Rate nephelometric measurement of wheat germ in pasta products.

An immunoassay for the rate nephelometric determination of wheat germ added to pasta products has been developed using a specific wheat germ antiserum. The method can be used to analyze germ from either soft or hard wheat. Specially prepared pasta products and commercial preparations were analyzed with the immunonephelometric method, in comparison with the radial immunodiffusion technique; good correlation was found between the results of the 2 assays.

A simple and rapid immunoenzymatic assay of prostatic acid phosphatase in serum.

A method is described for the quantitative assay of human prostatic acid phosphatase after its isolation from serum by a double antibody technique employing a specific rabbit antiserum and a goat anti-rabbit gammaglobulin serum. After centrifugation, the precipitate is analyzed for phosphatase activity using p-nitrophenylphosphate as substrate. The method allows a rapid, precise, specific and sensitive assay.

Radio-ligand-receptor assay for human chorionic somatomammotropin (hCS), ovine prolactin (oPRL) and human growth hormone (hGH) using rat ventral prostate particles.

A radio-ligand-receptor assay with a sensitivity of 6.5 x 10(-3) mIU of ovine prolactin per tube has been developed for human chorionic somatomammotropin (hCS) and other lactogenic hormones, using as specific ligand the microsomal pellet of rat ventral prostate. This particulate fraction has proved more suitable than the same fraction obtained from rabbit mammary gland in the evaluation of the prolactin-like activity of purified preparations of hCS.

Effect of the purification procedure on capacity of human chorionic somatomammotropin to bind rat ventral prostate particles.

The purification process can cause alterations in the structure and thus in the activity of the product undergoing purification. In the case of human chorionic somatomammotropin (hCS), a protein hormone of placental origin, the possible effects of purification are unknown, even though it is prepared under mild conditions. hCS is able to displace labelled ovine prolactin from rat ventral prostate membranes and we have measured this activity in a crude and in a purified preparation of the hormone. At the same weight level, both the crude and the purified hormone are equally active.

Chemical, biological and immunological characterization of separated electrophoretic components of human chorionic somatomammotropin (HCS).

Purified HCS preparations ofter show heterogeneity when assayed by electrophoresis, although they may be considered homogeneous according to other criteria (N-terminal amino acid analysis, gel filtration and ultracentrifugation patterns, etc.). Using DEAE-cellulose chromatography, the two main components corresponding to various degrees of hormone amidation were separated. The deamidation kinetic of the electrophoretically slower migrating component was studied and found to be sensitive to temperature and, to a lesser extent, to pH. Denaturing agents strongly increased the deamidation rate, while no effect was found by raising the ionic strength. No difference was evident between the two components and the parent hormone when compared (on a weight basis) in a specific HCS radioimmunoassay system. Even the precipitin reaction in agar gel with an anti-human growth hormone (HGH) serum did not show variations in cross-reactivity versus HGH. Moreover, groups of guinea pigs immunized with the single HCS components, gave antibodies reacting identically with the components and the parent HCS. The prolactin-like activity of the two components differed when determined by the pigeon crop-sac asay: the slower migrating component proved to be less active than the more deamidated one. On the other hand, the activity in two different radioreceptor assays (employing membranes from rabbit mammary gland and rat ventral prostate) was greater for the faster migrating component.

New hemolytic method for determination of antistreptolysin O in whole blood.

A new method is proposed for the determination of antistreptolysin O, based on the properties of streptolysin O. The reduced form of the toxin is hemolytic, whereas the oxidized form is not; this activity can be restored, however, by a reducing agent. Both forms maintain the capacity to bind specific antibodies. Unlike the classical method, the assay is performed on whole blood, using the patient's own erythrocytes as revealing agents. A procedure is proposed which limits the operation to a single dilution of a microvolume of blood (0.05 ml brought to 2 ml) and to the distribution of constant quantities in test tubes prefilled with different amounts of oxidized streptolysin O. After a few minutes, a reducing agent is added, and the reading is performed as in the classical technique after sedimentation of the erythrocytes. Correlation with the traditional method (r = 0.987) is evaluated on the basis of the results of samples from hospitalized patients.
