Preclinical activity of 17 beta-[N-[N'-(2-chloroethyl)-N'-nitrosocarbamoyl]-L-alanyl]-5 alpha-dihydrotestosterone (E91) against tumour colony forming units and haematopoietic progenitor cells.

E91 (17 beta-[N-[N'-(2-chloroethyl)-N'-nitrosocarbamoyl]-L-alanyl]-5 alpha-dihydrotestosterone) (CNC-ala-DHT) is a newly synthesised alkylating compound consisting of N-[N'-(2-chloroethyl)-N'-nitrosocarbamoyl]-L-alanine (CNC-ala) as the alkylating moiety and of 5 alpha-dihydrotestosterone (DHT) as a steroid carrier molecule. We studied the antitumour activity of E91 (final concentrations: 0.1, 1, 10 and 30 mumol/l) against freshly explanted human tumours, using an in vitro soft agar cloning system. A total of 54 tumour samples was evaluated using 1 h-exposure and 51 tumour specimens were studied using a continuous exposure for 21-28 days. In addition, the compound's activity was compared with other clinically used anticancer agents. After short-term exposure, 49 of 53 evaluable specimens (92%) had adequate colony formation, as compared with 49 of 50 (98%) after long-term exposure. After short-term exposure, E91 exhibited only marginal antitumour activity. However, in long-term exposure experiments, E91 had marked and concentration-dependent antitumour activity (P < 0.001). At concentrations of > 10 mumol/l, E91 was as active as the other clinically used antineoplastic agents and at 30 mumol/l, E91 was significantly more active than 5-fluorouracil (P = 0.041). E91 showed activity against a wide spectrum of tumour types. The highest activity was observed against colorectal carcinomas (3/4 tumour specimens inhibited at 30 mumol/l). Sensitivity was also high remarkable in breast cancer specimens with 3/6 specimens inhibited at 30 mumol/l. In vitro myelotoxicity was less than that of doxorubicin. At 30 mumol/l, E91 induced a reduction of colony forming units-granulocyte macrophage (CFU-GM) to only 53% of control and of CFU-GEMM to 20% of control. We conclude that because of broad activity and reduced myelotoxicity further clinical development of E91 appears warranted.

Automatic fixation facility for plant seedlings in the TEXUS Sounding Rocket Programme.

Automatic chemical fixation of plant seedlings within a 6 min period of reduced gravity (10(-4)g) was performed on three ballistic rocket flights provided by the German Sounding Rocket Programme TEXUS (Technologische Experimente unter Schwerelosigkeit = Technological Experiments in Microgravity). The described TEXUS experiment module consists of a standard experiment housing with batteries, cooling and heating systems, timer, and a data recording unit. Typically, 60 min before launch an experiment plug-in unit containing chambers with the plant material, the fixation system, and the temperature sensors is installed into the module which is already integrated in the payload section of the sounding rocket (late access). During the ballistic flight plant chambers are rapidly filled at pre-selected instants to preserve the cell structure of gravity sensing cells. After landing the plant material is processed for transmission electron microscopy. Up to now three experiments were successfully performed with cress roots (Lepidium sativum L.). Detailed improvements resulted in an automatic fixation facility which in principle can be used in unmanned missions.

Water and arteriosclerosis.

Consumption of hard water is presumed to protect against the development of arteriosclerotic diseases. Substantial epidemiologic correlations support the "hard water effect" as sufficiently valid to warrant alteration of public water supplies as a public health measure. A recently proposed theory that arteriosclerosis is an indolent, chronic, infectious disease caused by cyanobacteria suggests that an alternate explanation may be possible. The relationship between hard water and arteriosclerosis is reviewed, contradictions and exceptions noted, and an alternate theory of microbial infection is offered.

Arteriosclerosis in East Asians.

Arteriosclerosis is remarkably common in East Asians, both in the greater subcontinent and in their adopted countries after migration. Since arteriosclerosis is an acquired disease, East Asians must possess and preserve a common cultural practice related to the cause of arteriosclerosis and its complications. Therefore, an unusual opportunity is provided to examine East Asian migration and customs, especially diet, searching for a clue to the etiology of this disease. The result of this analysis provides additional support for a new theory proposing arteriosclerosis as an infectious disease caused by Cyanobacteria.

Milk and arteriosclerosis.

Milk consumption is related to arteriosclerosis. Recent landmark studies confirm a previously suspected close correlation between milk intake and arteriosclerotic heart disease. Support is therefore provided for a recently proposed novel hypothesis that arteriosclerosis is a chronic infectious disease caused by blue-green bacteria and that milk is a carrier vehicle for these contaminant organisms. A revisionist view of diet and milk in the causation of arteriosclerosis is developed. Previous hypotheses relating milk consumption to arteriosclerosis and advances in pasteurization techniques are discussed and integrated with this infection theory.

A possible cause of arteriosclerosis.

A new etiology of arteriosclerosis is proposed. This theory has emerged from the inability of current theories to account for the spontaneously occurring disease and from numerous factual anomalies uncovered by recent research. It is suggested that arteriosclerosis is a chronic infectious disease caused by an infestation of blue-green algae, and that the natural history, histopathologic changes, and many apparently contradictory facts associated with arteriosclerosis are explained by such an etiologic agent. If correct, a radical shift in our understanding of the disease is required. Certain observations and forecasts are made based upon this infection theory.
