Cortical responses to stimulation of two functionally different thalamic nuclei in rats.

In acute experiments on adult rats we studied cortical responses to stimulation of two thalamic nuclei--a specific somatosensory relay nucleus (the ventral dorsomedial nucleus, VDM) and a nucleus without specific cortical projection (the lateral anterior nucleus, LA). The responses evoked from the two nuclei by various stimuli have the same basic form--an initial positive phase (inconstant in stimulation of the LA) is followed by dominant negativity and a late slow negative wave; the response ends with a rhythmic afterdischarge. Rhythmic stimulation of the thalamic nuclei produces incremental responses in the cortex and both the positive and the negative phase of the evoked potential participate in the amplitude changes. Hippocampocortical responses (which were compared with thalamocortical responses) have neither a late negative wave nor a distinct rhythmic afterdischarge; in rhythmic stimulation the amplitude of the responses seldom waxes and wanes. Study of the given cortical responses failed to explain the basic difference in the ability of the two thalamic nuclei to produce a self-sustained afterdischarge after rhythmic stimulation, when a spike-and wave rhythm was always elicited from the VDM, while in most cases a "hippocampal" type of self-sustained afterdischarge was evoked from the LA.

Electrocorticographic activity elicited by metrazol during ontogenesis in rats.

Electrocorticographic changes induced by intraperitoneal injections of metrazol were studied in 72 male rats aged from 4 to 29 days. The initial paroxysmal signs varied according to age; 4- to 7-day-old rats: discharges of waves in the alpha and theta range; 11- to 20-day-old rats: spikes in the frontal area; rats aged 24 days and more: theta waves discharges (of about 5Hz) in the occipital area, i.e. a typical mature reaction. In 4- and 5-day-old rats, generalized seizures were manifested as slow waves only. A shift towards fast frequencies was observed during further development; a substantial part of this shift took place before the 14th day. After this age, bilateral synchrony of long-lasting seizure discharges was also present.

Seizures elicited by subcutaneous injection of metrazol during ontogenesis in rats.

Metrazol administered subcutaneously at a dose of 80 mg . kg-1 elicited genuine minimal seizures (predominantly clonic) in adult, 45- and 25-day-old rats. The same dose of metrazol resulted in long-lasting generalized tonic-clonic seizures in rats aged 15 days and younger. Eighteen-day-old rats represented a transitory stage in which both types of seizures were present: minor seizures at first and then, after a short resting period, major seizures. A different seizure pattern in young rats was not induced by their higher sensitivity to metrazol, but was caused by the level of neural maturation.

Automated quantitative gas-liquid chromatography of intact lipis. II. Accuracy, precision and reproducibility of results.

The effect of various factors on the precision and accuracy of the gas chromatographic determination of neutral lipids was studied in the concentration range where the correction factors are dependent on the amount analyzed. The mutual effect of individual components of the neutral lipid spectrum on the recovery was examined. A method is described which provides the stable recovery of the components present at low concentrations, using the addition of high-molecular-weight triglyceride (triarachidin) which does not interfere in the determination of the usual triglycerides. The validity of the correction factors measured with pure compounds was verified by hydrogenation of biological samples of various compositions. Hydrogenation of the sample also solves the problem of the determination of the triglyceride fraction of carbon number 46, which interferes under normal conditions with the determination of the cholesteryl ester fraction of carbon number 47. A method for the standardization of the gas chromatographic determination of neutral lipids is given using pure compounds instead of lyophilized biological samples. Long-term quality control was carried out using synthetic control samples. The results show sufficiently low values of the variation coefficients over the whole period. The values of the variation coefficients measured over an interval of 25 weeks are about 4% for the main components of the neutral lipid spectrum and 6.3% for the components present at concentrations up to 5%. Thw within-day variation for the most neutral lipid fractions and for lipid classes attains a value of 40-75% of the day-to-day variation. The most satisfactory values are obtained for the variation within a single series which amount to less than 2% for all substances except for triglyceride fractions 48 and 54. The correlation of the determination of total cholesterol and triglycerides by gas chromatography and by enzymatic methods shows a very good agreement between the results obtained by the two methods. Using quality control, it is possible to follow the accuracy of the calibration and to demonstrate objectively the necessity for system recalibration.

Ontogenetic development of pentobarbital-induced EEG pattern and sleeping time in rats.

