Comprehensive laboratory evaluation of impotent men.

A thorough diagnostic evaluation was done on 27 impotent men. In addition to a detailed history and physical examination each patient underwent a psychiatric interview, Doppler penile blood pressure determination and measurement of the bulbocavernosus reflex latency time. Abnormalities in penile blood pressure were detected in 67 per cent of the patients, neurologic abnormalities were found in 41 per cent and psychiatric disorders were noted in 35 per cent. Of the 27 patients 48 per cent had a combination of neurologic, psychiatric and vascular abnormalities. A definite cause-and-effect relationship between these abnormalities and impotence has not been established.

Initial, clinical trial of a new, specific 5-HT reuptake inhibitor, citalopram (Lu 10-171).

Ten endogenously depressed inpatients were treated once daily for at least 4 weeks with 40-50 mg citalopram (Lu 10-171) - a specific serotonin reuptake inhibitor. The Hamilton rating scale for depression and global assessment indicated pronounced or moderate response in seven patients and slight or no response in three. Side effects were few, mild and transient and neither anticholinergic nor cardiotoxic effects were observed. No difference was observed between responders and non-responders as regards psychopathology, plasma levels or uptake inhibition. It is concluded, that citalopram seems to possess antidepressant properties and that controlled trials are wanted to evaluate its therapeutic value as well as the underlying hypothesis of serotonin - deficient depressions.

Renal function after high-dose urography in patients with chronic renal insufficiency.

Renal function was assessed by frequent recordings of Ccr and serum creatinine in 22 patients with chronic renal failure before and after high-dose urography. Urography was followed by a significant, moderate and transient decrease in Ccr which was maximal on the second day; at the same time a significant increase in serum creatinine was observed which was maximal on the fourth day after urography. In all but two patients, Ccr and serum creatinine returned to pre-urographic levels within the next two weeks. Possible causes of the deleterious effects of the contrast media on the kidneys are discussed.

Division cycle of Myxococcus xanthus. 3. Kinetics of cell growth and protein synthesis.

The kinetics of cell growth and protein synthesis during the division cycle of Myxococcus xanthus was determined. The distribution of cell size for both septated and nonseptated bacteria was obtained by direct measurement of the lengths of 8,000 cells. The Collins-Richmond equation was modified to consider bacterial growth in two phases: growth and division. From the derived equation, the growth rate of individual cells was computed as a function of size. Nondividing cells (growth phase) comprised 91% of the population and took up 87% of the time of the division cycle. The absolute and specific growth rates of nondividing cells were observed to increase continually throughout the growth phase; the growth rate of dividing cells could not be determined accurately by this technique because of changes in the geometry of cells between the time of septation and physical separation. The rate of protein synthesis during the division cycle was measured by pulselabeling an exponential-phase culture with radio-active valine or arginine and then preparing the cells for quantitative autoradiography. By measuring the size of individual cells as well as the number of grains, the rate of protein synthesis as a function of cell size was obtained. Nondividing cells showed an increase in both the absolute and specific rates of protein synthesis throughout the growth phase; the specific rate of protein synthesis for dividing cells was low when compared to growthphase cells. Cell growth and protein synthesis are compared to the previously reported kinetics of deoxyribonucleic acid and ribonucleic acid synthesis during the division cycle.

Deoxyribonucleic acid synthesis during exponential growth and microcyst formation in Myxococcus xanthus.

Myxococcus xanthus in exponential phase with a generation time of 270 min contained a period of 50 min during which deoxyribonucleic acid (DNA) synthesis did not take place. After induction of microcysts by the glycerol technique, the DNA content increased 19%. Autoradiographic experiments demonstrated that the DNA made after glycerol induction was not evenly distributed among the microcysts. The distribution of grains per microcyst fits the following model of chromosome replication: in exponential phase, each daughter cell receives two chromosomes which are replicated sequentially during 80% of the divison cycle; after microcyst induction, no chromosomes are initiated. Mathematical formulas were derived which predict the kinetics and discrete probability distribution for several chromosome models.

An influenza A live attenuated reassortant virus possessing three temperature-sensitive mutations in the PB2 polymerase gene rapidly loses temperature sensitivity following replication in hamsters.

The purpose of the present study was to produce an influenza A H2N2 donor virus from which an attenuating PB2 gene bearing three discrete temperature sensitive (ts) mutations could be readily transferred to currently epidemic influenza A H1N1 and H3N2 viruses via genetic reassortment. An influenza A transfectant virus was first produced that contained site-directed ts mutations at amino acids 112, 265, and 556 in the PB2 gene of influenza A/AA/60 virus origin in a background of the other seven RNA segments from the influenza A/LA/87 (H3N2) virus. The A/LA/87 PB2 ts transfectant virus (clone 22B1) was mated with the A/AA/60 (H2N2) wild type virus, and six H2N2 ts reassortants were obtained. One reassortant virus, clone 25A1, possessed the triple ts PB2 gene in the context of all seven other genes of homologous A/AA/60 origin. Isolation of this reassortant permitted an examination of the contribution of the ts mutations present in a triple ts PB2 transfectant virus to its attenuation and phenotypic stability independent from an effect of the A/AA/60-A/LA/87 gene constellation on attenuation. It was found that the A/AA/60 triple ts reassortant virus was less ts, less attenuated, and less phenotypically stable than the A/LA/87 triple ts transfectant virus from which it was derived. The A/AA/60 reassortant possessing the PB2 gene containing three introduced ts mutations underwent rapid and significant loss of its temperature sensitivity following replication in the lungs of immunocompetent hamsters. This indicated that the A/AA/60-A/LA/87 gene constellation contributed significantly to the overall level of temperature-sensitivity, attenuation, and stability of the A/LA/87 triple ts transfectant virus. It is likely that the instability of the ts phenotype exhibited by the A/AA/60 triple ts reassortant virus would not be acceptable for a vaccine to be used in humans. The implications of these findings for the usefulness of ts mutations as the sole attenuating mutation in influenza virus vaccines is discussed.
