Influence of radioprotective agents on the activities of isoenzymes of blood serum alkaline phosphatase in irradiated dogs.

Changes of the total serum activity of alkaline phosphatase (AP) and its bone and intestinal isoenzymes were studied in dogs gamma-irradiated by 3.0 Gy. The activities of AP isoenzymes were determined by means of a heat inactivation-inhibition method. After irradiation the serum AP activities were lower in general. In case of the bone isoenzyme the decrease was most pronounced. The changes of the total AP activity and of the intestinal isoenzyme were less distinct. The i.m. administration of the radioprotective mixture cystamine with mexamine (24 mg/kg + 4 mg/kg) before irradiation led only to a moderate reduction of the radiation-altered AP values in the dogs. The present results indicate that the radioprotective way used here renders only an unsatisfactory protection for large laboratory animals against ionizing radiation.

Transient neonatal diabetes mellitus. Treatment with chlorpropamide.

Nonketotic diabetes mellitus developed in a "small-for-dates" baby at the age of 6 days. The disease was controlled initially with insulin but beginning at the age of 40 days, chlorpropamide was substituted gradually for insulin over a 24-day period. Treatment was stopped at the age of 13 weeks and the baby remained well thereafter. It is suggested that chlorpropamide might be useful in the treatment of transient neonatal diabetes.

Hemodynamic changes in rats after radioprotective combination of cystamine administered subcutaneously and 5-methoxytryptamine injected intramuscularly.

Administration of cystamine (20 mg base/kg s.c.) and 5-methoxytryptamine (10 mg base/kg i.m.) with 12 minutes delay to anesthetized rats induced the depression of whole cardiovascular system, hemoconcentration and great blood flow decrease in the radiosensitive tissues. Distributionof lowered cardiac output preferred the fractions to brain, heart, liver and lungs. Hemodynamic responses to protective combination may participate in its radioprotective action.

Acute cardiovascular responses to radioprotective mixture of cystamine and 5-methoxytryptamine in rats.

I.p. administration of radioprotective mixture of cystamine (18 mg base/kg) and 5-methoxytryptamine (3 mg base/kg) to anesthetized rats induced the depression of hemodynamics. Decrease of cardiac output, hypotension, bradycardia, increase in peripheral vascular resistance, the escape of plasma from the vascular stream, pronounced diminution of blood flow in the spleen and other tissues were determined. Pharmacological properties of the protective can contribute to its radioprotective efficiency in the whole mammalian organism.
