitogenic activity of MLC-MF. The effect of MLC-MF was not inhibited by hydrocortisone at concentrations that inhibited MLC-MF production, indicating that the MLC-MF effect is direct and is not mediated through further production of mitogenic factors. Experiments in which MLC-MF was produced in a MLC between a HLA-D homozygous individual and her child suggested that soluble alloantigens do not play a role in the MLC-MF effect. Studies in one individual with MLC responses higher than other individuals indicated average MLC-MF production but above average response to MLC-MF. We conclude that MLC-MF is directly mitogenic for unstimulated lymphocytes, without requiring the simultaneously presence of either mitogen or antigen. In addition, the proliferative MLC reaction of antigen. In addition, the proliferative MLC reaction of individuals with strong responses to MLC-MF may be influenced significantly by the nonspecific clones recruited by MLC-MF.

Changes in future time perception of day hospital psychiatric patients in response to small group treatment approaches.

Attempted to assess the effects of small group approaches on future time perception of psychiatric patients in a day hospital. The research employed two patient groups within the day hospital: A control group, (N = 12) which received routine day hospital treatment, and an experimental group, (N = 12) which received, in addition, the small group treatment. Measures of future time perception were administered to both groups before and after a 12-week treatment. Results indicated significant positive changes in response to group treatment on Future Events and the dimension of Importance. However, no positive significant differences were found on Time Extension, Directionality, and Quality of Affect. While it is not within the scope of this research to evaluate whether the secured change can be maintained, it was clear that in immediate posttesting important changes had resulted from the treatment strategy.

Rehabilitation of the Puerto Rican addict: a cultural perspective.

Drug addiction has assumed major proportions in the Hispanic population. This paper questions whether treatment programs in Puerto Rican communities adequately relate their rehabilitative services to the realities of Hispanic culture. The experience of a clinic in the South Bronx is reviewed, and it is suggested that programs need to be aware of cultural differences, provide Hispanic staff to treat Hispanic patients, and build up acknowledged strengths, values, and folkways.

Methadone treatment and physical complaints: a clinical analysis.

In this study 102 male patients on the Harlem Hospital Methadone Maintenance Treatment Program were asked about physical complaints which they attributed to taking methadone. The most common complaints were sweating, constipation, drowsiness, sexual problems, and aches in bones and joints. There were no statistically significant differences between new patients and long-term patients, but long-term patients appear more likely to be bothered by sweating than new patients, and constipation occurs most frequently during the initial stages of treatment. Complaints were found, in general, to be minor and did not constitute a barrier to patient retention in treatment.

Multidisciplinary team malfunctioning on a state hospital unit: a case study.

The use of multidisciplinary teams in the care of psychiatric patients can be countertherapeutic unless attention is paid to the dynamics of team functioning. The authors present a case study of team malfunctioning on an inpatient unit in a state hospital that resulted from staff's role confusion and insecurity. Patient care was relegated to second place as major interpersonal conflicts among the staff were played out along ethnic and cultural lines. Resolution of the conflicts required identifying their source, clarifying staff roles, and initiating a special inservice training program focused primarily on the needs of paraprofessional staff.

Tardive dyskinesia and institutional practice: current issues and guidelines.

Despite increasing public and legal pressures to minimize the occurrence of tardive dyskinesia, the practicing clinician often feels there are no alternatives to the use of neuroleptic medication in the treatment of psychosis. However, there are many clinical situations in which the use of neuroleptics may be avoided; guidelines for such situations, including the treatment of affective disorders and of chronic schizophrenia, are presented. The authors also discuss the use of alternative treatment programs and psychosocial interventions that may allow significant reduction in neuroleptics, and they describe new areas of research aimed at preventing and treating tardive dyskinesia.

The fiscal crisis in New York City: effects on the mental health care of minority populations.

The author describes the effect that severe budget cuts accompanied by guidelines stipulating that direct services be given priority over primary preventive services had on a community mental health center in New York City. He focuses his discussion on the effect these guidelines had on the mental health care services provided the minority populations served by the center. Before the budget cuts, 1 out of every 4 staff members was a nonprofessional indigenous worker. After the cutback, 2 out of every 3 of the people laid off were nonprofessionals. The author provides a number of recommendations to help government agencies plan budget reductions without sacrificing primary prevention efforts.

Suppression of cell-mediated tumor cell lysis and complement-induced cytotoxicity by trypan blue.

Different forms of cell-mediated cytotoxicity were suppressed in the presence of trypan blue. The systems affected included lysis of antibody-coated tumor cells by normal and C. parvum-stimulated mouse peritoneal cells and lysis of allogeneic targets by immune effector cells. The inhibition, measured in a 4-hr 51Cr release assay, was reversible and did not occur in the presence of 30% fetal calf serum or albumin. Binding between effector and target cells through Fc receptors was not affected, and lysis of allogeneic cells was inhibited at the lytic step rather than at the binding step. In contrast, lysis of sensitized erythrocytes was not inhibited by trypan blue, suggesting that lysis of these targets may not involve the steps required in tumor cell lysis. Trypan blue blocked the function of antibody before binding to target cells and also suppressed complement-induced cytolysis. Most individual complement components were susceptible to the inhibitory action of trypan blue. These results reveal an affinity of trypan blue for proteins in general that may be responsible for many of its biologic actions.

Cultural factors in the training of psychiatric residents in an Hispanic urban community.

This paper describes a resident's experience with his first Puerto Rican patient who practiced spiritualism, also elaborating on the major problems encountered, in so far as training for psychiatric residents is concerned. Furthermore, an attempt is made to analyze the role that residency training programs should play in preparing the student psychiatrists to assume a more sophisticated role when confronted with such patients, who are so common in the urban ghettoes of the United States.

Establishment and characterization of a cell line derived from a spontaneous murine lung carcinoma (M109).

The Madison lung (M109) tumor cell line, initiated from a "spontaneous", anaplastic murine lung carcinoma, has been propagated continuously in vitro for more than 300 cell generations. Cytogenetic analysis revealed a mouse karyotype with a mode of 78 chromosomes (2n = 40). Three distinct marker chromosomes were identified by trypsin-giemsa banding. The cells piled up in culture and had a short generation time and high plating efficiency. Electron microscopy revealed highly undifferentiated cells with little rough endoplasmic reticulum, an abundance of free polysomes, the presence of few and often odd-shaped mitochondria, lipid bodies and phagocytic vacuoles. Virus particles of the C-type were found frequently. The subcutaneous transplantation of M109 cultured cells at a relatively low cell inoculum produced highly metastatic tumors in syngeneic BALG/c mice.
