Expression of vascular endothelial growth factor receptors in human prostate cancer.

OBJECTIVES: We recently reported the expression and cytokine regulation of vascular endothelial growth factor (VEGF) in human prostate cancer (PCa). VEGF exerts its angiogenic and pro-tumorigenic properties by way of two high affinity receptors, fms-like tyrosine kinase 1 (FLT-1) and fetal liver kinase 1 (FLK-1). We hypothesized that these receptors are expressed and control VEGF functions in the PCa microenvironment. Herein, we evaluate the expression of these receptors in ex vivo PCa tissue, benign prostatic hypertrophy (BPH) tissue, and cultured PCa cell lines. METHODS: Ex vivo PCa specimens were obtained from patients undergoing radical retropubic prostatectomy. Specimens were selected to contain both PCa and BPH tissue (n = 15). Immunohistochemical analysis using antihuman FLT-1 and FLK-1 was performed and specimens were analyzed to characterize the expression and distribution of both receptors. Immunocytochemical analysis for FLT-1 and FLK-1 was also performed on cultured PCa cell lines (DU-145 and LNCaP). RESULTS: PCa cells expressed the VEGF receptor FLT-1 in 100% of specimens evaluated. Expression of FLK-1 was variable and related to tumor grade; high-grade tumors displayed little or no FLK-1 expression. Vascular endothelial cells (VECs) within areas of PCa consistently expressed both FLT-1 and FLK-1 receptors. FLT-1 and FLK-1 were both expressed in BPH tissue. FLT-1 was expressed in the glandular epithelial cells in BPH, but in most cases FLK-1 was localized specifically to the basal cell layer of hypertrophic glands. FLT-1, but not FLK-1, was expressed by the DU-145 and LNCaP cell lines. CONCLUSIONS: Although they are differentially expressed, both FLT-1 and FLK-1 are present in PCa and BPH. Expression of receptors on VECs of tumor vessels supports the well-established role of VEGF in paracrine stimulation of VECs in the tumor microenvironment. The expression of FLT-1 and FLK-1 on tumor cells themselves suggests a potential autocrine function for VEGF (such as regulating tumor cell proliferation). These findings imply that a novel dual role may exist for VEGF, such that it is involved in tumor cell activation (autocrine), in addition to paracrine actions whereby it regulates endothelial cell functions and subsequent neovascular development.

Localization of chloroacetaldehyde-induced DNA damage in human p53 gene by DNA polymerase fingerprint analysis.

Chloroacetaldehyde (CAA) reacts with DNA bases, forming hydroxyethano derivatives of different stability, which are subsequently converted into etheno (epsilon) adducts: epsilon A, epsilon C, epsilon G. DNA polymerase fingerprint analysis was used to study the distribution of CAA-induced modifications in the p53 sequence. A plasmid bearing cDNA containing the human p53 gene was reacted in vitro with CAA, then dehydrated for conversion of hydroxyethano into etheno adducts, and primer extension by T7 DNA polymerase in the presence of four dNTPs was performed. The DNA repair enzymes methylpurine-DNA glycosylase and Escherichia coli exonuclease III were used to convert epsilon A residues in the template into DNA strand breaks, which enabled precise localization of the epsilon A residues within the p53 gene. Hydroxyethano derivatives of adenine and cytosine in a template blocked T7 DNA polymerase and caused premature chain termination opposite adenine or one base before cytosine. After dehydration, both epsilon A and epsilon C were much more easily by-passed by T7 DNA polymerase. Formation of epsilon G was identified as 'stop bands' one base before guanine residues. Modification of cytosine and guanine was additionally recognized by weakening or disappearance of non-specific stops on an undamaged template, probably due to steric hindrance by the tertiary DNA structure for polymerase. Etheno adduction of cytosine and guanine relaxed the compact DNA structure and enabled DNA polymerase to by-pass. In exons 5-8 of p53, 143 out of 500 sites appeared to be damaged by CAA, with four particularly densely modified regions between codons 135-147, 218-222, 234-255 and 284-292. The pattern of modification followed the pattern of p53 mutations found in vinyl chloride-associated liver angiosarcomas in humans and rats, but only in regions that showed 100% homology with the human sequence. The factors that influence DNA damage and induction of mutations in the p53 gene by CAA and vinyl chloride are discussed.

