Oligotyping of celiac multiplex families with the 11th International Histocompatibility Workshop reagents.

Using PCR and SSO probes from the 11th International Histocompatibility Workshop, we oligotyped for HLA-DRB1 gene and DQA1*0501, DQB1*0201 alleles 10 celiac families each with 2 affected children. All families belong to the Italian population except for one, whose mother is originally from Cape Verde island. 8/10 sibling pairs share the DQA1*0501/B1*0201 heterodimer, inherited in cis or in trans arrangement. All the dimer-negative patients were DR4-positive.

Further researches about retinal damages and dietary protein imbalance in growing rats.

Following previous trials about hypo- and hyper-protein diets and their negative effects on some ocular structures when administered in selected stages of rats' growth, Authors have presented a comparative study of retinal alterations between light (LM) and electron (EM) microscopic analysis. LM showed a vacuolar degeneration of retinal optic fiber and ganglion cell layers where EM, performed at the same time, showed damages of the nervous tissue consisting of microtubule diminution, optic fiber molding and glial cell proliferation.

Functional and morphological abnormalities in the retina of albino rats fed with a low protein diet after weaning.

Previously, we have reported a morphological study of the retina in albino rats fed with a low protein diet. The aim of this study is to verify our previous data and to obtain further informations about morphological and functional damages of the retina following low protein diet in albino rats. For this purpose the retinas of a group of weanling albino rats fed with a hypoproteic diet were studied with Light Microscopy (LM), Electron Microscopy (EM) and Electroretinography (ERG). Our results pointed out a correlation between the anatomic and functional ocular damages and proteinic malnutrition. LM showed a vacuolar degeneration of the optic fiber layer; the EM confirmed these results showing an optic fiber cytoskeleton modification together with glial proliferation. The functional damage as well seems to be correlated to the nutritional imbalance: the scotopic ERG in the experimental group shows a significant decrease (p less than 0.001) of the b-wave amplitude.

HTLVI antibodies in multiple sclerosis and other neurological diseases.

The sera and the CSFs from 157 Multiple Sclerosis (MS) patients and 43 other neurological diseases (OND) cases have been evaluated for the presence of HTLVI antibodies. A commercial passive agglutination assay (PPA), an indirect fluorescence assay (IFA), and a Western Immunoblotting (WIB) performed in our laboratories have been employed. No OND samples showed HTLVI antibodies, while 14 sera and 1 CSF from MS patients resulted positive by PPA and 4 sera were positive with the IFA. When tested with WIB 6 MS sera showed a reactivity against one or more HTLVI proteins. Our results lead us to affirm that in a small number of MS patients, when sensitive tests are employed, it is possible to observe an antibody response toward proteins that share one or more epitope with HTLVI antigens.

Retinal and lens damages observed in young rats undergoing protein malnutrition in selected stages of their growth.

Ocular damages of young rats undergoing protein malnutrition in selected stages of their growth have been valuated. Malnutrition was induced by a purified, hypoproteinic diet given to mothers or to newborns after their weaning. The histologic results pointed out that protein lack damages ocular structures different in embryonal origin: retina and lens. In fact, vacuolar degeneration of retinal optic fiber layer and cataract of lens were observed in experimental groups of rats. Only lens damage appeared completely reversible after a prolonged nutritional rehabilitation as for rats malnourished during fetal stage and lactation and then rehabilitated for 50 days. On the contrary retinal degeneration was still present in these animals even if some improvement could be observed.

Circulating immune complexes in serum and in cerebrospinal fluid of patients with multiple sclerosis. Characterization and correlation with the clinical course.

We studied circulating immune complexes (IC) in the serum and cerebrospinal fluid (CSF) of patients with clinically defined multiple sclerosis (MS), in order to establish a correlation with the clinical course of the disease and to investigate the molecular composition of the IC isolated from patients in active phase of the disease. Serum IC levels were found to be significantly increased in patients from the progressive and active relapsing-remittent subgroups with both the CIC-conglutinin and C1q-binding methods. High levels of IC in CSF were detected only in the subgroup consisting of the relapsing-remittent patients in disease exacerbation when IC were determined by the C1q-binding test. No significant increase in serum or in CSF were found using the mRF-I test. The preliminary results of a qualitative investigation on serum IC in MS indicated that they are heterogeneous in nature, their size is mainly of the intermediate type, and they contain IgG, IgM, complement components and beta 2-microglobulins, the latter presenting an observation both new and interesting for studies on serum IC in MS patients.

Cytochemical study of larval skin in Amphibia: occurrence of ruthenium red positive cells in cutaneous gland anlagen.

