Light-stimulated phosphorylation of rhodopsin in the retina: the presence of a protein kinase that is specific for photobleached rhodopsin.

A protein kinase has been extracted from bovine rod outer segments by a mild procedure. The enzyme acts specifically on photobleached, not unbleached, rhodopsin and will not catalyze the phosphorylation of histones, phosvitin, or casein. We propose the name "opsin kinase" for the enzyme, which is not affected by cyclic nucleotides but which is inhibited by theophylline. Preparations of purified rod outer segments, however, appear to contain only low concentration of opsin phosphatase activity.

Effects of synthetic tripeptide on the differentiation of dissociated cerebral hemisphere nerve cells in culture.

Dissociated cells from 7-day-old chick embryo cerebral hemispheres were cultivated on collagen in Falcon Petri dishes in the presence of various concentrations of fetal calf serum and of a chemically synthesized tripeptide Gly-His-Lys. Four different culture conditions were employed in the composition of the nutrient medium in which the cells were cultivated: a low serum concentration of 1, 2 or 5% (group A), a low serum concentration with 200 ng/ml tripeptide (group B), a serum concentration of 10 to 20% (group C) and a serum concentration of 10 or 20% with 200 ng/ml tripeptide (group D). Within the first 24 h of cultivation the cells settled on the collagen substrate and outgrowth of neuronal processes started in all four culture conditions. After 48 h in culture, differences between the groups became evident. In group A most isolated nerve cells had disappeared and glial cells proliferated from the remaining clumps. In group B the neurons had differentiated in absence of glial cells, the proliferation of which was greatly suppressed. In group C and D a differentiation of neurons occurred in a similar way to group B, but in addition the glial cells had proliferated. After 7-8 days in culture the cells in group A and B suddenly degenerated. In group C and D the nerve cells maintained for up to 3 weeks. The optimum concentrations of tripeptide in which the neuroblasts grew fibers and maintained in culture during 7-8 days were in the range of 100-400 ng/ml. The role of the tripeptide in the differentiation and maintenance of nerve cells is discussed.

Morphological, histochemical and chromosomal patterns of neuroblastoma parental and purine resistant lines.

Several neuroblastoma clones and the same clones adapted to proliferation in a medium containing 15 mug/ml of 8-azaguanine and 6-thioguanine are characterized with respect to their morphology, acetylcholinesterase activity, catecholamine content and chromosomal pattern. Interclonal as well as intraclonal heterogeneity was found for the cell parameters studied. A reduction in the number of catecholamine-containing cells was observed in the azaguanine and thioguanine resistant adrenergic (M1, N115) cells compared with their parental lines. An increase of choline acetyltransferase activity was found in the M5 cholinergic clone, and a decrease of the same activity in the S21 cholinergic line selected in the medium with the purine analogues. Furthermore, a striking change in the distribution of chromosomes and chromosomal markers appeared in the resistant cells of all clones.

[Purification of rat brain succinate-semialdehyde dehydrogenase and study of its inhibition by branched chain fatty acids].

Rat brain succinic semialdehyde deshydrogenase has been purified 1300 fold. This enzyme is inhibited non competitively by the same branched chain fatty acids which inhibit GABA-transaminase competitively with respect to GABA. The respective activities of GABA-T and SSADH found in rat brain indicate that at anticonvulsant doses, the acids dipropylacetic and 2-methyl 2-ethyl caproic preferentially inhibit GABA-transaminase thus inducing a rise in cerebral GABA level. This increase is therefore not due to metabolism of the succinic semialdehyde by GABA-T.

[Sulfurtransferase activity in cultured neuronal clone cells].

The activity of 3'-phosphoadenosine-5'-phosphosulfate : galactocerebroside sulphotransferase (PAPS - CST, EC 2.8.2.11), which catalyzes the synthesis of sulphatides, was measured in cloned cells (NIE 115) derived from mouse neuroblastoma C-1300. This activity was of the same order of magnitude as that observed in adult mouse brain. The cell density had no effect on the specific activity of the PAPS-CST.