The latency of onset and the duration of sleep induced by pentobarbital was studied in rats from 9 to 35 days of age, as well as in adult rats. Sleep latency was prolonged and sleep duration shortened with increasing age and maturation. Electrocorticographic changes after pentobarbital administration were studied in rats of 5-45 days of age and in adult rats. In rats 9 days of age or less the electrocorticogram manifested only depression of activity, whereas from the 12th day onward all characteristic barbiturate-induced phenomena were registered: spindles, slow waves, and isolated spikes with suppression of background activity. Spindles were the most conspicuous of the pentobarbital-induced phenomena and were the 1st to appear as a function of age, occurring only in the frontal areas of 12 days of age but in both frontal and occipital areas at 15 days of age. Frequency of the elements comprising spindles increased with from 2.5-3.5 c/sec in 12-day-old rats to 5-10 c/sec in adult animals. Barbiturate spindles could be used as a model for rhythmic thalamocortical phenomena even at early stages of development.

Two types of epileptic cortical after-discharges evoked by the stimulation of the hippocampus and thalamic nuclei in rats.

Rhythmic cortical phenomena were evoked by stimulation of hippocampus and/or thalamus in rats. Electrical stimulation of thalamic nuclei (nc. ventralis dorsomedialis VDM and nc. lateralis anterior--LA) by single pulses elicited rhythmic after-discharge ("spindle") as a late component of the cortical response, whereas identical stimulation of hippocampus did not trigger such an activity. Rhythmic stimulation of thalamic nuclei elicited cortical incremental responses more often than stimulation of hippocampus. Epileptic self-sustained after-discharges (AD) occurred after the endo of rhythmic stimulation in 66% of hippocampal stimulations, in 35% of VDM stimulations and in 30% of LA stimulations. AD evoked by stimulation of the hippocampus (long duration discharges of slow serrated waves) differed characteristically from AD following stimulation of the VMD (short duration spike and wave complexes). Stimulation of the LA in half of the cases led to the "hippocampal" pattern of serrated wave after-discharges, in the remaining cases spike-and-wave complexes or a combined AD pattern was recorded.

Ontogenetic development of electrocorticogram in the rat.

Spontaneous electrocorticogram (ECoG) was recorded in frontal (sensorimotor) temporal (auditory) and occipital (visual) cortical regions of 86 male rats (immobilized with d-tubocurarine) aged from 3 days to adulthood. Activity which could be classified as ECoG was for the first time recorded in 5-day-old rats; it was formed by groups of slow waves with unstable frequency intermingled with periods of isoelectric line. Discontinuous ECoG activity was regularly registered even in 10-day-old rats, exceptionally in 12-day-old rats. During further maturation of the continuous ECoG an increase in frequency and an establishment of a basic rhythmic activity synchronous over both hemispheres took place, so that 25- and 30-day-old rats did not differ from the adult ones. Autocorrelagrams and power frequency spectra demonstrated a broad frequency range of the basic rhythm as well as delay in the development of occipital cortical areas in comparison to frontal areas.

The influence of GABA on discharges of cortical epileptogenic focus.

GABA, when applied locally, acted similarly on both primary and mirror cortical focus: the negative component of the spike discharge was suppressed or inverted in polarity, whereas the late slow negative wave was strongly potentiated. Recordings from deep cortical layers suggested a different origin of these 2 surface-negative components of focal discharges.

Automated quantitative gas--liquid chromatography of intact lipids. I. Preparation and calibration of the column.

A method for quantitative gas chromatographic determination of plasma lipids (free cholesterol, cholesteryl esters and triglycerides) in the low concentration range is described. This method permits a determination of not only the lipid classes mentioned above, but also their fractions according to molecular weight, down to 10 ng, without previous derivatization. Special attention was devoted to the preparation of columns with high efficiency and minimal losses of the test substances. The best results were obtained with a glass column 0.5 m x 2.0 mm I.D., packed with 1% OV-1 on Gas-Chrom Q (100--120 mesh). The processing of results is fully automated, using an MDS-2400 computer and includes the calculation of a non-linear calibration plot for each substance analyzed, accuracy control of the measured values, tabulation of the fwr values and the calculation for analyses of biological samples. For the calibration, the pure substances were used at 15 concentrations within a range of 10--1000 ng. The coefficient of variation calculated from 20 duplicate measurements of the calibration mixture did not exceed 5% for any component in the interval from 10 to 100 ng or 3% within range from 100 to 1000 ng.
