Synthesis, antibacterial activity and structure-activity relationships of N-substituted 4-diazo-pyrazole-5-carboxamides. 2.

A series of 4-diazopyrazole-5-carboxamides 1 d-n has been synthesized and their antibacterial activity against a number of Gram-negative and Gram-positive strains has been tested. Some of the compounds resulted quite active and the whole set allowed to further study the SAR of the class. Substituents in position 5 affect Gram-negative and Gram-positive activities via bulk and electronic properties respectively; position 3 mostly affects the Gram-negative activity, while the presence of the charged diazo group in position 4 is crucial for both antibacterial activities.

Protection by pyroglutamic acid and some of its newly synthesized derivatives against glutamate-induced seizures in mice.

The protection by pyroglutamic acid (CAS 98-79-3) and derivatives Ia-i (injected i.p.) against glutamate- and NMDA (N-methyl-D-aspartate) (i.c.v.) induced seizures in mice has been studied in comparison with known antiepileptics and antagonists of excitatory aminoacids. The potency of pyroglutamic acid and some derivatives (Id,f,g,h) against glutamate-induced convulsions was similar to that shown by glutamic acid diethylester and by valproic acid. Interestingly, pyroglutamic acid did not affect NMDA-induced convulsions which were well antagonized by both 2-amino-5-phosphono valeric acid and by diazepam. Thus, pyroglutamic acid may represent the starting for synthesis of excitatory aminoacid antagonists acting at non NMDA receptors.

[Synthesis and evaluation of the antifungal activity of a new series of N-acylureas].

Four new series of N-Acylureas (IIIa-h), (IV a-h), (Va-h), (VIa-h) were synthesized and tested for phytoiatric antimycotic activity. Only few of the tested compounds (IIIg), (IIIh), (IVf), (IVh), (Vg), (Vh), (VIg) showed a fairly good activity against P. irregulare, whereas no compounds exhibited any activity in the in vivo tests (in the preventive phase).

Synthesis and cytostatic activity of geiparvarin analogues.

In an attempt to determine some of the structural features of geiparvarin (1) that account for its cytostatic activity in vitro, a series of geiparvarin analogues (4a-g) modified in the 3(2H)-furanone moiety have been designed and synthesized. The preparation of 4a-g was achieved through a new approach to the 3(2H)-furanone ring based on the elaboration of isoxazole derivatives. Among these synthetic analogues, 4b, the 5-methyl-5-ethyl derivative, proved as active as 1 in inhibiting the proliferation of murine and human tumor cell lines in vitro. As a rule, substitutions at the C5 atom of the 3(2H)-furanone moiety of 1 slightly decreased the cytostatic activity of geiparvarin. Several geiparvarin analogues described in this study (i.e. the 5-methyl-5-ethyl derivative 4b, 3(2H)-furanimine 4c, 5-methyl derivative 4f, and 5-ethyl derivative 4g) showed such activity in vitro and deserve further investigation for their antitumor potentials in vivo.

Synthesis, antibacterial activity and structure-activity relationships of N-substituted 3-methyl-4-diazo-5-pyrazolecarboxamides.

A number of N-substituted 3-methyl-4-diazo-5-pyrazolecarboxamides (IVa-p) have been synthesized and tested for their in vitro antibacterial activity. All the compounds have been assayed against several representative Gram-negative and Gram-positive bacteria, as well as against some intestinal bacterial species. Some of the 4-diazopyrazoles IVa-c have shown a quite interesting broad-spectrum activity, while they are ineffective against the "protective" intestinal flora. The structure-activity relationships of the series have been studied quantitatively, via both univariate and multivariate methods: the results are consistent and permit some rationalization of the behaviour of the compounds investigated.

Synthesis of 1H-pyrazolo[4,3-d]pyrimidine-7(6H)-ones and pyrazolo-5-carboxamides and interaction with benzodiazepine and adenosine A1 receptors in rat cerebral cortex.

The effects of several newly synthesized pyrazolo [4,3-d] pyrimidine-7-ones and pyrazolo-5-carboxamides on [3H]-flunitrazepam and [3H]-cyclohexyladenosine binding in rat cerebral cortex membranes have been studied. The compounds under examination, displaying affinities in the micromolar range for both benzodiazepine and adenosine A1 receptors, could be of interest in the study of a possible link between the two receptor systems.

Synthesis and interaction of 5-(substituted-phenyl)-3-methyl-6,7-dihydropyrazolo[4,3-e] [1,4]diazepin-8(7H)-ones with benzodiazepine receptors in rat cerebral cortex.

On the basis of the anxiolytic property of ripazepam, 1-ethyl-4,6-dihydro-3- methyl-8-phenylpyrazolo[4,3-e][1,4]diazepin-5(1H)-one (1), a series of isomeric 5-(phenyl-substituted)pyrazolo[4,3-e][1,4] diazepin-8-ones 3a-f were prepared and tested for their ability to bind to the benzodiazepine receptor. All compounds 3a-f display affinities for the benzodiazepine receptor in the microM range of concentration; in particular 5-phenyl-3-methyl-6,7-dihydropyrazolo[4,3-e][1,4] diazepin-8(7H)-one (3a) is 2 orders of magnitude less potent in inhibiting [3H]flunitrazepam binding than diazepam and displays an affinity for the benzodiazepine receptor practically comparable to that of its structural isomer, ripazepam, and to that of chloriazepoxide.

