T-cell subsets: effects of cryopreservation, paraformaldehyde fixation, incubation regime and choice of fluorescein-conjugated anti-mouse IgG on the percentage positive cells stained with monoclonal antibodies.

The percentages of positive lymphocytes stained with OKT3 (total), OKT4 (helper/inducer) or OKT8 (suppressor/cytotoxic) monoclonal antibodies have been determined in normal individuals. Under a variety of experimental conditions, the results were found to be unaffected by cryopreservation, paraformaldehyde fixation or incubation regime. In contrast, the results were affected, both quantitatively and qualitatively, by the choice of fluorescein-conjugated anti-mouse IgG as the second step reagent.

Testosterone induction of microsomal acyl-CoA reductase and a cytosolic regulatory protein in mouse preputial glands.

Alkyl and alk-1-enyl (plasmalogens) ether-linked glycerolipids are prominent components of many mammalian cells; moreover, an acetylated form of an alkyl phospholipid was recently found to possess potent hypotensive, inflammatory and allergic properties. In our studies, preputial glands of mice were selected as a model to investigate the regulation of factors involved in the biosynthesis of ether-linked lipids, since these glands contain high concentrations of ether-linked neutral lipids that are under the influence of hormonal control. We found that a key enzyme in the ether-lipid metabolic pathway, microsomal acyl-CoA reductase that catalyzes the formation of long-chain fatty alcohols (precursor of the O-alkyl chain), was increased 16-fold after injecting testosterone into male, castrated mice. This induction was highly specific, since testosterone did not affect another microsomal enzyme, NADPH-cytochrome c reductase. Based on kinetics of enzyme activity changes, the half-life of acyl-CoA reductase was calculated to be 61-70 h. In addition, the activity of a cytosolic stimulatory protein for the acyl-CoA reductase (but not for a different cytosolic protein, lactate dehydrogenase) was also enhanced in the testosterone-treated, male, castrated mice. These findings indicate that acyl-CoA reductase is an important regulatory enzyme in the reactions that lead to the formation of the ether bond in glycerolipids and that it is modulated through hormonal control.

Commercial factor VIII associated hepatitis, 1974-75, in the United Kingdom: a retrospective survey.

A retrospective survey of transfusion hepatitis associated with a brand of commercial Factor VIII was carried out in 24 Haemophilia Centres from January 1974 until December 1975. Of 371 patients who were transfused with this product, and were followed up, 78 cases of hepatitis affecting 66 patients were found (17.7%). Two types of hepatitis were observed: hepatitis B and non-B hepatitis, the latter with an incubation period of between 8 and 60 days. Twelve patients contracted two types of hepatitis, non-B followed by hepatitis B. Only one patient died after contracting hepatitis B. Four of the suspect batches of concentrate were found to be positive for HBsAg by radioimmunoassay. There was evidence that the presence of hepatitis B surface antibody in a patient's serum prior to exposure was associated with immunity to hepatitis B. Evidence was presented suggesting that the non-B hepatitis observed was not due to hepatitis A. The factors affecting the incidence of transfusion hepatitis in haemophiliacs were discussed.

Testing the relative conspicuity of safety garments for New Zealand forestry workers.

The relative conspicuities of six test garments (fluorescent orange, green/red, high-contrast, fluorescent lime-yellow, white, black) were assessed to guide the selection process of a standard upper body safety garment to be used within the New Zealand logging industry. Six male and four female participants, aged between 18 and 26 years, volunteered to perform 10 trials each on a demanding, central (tracking) task, while peripherally searching colour slides for test garments displayed on each of eight positions, against a pine forest background typically found in the New Zealand forestry. During each trial, transparency luminance was first gradually increased from darkness to daylight and then decreased to darkness again within 180 s. A head mounted ASL eye tracking system (4000SU) recorded the eye line of gaze for each participant, enabling a rank order of detection to be obtained for the tested garments in each trial. The fluorescent lime-yellow, fluorescent orange and white test garments were detected earlier than any other test garments across all trials, and the fluorescent lime-yellow test garment was detected first with a higher frequency than the white test garment. It was concluded that while white may be the most visible colour in near darkness conditions, as it provides highest contrast, fluorescent lime-yellow stands out better in twilight and daylight conditions against the pine forest background. The results of this study led to the promotion of fluorescent lime-yellow as the standard safety colour used in upper body garments within the New Zealand forest industry.
