Malotilate reduces collagen synthesis and cell migration activity of fibroblasts in vitro.

Diisopropyl-1,3-dithiol-2-ylidenemalonate (malotilate) was studied for and compared with cyanidanol, hydrocortisone and colchicin on its impact on fibroblast cultures. Under in vitro conditions, malotilate specifically reduces collagen synthesis of fibroblasts. In addition, malotilate is an efficient inhibitor of fibroblast migration as tested by a chemotaxis assay in a modified Boyden chamber. Our data may support the notion that malotilate is of potential interest for interfering with fibrotic processes.

Effect of retinoids on collagen production by chondrocytes in culture.

The effect of vitamin A and of some of its derivatives on chondrocytes in culture has been studied. In the presence of retinoids the proliferation of the cells decreased and they lost their characteristic polygonal shape and assumed a fibroblast-like morphology. All retinoids also caused dedifferentiation of chondrocytes as indicated by the induction of types I and III collagen. 13-cis retinoic acid (= isotretinoin) was the most active derivative in this aspect. Since appropriate control of the synthesis of extracellular matrix proteins is a prerequisite for their normal physiological function, alterations such as those observed here may be involved in the pathogenesis of side effects which are observed during the treatment of dermatological disorders with retinoic acid derivatives.

Lipoperoxidation of lung lipids in rats exposed to nitrogen dioxide.

Absorption spectra characteristic of diene conjugation and typical for peroxidized polyenoic fatty acids can be induced in rat lung lipids after the rats have been exposed to a scant amount of nitrogen dioxide (1 part per million) for 4 hours. The peroxidative changes do not occur immediately but appear to reach a maximum between 24 and 48 hours after exposure. The prooxidant effect of this atmospheric pollutant in rat lung lipids may be partially prevented by prior treatment of the animal with large doses of alpha-tocopherol.
