A novel dodecapeptide from a combinatorial synthetic library exhibits potent antifungal activity and synergy with standard antimycotic agents.

There has been a marked expansion in the discovery of new antifungal peptides. This paper describes a novel dodecapeptide, H-Arg-Trp-Trp-Arg-D-Trp-D-Phe-Ile-D-Phe-His-Trp-Arg-Trp-NH(2), derived from a previously described nonapeptide and synthesized by the combinatorial approach. Further, interaction of this peptide with antifungals such as amphotericin B, flucytosine and fluconazole was studied by checkerboard analysis and time-kill assay to obtain the dynamic picture with respect to time. The best synergistic activity was observed with a combination of peptide and fluconazole, followed by peptide and flucytosine.

New antifungal flavonoid glycoside from Vitex negundo.

Flavonoids are ubiquitous in photosynthesizing cells and are common part of human diet. For centuries, preparations containing these compounds as the principal physiologically active constituents have been used to treat human diseases. Increasingly, this class of natural products is becoming the subject of anti-infective research. Our bioactivity guided fractionation of ethanolic extract of leaves of Vitex negundo resulted in the isolation of new flavone glycoside (4) along with five known compounds 1-3, 5 and 6. All the isolated compounds were evaluated for their antimicrobial activities. The new flavone glycoside 4 and compound 5 were found to have significant antifungal activity against Trichophyton mentagrophytes and Cryptococcus neoformans at MIC 6.25 microg/ml.

Instability and dynamics of two nonlinearly coupled laser beams in a two-temperature electron plasma.

We consider nonlinear interactions between two colliding laser beams in an electron plasma, accounting for the relativistic electron mass increase in the laser fields and radiation pressure driven electron-acoustic (EA) perturbations that are supported by hot and cold electrons. By using the hydrodynamic and Maxwell equations, we obtain the relevant equations for nonlinearly coupled laser beams and EA perturbations. The coupled equations are then Fourier analyzed to obtain a nonlinear dispersion relation. The latter is numerically solved to show the existence of new classes of the parametric instabilities in the presence of two colliding laser beams in a two-electron plasma. The dynamics of nonlinearly coupled laser beams in our electron plasma is also investigated. The results should be useful in understanding the nonlinear propagation characteristics of multiple electromagnetic beams in laser-produced plasmas as well as in space plasmas.

Instability and evolution of nonlinearly interacting water waves.

We consider the modulational instability of nonlinearly interacting two-dimensional waves in deep water, which are described by a pair of two-dimensional coupled nonlinear Schrödinger equations. We derive a nonlinear dispersion relation. The latter is numerically analyzed to obtain the regions and the associated growth rates of the modulational instability. Furthermore, we follow the long term evolution of the latter by means of computer simulations of the governing nonlinear equations and demonstrate the formation of localized coherent wave envelopes. Our results should be useful for understanding the formation and nonlinear propagation characteristics of large-amplitude freak waves in deep water.

Antifungal anthraquinones from Saprosma fragrans.

A new 3,4-dihydroxy-1-methoxy anthraquinone-2-corboxaldehyde (1) together with a known anthraquinone, damnacanthal (2), were isolated from the chloroform fraction of the aerial part (whole plant without root) of Saprosma fragrans. The isolated anthraquinones (1) and (2) were found to exhibit antifungal activity against Trichophyton mentagrophytes and Sporitrichum schenckii. Their structures were established by chemical and spectral analysis.

Formation and dynamics of dark solitons and vortices in quantum electron plasmas.

We present simulation studies of the formation and dynamics of dark solitons and vortices in quantum electron plasmas. The electron dynamics in the latter is governed by a pair of equations comprising the nonlinear Schrödinger and Poisson system of equations, which conserves the number of electrons as well as their momentum and energy. The present governing equations in one spatial dimension admit stationary solutions in the form a dark envelope soliton. The dynamics of the latter reveals its robustness. Furthermore, we numerically demonstrate the existence of cylindrically symmetric two-dimensional quantum electron vortices, which survive during collisions. The nonlinear structures presented here may serve the purpose of transporting information at quantum scales in ultracold micromechanical systems and dense plasmas, such as those created during intense laser-matter interactions.

Filamentational instability of partially coherent femtosecond optical pulses in air.

The filamentational instability of spatially broadband femtosecond optical pulses in air is investigated by means of a kinetic wave equation for spatially incoherent photons. An explicit expression for the spatial amplification rate is derived and analyzed. It is found that the spatial spectral broadening of the pulse can lead to stabilization of the filamentation instability. Thus optical smoothing techniques could optimize current applications of ultrashort laser pulses, such as atmospheric remote sensing.

Modulational instability of partially coherent signals in electrical transmission lines.

We present an investigation of the modulational instability of partially coherent signals in electrical transmission lines. Starting from the modified Ginzburg-Landau equations and the Wigner-Moyal representation, we derive a nonlinear dispersion relation for the modulational instability. It is found that the effect of signal broadbandness reduces the growth rate of the modulational instability.

Synthesis and antibacterial evaluation of ureides of Baylis-Hillman derivatives.

The synthesis of several 1-(2-cyano-3-aryl-allyl)-3-aryl-urea(thiourea) constructed from the reaction between allylamines generated from Baylis-Hillman acetates and substituted isocyanates and isothiocyanates has been described. Further, their cyclization in the presence of a base led to the formation of 5-arylmethyl-4-imino-3-aryl-3,4-dihydro-1H-pyrimidin-2-ones. All compounds were tested for their antibacterial activity. Few of the compounds showed superior activity or were equipotent to the standard antibacterial agents.

