In male rats hypophysectomy reduced the wet weight of hearts per 100 g body weight. The yield of mitochondrial protein per g heart was also decreased which is in contrast to that in liver. Adiminstration of bGH to hypophysectomized rats reversed these changes towards normal. It appears that some of the effects of growth hormone may vary in different tissues.
A mildly obese 15-year-old boy had short stature with rounded facies and short, stubby hands and toes. He had the fully expressed syndrome of pseudohypoparathyroidism but was the only member of his family who had all the somatic characteristics of this disease. The serum parathyroid hormone level was substantially elevated. Urinary excretion of cyclic adenosine monophosphate and phosphate failed to increase following intravenous infusion of parathyroid hormone. However, he did not have hypocalcemia. The present entity is probably a transient form of pseudohypoparathyroidism with partial responsiveness of skeletal adenyl cyclase to parathyroid hormone.
Urinary xanthurenic and kynurenic acid levels were measured in five patients while they were receiving 50 mg and 100 mg of pyridoxine. The levels of tryptophane metabolite decreased progressively as the dose was increased but remained above basal levels. There was marked clinical improvement in these patients while receiving the higher dose only. The double-blind study with 76 asthmatic children followed for five months indicated significant improvement in asthma following pyridoxine therapy (200 mg daily) and reduction in dosage of bronchodilators and cortisone. The data suggest that these children with severe bronchial asthma had a metabolic block in tryptophane metabolism, which was benefitted by long-term treatment with large doses of pyridoxine.
The effect of cortisol and estradiol on the stimulation by growth hormone of liver mitochondrial protein synthesis in hypophysectomized rats has been studied by measuring radioactive leucine incorporation in vivo and in vitro. Estradiol inhibited the stimulatory effect of growth hormone on incorporation into mitochondrial and microsomal proteins. Cortisol blocked the stimulatory effect of growth hormone on mitochondrial protein synthesis. The stimulatory effects of cortisol and growth hormone on microsomal incorporation were not additive. Gain in body weights corresponded with their effects on mitochondrial and microsomal protein synthesis. These results show that steroid hormones may act as regulators of the growth hormone effect.
Seventeen out of 21 children with severe asthma had an abnormal increase in urinary Kynurenic (KA) and Xanthurenic (XA) acid following oral tryptophane administration. In a second study 11 asthmatic children had significantly greater increases in XA and KA than nine normal healthy children. A micro-assay of KA and XA by column chromatography was performed. The data suggest the presence of a partial block in the metabolism of tryptophane in some children with bronchial asthma.
