Prostaglandin F2alpha as a mediator of pulmonary changes during platelet aggregation.

Increases in pulmonary arterial pressure, tracheal insufflation pressure, and blood levels of the prostaglandin F2alpha metabolite, 15-keto-13, 14-dihydro F2alpha, were observed after protamine chloride or thrombin-induced platelet aggregation and release reaction in dogs. These effects were largely eliminated after administration of acetylsalicylic acid, an inhibitor of prostaglandin synthesis. The platelet aggregation was not noticeably affected. It is suggested that release of prostaglandin F2alpha from platelets is an important factor for the pulmonary changes during induced platelet aggregation. The necessity of measuring metabolite levels, rather than prostaglandin F2alpha levels, in blood during in vivo conditions is demonstrated.

Effects of local thrombinaemia and endotoxinaemia on hepatic circulation in the anaesthetized dog.

The acute response of the hepatic circulation to thrombinaemia and endotoxinaemia were investigated in the anaesthetized dog. Small amounts of thrombin were infused into the hepatic artery and portal vein. The effects of thrombin were compared to those of minor amounts of portally infused endotoxin. Portal pressure increased significantly following portal infusion of thrombin and endotoxin. Both substances decreased the blood flow markedly in the hepatic artery, while that in the superior mesenteric artery and portal vein decreased only slightly. Heparin and polyphlorethin phosphate (PPP) abolished all effects of thrombin. Bradykinin infused simultaneously inhibited only the effect on the hepatic artery. Vasoactive intestinal peptide, which has the same vasodilatory effect in the liver, did not. It is believed that thrombin and perhaps endotoxin exert a vasoconstrictive effect mediated by products released from platelets.

Interference on the haemostasis in dogs with induced coagulopathies by drugs affecting the platelet function.

Acetylsalicylic acid and phenylbutazone increased the bleeding significantly from standardized wounds in dogs defibrinogenated with Defibrase. Administration of dipyridamole and Xylocain had no effect. The results were the same in dogs treated with warfarin sodium. The results demonstrate the value of using laboratory animals with an induced coagulopathy when establishing the effect on the haemostasis exerted by drugs known in vitro to interfere with platelet functions. Acetylsalicylic acid and dipyridamole did not alter the activity of the K-vitamin-dependent coagulation factors, factors V, VIII, platelet count or fibrinogen concentration.

Patch grafts or tubular grafts in the repair of coarctation of the aorta. A follow-up study.

The occurrence of aneurysms after repair of aortic coarctation with patch graft and tubular graft techniques was studied. In the patch graft group, 3 out of 6 re-investigated patients developed aneurysms; 2 of them after several years. It is suggested that the causes of aneurysms originate from mechanical factors. In the tubular graft group, 2 out of 18 patients developed aneurysms. In this group, the aneurysms seemed to be due to infection.
