Deletions involving two distinct regions of 6q in B-cell non-Hodgkin lymphoma.

The recurrent loss of genetic material from a specific chromosomal region in a given tumor type suggests the presence of a tumor-suppressor gene, the loss or inactivation of which may be relevant for tumorigenesis. In this study, we provide molecular evidence for the recurrent association between deletions on the long arm of chromosome 6 and B-cell non-Hodgkin lymphoma (B-NHL). Normal and tumor DNAs from 71 cases of B-NHL were studied for loss of constitutional heterozygosity (LOH) at 19 loci on chromosome 6 using a panel of restriction fragment length polymorphism (RFLP) probes. LOH, indicating deletion of all or part of 6q, was detected in 16 of 71 cases (22.5%), ranging from low-grade to high-grade B-NHL. The isolated loss of 6p or the loss of other chromosomes (8, 17, 22) tested as controls for specificity was not observed in any case. Comparison of the extent of the deletions among different cases allowed the identification of two distinct regions of minimal deletion (RMD) at 6q25 to 6q27 (RMD-1) and at 6q21 to 6q23 (RMD-2), respectively, suggesting the existence of two tumor-suppressor genes. These data support a role for 6q deletions in B-NHL pathogenesis and provide a basis for identifying the corresponding tumor-suppressor genes.

Cytogenetic evidence for extramedullary blast crisis with t(8;13)(q11;p11) in chronic myelomonocytic leukemia.

A 27-year-old male developed massive generalized lymphadenopathy with chronic myelomonocytic leukemia (CMML) presenting as extramedullary blast crisis mimicking a lymphocytic lymphoma. On presentation, a consistent chromosomal abnormality involving chromosomes 8 and 13, i.e. 46,XY,t(8;13) (q11;p11), was present in lymph node tissue, bone marrow and unstimulated peripheral blood. The appearance of trisomy 21 in addition to the presence of the original cytogenetic abnormality is simply regarded as clonal evolution, i.e. 47,XY,t(8;13)(q11;p11),+21. The importance of the cytogenetics lies in finding the same abnormality in bone marrow and lymph node, adding evidence that the immunologically similar cells in the two sites have arisen from a common progenitor cell. To our knowledge, this novel chromosomal abnormality has not been reported in association with a unique case.

Anatomical studies on the prediaphragmatic portion of systema nervosum pars sympathica of the buffalo (Bubalus bubalis).

The gross disposition of the prediaphragmatic portion of systema nervosum pars sympathica was studied in six male buffalo calves after careful dissection. The cranial cervical ganglion, the cervicothoracic ganglion and the ansa subclavia were present bilaterally. Rarely a middle cervical ganglion was also encountered. The vertebral ganglia were present in most of the animals. The cranial cervical sympathetic trunk joined the N. Vagus at the level of atlas and the caudal cervical sympathetic trunk separated from the N. vagus at the sixth cervical vertebral level. The ansa subclavia joined the vertebral and the cervicothoracic ganglia. The myelinated fibres in the sympathetic trunk showed a fascicular arrangement. They were mostly of 2 to 4 mum in external diameter. The number (mean value) of myelinated fibres in the cranial cervical trunk was 8,981 +/- 2,016 and their density was 3,379/mm2. In the caudal cervical trunk, there was 10,345 +/- 3,424 myelinated fibres with a density of 3,679/mm2. In the ansa subclavia, the corresponding values were 3,301 +/- 421 and 2,456/mm2 respectively. The interganglionic trunk between the vertebral and the cervicothoracic ganglia revealed 9,843 +/- 4,602 myelinated fibres and their density was 3,698/mm2. In the interganglionic trunks between the third and the fourth and the fourth and fifth thoracic ganglia, the total number of myelinated nerve fibres was 10,540 +/- 4,127 and 9,217 +/- 1,367 respectively and their density values were 2,750/mm2 and 2,808/mm2 respectively. The corresponding values for the N. transversarius were 26,140 +/- 3,807 and 2,433/mm2 respectively. Ganglion cells were found along the caudal cervical sympathetic trunk and the thoracic sympathetic trunk. A fascicle of umnyelinated fibres was also found in the cervical sympathetic trunks.

Histological studies on certain autonomic ganglia in the buffalo (Bubalus bubalis).

In the buffalo the sympathetic ganglionic neurons were multipolar and contained finely granular or vesicular Nissl material and eccentric nuclei with more than one nucleolus. They measured (mean values) 23.62 X 14.30 mum in the cranial cervical ganglion, 22.51 X 13.58 mum in the vertebral ganglion, 25.25 X 15.75 mum in the cervicothoracic ganglion and 22.84 X 14.27 mum in the fourth thoracic ganglion. Their density values were 256 neurons/mm2 in the cranial cervical ganglion, 250 neurons/mm2 in the vertebral ganglion, 248 neurons/mm2 in the cervicothoracic ganglion and 252 neurons/mm2 in the fourth thoracic ganglion.