Candidacidal activity of a monoclonal antibody that binds with glycosyl moieties of proteins of Candida albicans.

The incidence of life threatening mycoses caused by opportunistic fungi has increased dramatically in recent years with Candida and Aspergillus being the most commonly encountered species. Candida albicans ranks among the four most common causes of bloodstream infections and is responsible for vulvovaginal candidiasis in the majority of women in their reproductive years. Limited spectrum of antifungal activity of currently available antifungals and emergence of resistance has become a serious problem. Therefore, in search of an alternative form of treatment of candidiasis, in the present study a monoclonal antibody (MAb-G5) of IgA isotype was identified from the hybridoma produced by the fusion of lymphocytes of C. albicans immunized mouse with Sp2/O cells. The MAb-G5 exhibited in vitro candidacidal activity and was also found to be useful for treatment and prophylactic use under experimental conditions in vivo.

Nonlinear propagation of electromagnetic waves in negative-refraction-index composite materials.

We investigate the nonlinear propagation of electromagnetic waves in left-handed materials. For this purpose, we consider a set of coupled nonlinear Schrödinger (CNLS) equations, which govern the dynamics of coupled electric and magnetic field envelopes. The CNLS equations are used to obtain a nonlinear dispersion, which depicts the modulational stability profile of the coupled plane-wave solutions in left-handed materials. An exact (in)stability criterion for modulational interactions is derived, and analytical expressions for the instability growth rate are obtained.

Formation and dynamics of finite amplitude localized pulses in elastic tubes.

We present theoretical and simulation studies of the formation and dynamics of finite-amplitude localized pulses (solitary waves) of an incompressible fluid in an elastic tube. Starting from a set of hydrodynamic equations, we derive a Hamiltonian which represents the energy integral of our system. The energy integral is analyzed to obtain explicit profiles of finite-amplitude solitary pulses. Also studied are the excitation and dynamics of solitary pulses by using computer simulations. It is found that a train of solitary pulses can be excited by the nonlinear self-steepening at shock fronts. The relevance of our investigation to blood solitary waves in arteries is discussed.

Discovery of substituted isoxazolecarbaldehydes as potent spermicides, acrosin inhibitors and mild anti-fungal agents.

BACKGROUND: The continued endeavour to design novel, non-detergent molecules that can be useful as topical, prophylactic contraceptives has led to the discovery of substituted isoxazolecarbaldehydes as a new class of compounds exhibiting both spermicidal and acrosin inhibitory activities simultaneously. METHODS: Normal human semen samples were used to detect the spermicidal and acrosin inhibitory activities of the new compounds. Lactobacillus, HeLa and Candida cultures were used to determine the safety of compounds towards normal vaginal flora, their cytotoxicity and anti-fungal activity. Supravital staining and the hypo-osmotic swelling test (HOST) were used to detect the effect on sperm membrane integrity. Nonoxynol-9 (N-9) was used as a reference standard. RESULTS: The 5- and 3-substituted isoxazolecarbaldehydes showed significant spermicidal [minimum effective concentration (MEC)=0.005-2.5%] and acrosin inhibitory (IC50=3.9-58 x 10(-4) mol/l) activities in several molecules along with weak fungicidal activity against Candida albicans. Lineweaver-Burk and Dixon plot analysis of a representative structure showed non-competitive inhibition of human acrosin enzyme, and the most potent acrosin inhibitors also considerably diminished the induction of the acrosome reaction by Ca2+ ionophore. Some compounds were found to be significantly safer than N-9 towards Lactobacillus acidophilus in vitro at their respective spermicidal MECs. In the cytotoxicity assay, the IC50 of these compounds towards the HeLa cell line was of the same order as N-9 (0.9-0.1 mmol/l); however, in contrast, the compounds exhibited only a moderate effect on sperm membrane integrity. CONCLUSIONS: This study indicates that 5- and 3-substituted isoxazolecarbaldehydes are 'first generation' multifunctional, spermicidal molecules that hold promise for development as topical contraceptives with useful associated activities that can add considerably to their effectiveness, safety and prophylaxis.

Quasiparticle approach to the modulational instability of drift waves coupling to zonal flows.

The interaction between broadband drift mode turbulence and zonal flows has been studied through the wave-kinetic approach. Simulations have been conducted in which a particle-in-cell representation is used for the quasiparticles, while a fluid model is employed for the plasma. The interactions have been studied in a plasma edge configuration which has applications in both tokamak physics and magnetopause boundary layer studies. Simulation results show the development of a zonal flow through the modulational instability of the drift wave distribution, as well as the existence of solitary zonal flow structures about an ion gyroradius wide, drifting towards steeper relative density gradients.

Solitary phase-space holes in pair plasmas.

We present theoretical and computer simulation studies of the formation and dynamics of solitary phase-space holes in pair plasmas, which can be applied to both electron-positron plasmas and to plasmas containing positively and negatively charged macro-ions or macro-particulates (charged dust grains). We apply our numerical treatment to the parameters used in a recent series of experiments in a pair ion plasma whose constituents are negatively and positively charged fullerene carbon nanotubes. New experiments should be conducted to confirm our theoretical and numerical predictions.
