[Reactivity in vitro of the leukocytes of the earthworm Eisenia foetida Sav. to several mitogenic substances].

Earthworm leukocytes were stimulated by various mitogens including LPS, PHA and Con A. Con A could stimulate leukocytes when they were cultivated in totality. After separation into sub-populations, small-non-adherent-leukocytes and large-adherent ones showed specific reactions with mitogens. Both PHA and LPS reacted with small leukocytes but LPS had no action on large adherent leukocytes.

Evidence of depolarization-induced cAMP increase in the superior cervical ganglion of several mammalian species.

1. The activation of cAMP synthesis resulting from exposure of superior cervical ganglion (SCG) tissue to high K+ solutions observed in calf has been found to occur in three other mammalian species, namely rat, cat, and rabbit. 2. The cAMP accumulation in the ganglion of calf is much less pronounced than that occurring in the other species. The enhancing effect oftheophylline on the increase of cAMP was found to be much greater in the SCG of calf than in that of other species. When veratridine was used as the depolarizing agent the species differences were found to be similar to those observed after potassium stimulation. 3. The effectiveness of several other depolarizing agents in causing an increase in the cAMP level was tested in rabbit SCG.

The agglutinating and immunofluorescent activities of antileptospiral antibodies of human sera and of immunoglobulins M and G.

Microagglutinating and immunofluorescent activities of sera of nine patients with clinically diagnosed leptospirosis were investigated. L. grippotyphosa was found in seven patients, L. sejroe in one and L. icterohaemorrhagiae in one patient. The agglutinating activities of IgM approximated the activities of the whole sera. In five patients the agglutinating activities were found in IgG; in four of them two to three months after the onset of the disease, in one patient a month after its onset. In one patient only the activity of IgG reached the activity of IgM after six months. Immunofluorescent activity was found in IgG in eight patients two to five weeks after the onset of the disease. In seven of these patients the activity of IgG reached or exceeded the activity of IgM three to ten weeks after the onset of the disease. IFAT was found to be poorly specific because of high heterologous titres. In 5 out of 7 patients with L. grippotyphosa, heterologous IFAT titres were reduced or eliminated by carrying out the reaction with isolated IgM and IgG. However, in patients with L. icterohaemorrhagiae and with L. sejroe heterologous IFAT activity persisted in both IgM and IgG. The study points out the possibility of differentiating an early stage of the infection from convalescence.

Effect of depolarizing agents on the adenosine-3',5'-monophosphate content of the bovine superior cervical ganglion.

The cAMP content of the bovine superior cervical ganglion increases rapidly when the tissue is incubated in media containing high concentrations of potassium and theophylline. This effect of high K+ is mimicked by Rb+, Cs+ and NH4+. The depolarizing substances veratridine and ouabain also cause an increase in the concentration of ganglionic cAMP; the effect of ouabain develops more slowly than that of veratridine. Tetracaine, in high concentrations, reduces the response to potassium, whereas it reduces the response to veratridine at much lower concentrations. Tetrodotoxin, at a concentration that barely affects the response to potassium, prevents the response to veratridine.