Pyrazolo[4,3-d]pyrimidine nucleosides. Synthesis and antiviral activity of 1-beta-D-ribofuranosyl-3-methyl-6-substituted-7H-pyrazolo[4,3-d]pyr imi din-7-ones.

N1 analogues of formycin B, with substituents at the 3 and 6 positions of the pyrazolo[4,3-d]pyrimidine moiety were synthesized by the direct SnCl4-catalyzed ribosylation method. The site of the glycosidic linkage and the anomeric configurations were established on the basis of X-ray crystallography, as well as 1H and 13C nuclear magnetic resonance spectroscopy. Preliminary results of the antiviral testing of these derivatives in vitro are described.

Azaprostaglandin analogues. Synthesis and biological properties of 11-azaprostaglandin derivatives.

New nitrogen analogues of prostaglandins (11, 11a, 12, and 12a) have been synthesized starting from a 4,5-disubstituted 2-pyrrolidinone nucleus (5 and 5a) containing one side chain and a suitable functionality for elaborating the second one. These analogues had no better activity than natural prostaglandins in vitro [guinea pig ileum and trachea, rat stomach fundus strip, uterus and portal vein, ADP-induced guinea pig platelet-rich plasma (PRP) aggregation]. They similarly lacked any interesting activity in vivo [anesthetized rat blood pressure, stress, and acetylsalycilic acid (ASA) induced gastric lesions in rat].

Carboxyl activation in peptide synthesis using 4-oximino-pyrazol-5-ones.

4-Oximino-pyrazol-5-ones 2a, b and their esters with N-protected amino acids 3a, b have been studied for carboxyl activation in peptide synthesis. 2a, b and 3a, b appear to be diastereoisomeric mixtures whose configurations have been assigned on the basis of spectroscopic and X-ray data. Some peptides obtained using these pyrazolone derivatives are reported: no racemization was noted by the Weigand test.

Synthesis and prostaglandin-like activity of 2-(trans-3-hydroxy-1-octenyl)-3-indoleheptanoic acid.

The synthesis of 2-(trans-3-hydroxy-1-octenyl)-3-indoleheptanoic acid (1) is described. The title compound appeared to show a weak prostaglandin-like activity in two different systems. It contracted rat stomach fundus strips and guinea-pig ileum preparations only at concentrations about 10(3)- and 10(2)-fold higher, respectively, than PGE1. Moreover, it stimulated adenylate cyclase from rat liver plasma membrane, but the relative potency was 4--5 X 10(2)-fold lower than the natural compound. The title compound showed also a certain degree of PGE1 antagonism.

Selective adenosine A2A receptor antagonists.

In the early 1990s it became clear that the A2A adenosine receptor had characteristics that made it distinct from the other A1, A2B and A3 adenosine receptors. Great progress has been made with the discovery of selective A2A receptor antagonists. A variety of synthetic substitutions on the xanthine moiety led the chemists of Kyowa-Hakko to discover that introduction of the styryl group in the 8 position of xanthines was critical in achieving compounds endowed with selective A2A receptor antagonistic properties. One compound, KW 6002, (E)1,3-diethyl-8-(3,4-dimethoxystyryl)-7-methylxanthine, is currently being developed for treatment of Parkinson's disease. A number of non-xanthine heterocycles have also been synthesized starting from the non-selective adenosine antagonist CGS 15943, a triazoloquinazoline. Thus, replacement of the phenyl ring of CGS 15943 with a heterocyclic ring such as pyrazole or imidazole, led to a series of interesting compounds whose prototype, SCH 58261, 7-(2-phenylethyl)-5-amino-2-(2-furyl)-pyrazolo[4,3-e]-1,2,4-triazolo[1,5-c]pyrimidine, has become a reference A2A receptor antagonist. Modification of N7 substituents has progressed to optimize A2A receptor selectivity and pharmacokinetic characteristics. A related class of compounds having a bicyclic instead of the tricyclic ring structure is also of interest. The prototype of these triazintriazolo derivatives, ZM 241385, is a potent A2A receptor antagonist; however, it also shows interactions with A2B receptors. The relevance of the A2A receptors in specific disease states, especially in the central nervous system, makes this class of adenosine receptor blockers of interest for treatment of neurodegenerative disorders such as Parkinson's disease.

DNA minor-groove binders: results and design of new antitumor agents.

DNA minor-groove binding drugs have been extensively studied in the last years in order to influence the regulation of gene expression in neoplastic disorders by means of specific interactions with DNA bases. Pyrrolo[2,1-c][1,4]benzodiazepines (PBDs), CC-1065 and distamycins are three classes of minor-groove alkylating agents which showed interesting cytotoxicity profiles, but they cannot be used in humans for various toxicity problems. For this reason many groups applied heterocyclic substitutions extensively, in order to either modify the reactivity profile or introduce extra interactions within the minor groove, thus changing the binding site or modulating the binding sequence.
