[Poisoning due to tricyclic antidepressants in children and adolescents].

During the last decade there has been a substantial increase in the number of poisonings due to tricyclic antidepressants in both children and adults. The main toxic manifestations are of anticholinergic origin, and the most frequent constitute the so-called central anticholinergic syndrome. Most of the deaths result from the cardiac complications. Three cases are presented that illustrate chiefly the toxic manifestations related to the central nervous system. The therapeutic management is reviewed in the light of the metabolism of the tricyclic antidepressants, their pharmacologic action and the mechanism of their toxic effects.

Pyruvate dehydrogenase, lipoamide dehydrogenase and citrate synthase activity in fibroblasts from patients with Friedreich's and Charlevoix-Saguenay ataxia.

The activity of lipoamide dehydrogenase and two closely related enzymes was studied simultaneously in early, mild, and late passage fibroblast cultures. Friedreich's ataxia fibroblasts tended to lose pyruvate dehydrogenase and citrate synthase activities, while lipoamide dehydrogenase activity remained constant with aging of the cells. Mean pyruvate dehydrogenase activity was lower over-all in fibroblasts from ataxics. Mean citrate synthase activity was higher in ataxic fibroblasts. Present tissue culture media do not represent the best conditions in which to reproduce cofactor binding defects such as those found in other genetic diseases with structural enzyme mutations.

[Structure of chromogen I of the Morgan-Elson reaction].

Alkaline treatment of 2-acetamido-2-deoxy-3,4:5,6-di-O-isopropylidene-aldehydo-D-glucose, or of the analogous D-mannose derivative, gave the same crystalline material, unambiguously identified as 2-acetamido-2,3-dideoxy-alpha-D-erythro-hex-2-enofuranose (3) by n.m.r. and mass spectrometry, and crystallography. Treatment of 3 with methanol in acidic medium gave methyl 2-acetamido-2,3-dideoxy-alpha,beta-D-erythro-hex-2-enofuranoside (11). The identical compound was obtained from Chromogen I under identical conditions.
