Identification of viral structural polypeptides of Thogoto virus (a tick-borne orthomyxo-like virus) and functions associated with the glycoprotein.

Thogoto (THO) virus is a tick-borne virus which shares morphological and genetic features with members of the Orthomyxoviridae family although the viral glycoprotein appears to be related to gp64 of baculoviruses. Characterization of THO virus was undertaken to clarify its taxonomic position. Purified virus preparations contained at least six virus-encoded polypeptides with apparent M(r) values ranging from 29K to 92K. A 75K polypeptide was identified as an envelope-associated glycoprotein by Triton X-100 and salt dissociation studies, and by proteolytic degradation of the exposed proteins of the virion. By the same criteria, the nucleoprotein and the matrix protein were identified as the 52K and 29K polypeptides, respectively. Immunofluorescence studies using monoclonal antibodies (MAbs) located the glycoprotein on the external cell membrane and the nucleoprotein in the nucleus of infected cells indicating that virus replication involved a nuclear phase. In addition, the virus displayed haemagglutination and haemolytic activities with an optimum at pH 6. These activities are functions of the viral glycoprotein since they were inhibited by anti-glycoprotein MAbs. The data reported here support the notion that THO virus is a member of the Orthomyxoviridae family but that it should be classified in a group distinct from the other influenza viruses.

Technetium 99m-labelled-sucralphate isotope scanning in the detection of peptic ulceration.

An isotope scan using sucralphate labelled with technetium-99m has been compared with barium meal in the detection of peptic ulceration in 18 patients with endoscopically proven upper gastrointestinal lesions and eight controls. Gastric (75%) and duodenal (67%) ulcers were detected with identical frequency by isotope scan and barium meal but both were inferior to endoscopy. Structural, non-ulcerated lesions, were not identified by isotope scanning. The scan has potential value in duodenal ulcer disease, the serial assessment of ulcer healing and in paediatric gastroenterology.

Cystic fibrosis: adolescent and maternal concerns about hospital and home care.

The purpose of this study was to describe problems occurring in adolescents with cystic fibrosis that related to home or hospital care, maternal concerns, and the role of the health-care professional in promoting care. Forty-question semistructured interviews of adolescents (N = 20) and their mothers (N = 12) were used. Interrater reliability (90%) was established through analysis of 8 audiotaped interviews and agreement on content analysis of the 32 interviews. Results are discussed according to five categories of concerns identified by adolescents: (a) knowledge deficits, (b) self-care/decision making, (c) home care management, (d) hospital care management, and (e) suggestions for health professionals. Mothers described six categories of concern: (a) fear, (b) effect on self, (c) disruption of family life, (d) health-care management at home, (e) health-care management in the hospital, and (f) suggestions for health professionals.

Adolescent

Maternal responses to home apnea monitoring of infants.

The purpose of this study was to describe types of upset experienced by mothers of infants who received apnea monitoring for 1 to 17 months. A 38-question, semistructured interview was used. Interrater reliability (90%) was established through analysis of 15 videotaped interviews and agreement on content analysis of 74 transcribed interviews. Results are discussed according to nine categories of upset identified by the mothers: (a) fear, (b) lack of credibility, (c) problems with the monitor, (d) disrupted family life, (e) emotional effects, (f) lack of support, (g) concerns for the infant, (h) unresolved problems, and (i) unhelpfulness of health professionals.