High-Resolution Study of the Near-Infrared A2Pi1/2-X2Sigma+ Transition of YS.

A high-resolution excitation spectrum of gaseous YS has been recorded by laser excitation spectroscopy in a supersonic molecular beam. The YS molecules were produced by the reaction of laser-ablated yttrium atoms with CS2. The band of the A 2Pi1/2-X 2Sigma+ transition near 750 nm was rotationally analyzed, and a set of molecular constants is given for both electronic states. Copyright 1998 Academic Press.

The Molecular Constants and Potential Energy Curve of the Ground State X1Sigma+ in KLi.

The ground state X1Sigma+ of the diatomic molecule KLi has been studied by analyzing spectra of the B1Pi-X1Sigma+ system, simplified by polarization labeling. Rotational and vibrational constants are reported for the X1Sigma+ state covering about 45% of its potential well depth and the potential curve is constructed by the Rydberg-Klein-Rees (RKR) procedure. Comparison with previous experimental and theoretical work is made. Copyright 1998 Academic Press.

[Ventilation using the laryngeal mask and intubation tube during general anesthesia for ophthalmic surgery].

PURPOSE: The aim of the investigation was to compare efficacy of using for airway maintainance and controlled ventilation the laryngeal mask versus orotracheal intubation. MATERIALS AND METHODS: We carried outs this examination in the group of 50 patient of both sex scheduled for elective operations in ophthalmic surgery. All participants were of I or II class of perioperative risk according to ASA scale. We tried to estimate efficacy of using laryngeal mask versus orotracheal intubation during anaesthesia with controlled ventilation. We examined capillary blood hemoglobin saturation, carbon dioxide pressure in end-tidal expired gas and peak inspiratory pressure while laryngeal mask or orotracheal tube was applied. RESULTS: We found no differences of clinical importance in both group of patients in the range of measured parameters. CONCLUSION: We conclude that ventilation with the laryngeal mask inserted is safe and effective providing that patients are carefully selected.

Study of the 41Piu, 51Piu, 61Piu, and 61Sigmau+ States of K2 by Polarization Labeling Spectroscopy

The polarization labeling spectroscopy technique is used to study excited singlet ungerade states of K2 in the energy range 26 400-30 200 cm-1 above the bottom of the ground state potential. Three states, 4-6(1)Piu, are characterized by sets of Dunham coefficients. The 6(1)Sigmau+ state with an unusual shape of the potential curve is also partially analyzed. Comparison with theoretical calculations is done. Copyright 1997 Academic Press. Copyright 1997Academic Press

[Midazolam administered orally as premedication in children in the ophthalmology department].

The aim of research was using midazolam for preoperational medication in children undergoing diagnostic and minor surgical procedures in ophthalmic surgery and estimation of its properties for deep sedation. We used midazolam in children of both sex, from 2 to 10 years of age. All the patients were considered as I or II grade of ASA scale of perioperational risk. Patients were given midazolam orally at dose 0.5 mg per kg of body weight. Sedation degree was assessed according to Addenbrooke's Hospital Scale of Sedation as well as to changes in systolic (SAP) and diastolic (DAP) arterial pressure, pulse oximetry of capillary blood (SpO2) and heart rate (HR). Sedation in all children subjected to the trial was good and optimal conditions to perform the procedures were achieved. We found the level of sedation satisfactory and safe. Measurement of SAP, DAP, SpO2 and HR revealed stability of circulation in the whole perioperative period. We found the sedative properties of midazolam as excellent in the described range of age and useful for the diagnostics and surgery respectively.

CoMFA analysis of the interactions of antipicornavirus compounds in the binding pocket of human rhinovirus-14.

A CoMFA analysis of eight compounds related to disoxaril whose X-ray structures bound to HRV-14 had been determined resulted in a strong positive correlation of activity with steric effects of the compounds, particularly toward the pore end of the compound binding site, and no correlation with electrostatic effects. These results confirm what had been previously found, that the activity of these compounds was highly dependent upon their hydrophobic nature as expressed by log p. The CoMFA study also confirmed the results from the comparison of a series of active and inactive compounds using volume maps which showed that bulk at the pore end of the molecule was conducive to high levels of antiviral activity while excessive bulk around the ring led to poor activity.
