Determination of thiamine and thiamine phosphates in excitable tissues as thiochrome derivatives by reversed-phase high-performance liquid chromatography on octadecyl silica.

The analysis of thiamine and thiamine phosphates by high-performance liquid chromatography owes its high sensitivity to the fluorescent derivatives or thiochromes obtained by chemical oxidation in alkaline medium. The possibility of performing precolumn oxidation with potassium ferricyanide instead of using the hazardous cyanogen bromide has been investigated. The derivatization step has been optimized with respect to the following parameters: concentration of alkali and oxidant, presence of methanol and stability of the thiochromes . A gradient separation with 25 mM phosphate buffer (pH 8.4) and methanol as mobile phase components and an octadecyl silica column as stationary phase has been set up. The analytical run takes 14 min with the following elution order: thiochrome triphosphate, thiochrome pyrophosphate, thiochrome monophosphate and thiochrome. The minimum detectable amount is 0.05 pmol. The method was found suitable for the determination of thiamine compounds in excitable tissues such as nerves and electric organs as well as in proteins extracted from membranes of these organs. It may be useful to study the role of thiamine in the electrical activity of these tissues at the molecular level.

[Swiss continuing education meetings for school physicians. An evaluation of preferences for selected subjects].

The group "school health physicians" of the Swiss Society for Social- and Preventive Medicine has organized since its foundation in 1981 two meetings; Basle in 1982, and Zurich in 1983. Questionnaires were handed out on both occasions on source and extent of experience as school health doctors. The questions included views on the meetings in general and on the various topics discussed. Participants were also asked to write down suggestions for future meetings. The following subjects discussed were voted to be "important" or "very important" by at least two thirds of the participants: ophtalmologic mass examinations; health education as an important new factor for school health physicians as evidenced by the anti-smoke-campaign in Baselstadt schools; round-table-discussion on the currant importance of school-health medicine; certification of fitness for schooling by the school doctor; growth and development of the child on startin school; psychomotor disorders and their effects on children starting school. The third meeting, scheduled for September 27th 1984 in Lausanne, which will include for the first time a morning session, will deal with topics selected by the participants of the Basle and Zurich meetings: orthopedic problems at school age and their prevention; a report on school sports and high-performance sports.

[Cystic abdomino-mediastinal lymphangioma. Apropos of a case. Review of the literature].

A further case of cystic lymphangioma is reported, characteristic features being the multiple abdominal and thoracic localizations of the lesions. Apart form splenic sites, it is rare to detect lesions in the mesogastric and mediastinal regions, particularly as propagation of abdominal lymphangioma towards the mediastinum is an exceptional occurrence. A literature review provides data on the various regions in which cysts have been reported in the body, and the diagnostic problems they raise.

[Association of streptococcal spondylitis and endocarditis. 2 new cases].

Two new cases of spondylitis associated with endocarditis are reported. Approximately 30 cases have already been published in the medical literature. Although this association is infrequent, the possibility of its occurrence calls for repeated auscultation in patients with spondylitis and focal roentgenograms in patient with bacterial endocarditis who develop vertebral pain.

[School medicine: dynamic aspects of a discipline].

School health is evolving according to new knowledge, variations of pathology and epidemiology, scattering of responsabilities as to diagnosis and treatment, progressive coverage of school career, and diverse professional training of the physicians involved. As part of social and preventive medicine does it still correspond to a common denominator? If we pretend so we give the school physician "sensu stricto" a primordial position. But then we ought to formulate new directions inspired by the light of permanent reflexion what youth are expecting from the physician in their situation of students. Today pupil adaptation to school and adaptation of the latter to our young pupils ranks first, with a specific medico-social and pedagogical assistance focused on primary prevention. Therefore collective awareness of school physicians under the auspices of the Swiss Society of Social and Preventive Medicine is welcome.

Peripheral lymphocytes changes in the anamnestic response to tetanus toxoid challenge.

Few data are available on the blood lymphocyte response to revaccination in man. The anamnestic response to tetanus toxoid challenge was evaluated by a variety of techniques during the first week after revaccination. Out of twenty subjects used, eight were evaluated before and 5 days after the injections (days 1--8). Analytical cell electrophoresis showed important variation in the B and two T lymphocyte populations. The B cell percentages, assessed by EAC-rosettes and electrophoretic mobility, were found to decrease by days 2 and 3, and return to former levels by day 8, when a rise in specific antibodies was detected. A similar response was found in the T1 population generally considered to be composed of low affinity E-rosette-forming cells. Conversely, a significant increase (50--100%) in circulating T2 lymphocytes (active rosette-forming cells) was found by days 2 and 3, followed by a rapid decrease of these 'differenciated' cells. The increase in the T2 lymphocytes appeared earlier in skin test positive subjects. These changes were correlated with E-rosettes, mitogen stimulation, peripheral leucocyte migration inhibition and transformation in the presence of the antigen. EA-IgG rosettes and ADCC varied similarly. These results may indicate a significant non-specific cell mobilization following revaccination.

Familial correlations of longevity: an isolate-based study.

Familial correlations for age at time of death have been computed in a French Canadian isolate. We show that parent-offspring correlations as well as sib correlations are of the same order of magnitude as that between spouses for various age groups at death. It is suggested that heritability of survival is nearly zero. Observed variability in survival is interpreted as the effect of environmental differences acting upon age-dependent genes.
