[Rise in neutrophils after hydrocortisone seen as bone marrow reserve mass. Dependency on spleen size].

In 25 patients with a wide range of postmitotic marrow neutrophil mass a linear relationship was observed between the number of postmitotic marrow neutrophils and the rise of blood neutrophils after intravenous injection of 3 mg hydrocortisone/kg. Related to marrow neutrophil cellularity, the response curve was significantly higher in 10 splenectomized patients but greatly depressed in 10 patients with splenomegaly.

[Protein hydrolysis by immobilized enzymes].

The enzymic hydrolysis of some proteins (insulin-B-chain-S-sulfonate, S-aminoethylated lysozyme, bovine serum albumin) by immobilized peptidolytic enzymes is reported. Sepharose-bound pronase, trypsin and a protease from Thermoactinomyces sp. (MP), the latter both cross linked by glutaric dialdehyde and an exopeptidase mixture containing Sepharose-bound leucine aminopeptidase, carboxypeptidase A and a crude preparation of prolidase were used. After enzymic hydrolysis nearly all amino acids, except proline, were recovered in a 100% yield compared to the value of an acid reference hydrolysate. Tryptophan and methionine, which are partially destroyed by acid hydrolysis in the presence of oxygen could be recovered completely.

[Neutropenia in disorders of the neutrophil motility].

A 51-year-old patient exhibited severe neutropenia over a period of 12 months following lobectomy for carcinoma of the lung. Marrow neutrophil mass and marrow neutrophil production were measured quantitatively and were within normal limits. Mobilization into a skin window was absent. A defect in neutrophil motility could be demonstrated in vitro and was confirmed in vivo by poor neutrophil release from the marrow following injection of hydrocortisone; it thus represented a lazy leukocyte syndrome of the adult.

[Familial autoimmune hemolytic animia (AIHA) with negative Coombs test, lymphocytopenia and hypogammaglobulinemia].

A family with several cases of autoimmune hemolytic anemia and dysgammaglobulinemia is described. Two sibling had documented antierythrocyte antibodies, but a third had repeatedly negative Coombs tests. Diminished immunoglobulin-producing cells and a hypogammaglobulinemia were found, but no functional defect of humoral or cell-mediated immunity could be detected.

Application of ICP-MS for the assessment of thorium excretion in urine.

A method for rapid and sensitive determination of thorium in urine by inductively coupled plasma mass spectometry (ICP-MS) is described. The method is sufficiently sensitive to detect 1 ng/L 232Th in urine without any sample preparation. The mean urinary 232Th excretion in 23 unexposed subjects was 6.2 +/- 3.3 ng/d.

Studies on electrolyte and trace metal metabolism applying stable isotopes as tracers.

Tracer-kinetic investigations provide a useful tool for the assessment of body composition regarding electrolytes and trace elements. This paper demonstrates the feasibility of stable isotopes as tracers in studies on calcium and iron metabolism in risk groups like children. By applying both radioisotopes and stable isotopes simultaneously, in adults, a good intra-individual agreement was obtained for the derived figures of intestinal absorption. Using only stable isotopes, intestinal calcium absorption in children with renal failure was found to follow a similar pattern to that in adult patients.

Calcium kinetic investigations and their clinical use.

Pool sizes and internal fluxes of minerals can be quantitatively determined by tracer-kinetic investigations and compartmental analysis. Investigations performed in 252 patients revealed characteristic patterns for calcium pools and fluxes with respect to disease. The method provides a useful diagnostic tool in the early detection of metabolic bone disorders and in problems of differential diagnostics.

Fluorescent neoglycoproteins: antibody-aminodextran-phycobiliprotein conjugates.

New, highly amino-substituted dextran or aminodextran (hereafter denoted Amdex) of various sizes between about 20 and 1000 kDa molecular mass and degrees of amino-substitution between 7 and 40% were prepared and characterized by elemental analyses and polyacrylamide gel electrophoresis. These aminodextrans together with others commercially available were shown by static light scattering, viscosity, and refractive index measurements to adopt a globular structure in aqueous salt solutions. Antibody and fluorescent protein dye, phycoerythrin, or its tandems with cyanin 5. 1 and TEXAS RED, were covalently conjugated to the aminodextrans. The conjugates contained multiple dye molecules and were shown by dynamic light scattering and scanning electron microscopy to assume either globular structure or aggregates thereof. Streptavidin could be substituted for antibody to prepare streptavidin-aminodextran-PE conjugates, which were then used with biotinylated antibody to label subpopulations of white blood cells. The conjugates yielded up to 20-fold amplification of fluorescence intensity over direct antibody-dye conjugates in labeling white blood cells for flow cytometry.