During a cytochemical investigation into the development of cutaneous gland buds in amphibian larvae (both anuran and urodele), ruthenium red (RR) positive cells were found in the anlagen. These cells, with diffuse deposits of the polycation dye in their cytoplasms, were first detected on the lateral surface of still intraepidermal gland buds. Later, during segregation of anlagen from the epidermis, the reactive cells were followed whilst they converged towards the apex of the gland nest. These findings of a remarkable penetration of RR through intact plasma membranes are discussed in the light of the staining characteristics of the polycation dye. Furthermore, the mechanism regulating morphogenesis of amphibian cutaneous glands is briefly compared with the morphogenetic patterns of large exocrine glands in higher vertebrates. It is suggested that during fixation RR penetrates morphogenetically active cells, following the same pathways as calcium under physiological conditions. Calcium inflow correlates with the occurrence of thin, possibly contractile, filaments in these cells and also agrees with cell migration during morphogenesis.

Ocular alterations induced by hyperproteinic diet in growing rats.

According to literature reports about experimental damages in several organs with high metabolic rate induced by a rich animal protein diet, authors studied in rats, which had been exposed to hyperproteinic diet (casein 40%, lactalbumin 20%) since fetal life, the eyeball and external ocular muscle modifications. Histologic studies at different times have shown that in 60 day old rats a vacuolar degeneration was present in both rod and cone layer and outer nuclear layer of the retina. In 80 day old rats, together with a complete degeneration of the whole retina, external ocular muscles appeared severely fragmentated showing lipid degeneration areas along their bundles.

HTLV-I infection of cerebrospinal fluid T cells from patients with chronic neurologic disease.

Antibodies reacting with HTLV-I, the etiologic agent of acute T cell leukemia/lymphoma and a transforming agent for T4-positive lymphocytes in vitro, have recently been described in sera of patients with chronic neurologic disease in the absence of lymphoproliferative disorders. The largest number of such cases was described in Japan and in the Caribbean and parts of South America. We report here two cases of patients with chronic neurologic disease whose cerebrospinal fluid (CSF)-derived T cells contain HTLV-I specific RNA sequences and antigens and are expressing retroviral particles. Only one of these patients has demonstrable antibody to HTLV-I in serum or CSF.

The Italian Cooperative Multiple Sclerosis case-control study: preliminary results on viral antibodies.

Viral antibodies to measles, HSV-1, HSV-2, EBV and HTLB-III have been tested in 120 sera and 90 CFS of 78 MS patients, 21 other neurological diseases and 21 non neurological disease controls included in the Italian Multicenter MS case-control study. A significantly higher frequency of HSV-2 antibodies was found in the MS cases than in the two control groups. Moreover the HSV-2 and the RBV-VCA cantibody levels were higher in the MS patients than in those with non neurological diseases. A clear or weak antibody response to the HTLV-III proteins was shown, using the Western Immunoblotting method, in 10.3% of the MS sera or CSF:8.6% of the sera and 1.8% of the CSF were positive. The significance of these findings in relation to the design of the multicenter case-control study is discussed.

Longitudinal survey of T-lymphocytes and viral antibodies in MS patients.

Our study focused on the relationship between lymphocyte H/S ratios and antiviral antibodies. We have concluded a longitudinal survey of 29 clinically definite MS patients for 8 months seeking a possible correlation among clinical modifications, fluctuations of T-cell subsets and antiviral antibody titers. For this purpose in each patient the following parameters were recorded monthly:--clinical history and neurological examination;--blood T-cell subpopulation as defined by monoclonal antibodies of the OKT series;--serum antibody titers against measles, herpes simplex type 1 and 2, HTLV-1 viruses. All these data were compared with the different MS subgroups as defined by clinical course: stable, relapsing, progressive. We found that constantly normal OKT4+/OKT8+ ratios correlate with a stable clinical course; that highly fluctuating ratios parallel clinical exacerbation; that constantly high ratios are associated with a chronic-progressive course. These results are discussed with special reference to the correlation between OKT8+ lymphocyte percentages and anti-HTLV-1 antibodies, since this last virus has recently been claimed to be of pathogenetic importance in MS.

Hyper- and hypoproteic diet in growing rats: comparison between effects and evaluation of damages.

Authors have studied the effects of purified hyper- and hypoproteic diets on fetal and postnatal (not further than 40th day) stage of the rat, when cell subdivision process is faster in all the organs. Litters were grouped according to mother's and rats' (after lactation) diet. Growth of rats, brain DNA and hystology of organs were evaluated. Results of the study have shown that both diets cause alterations on growth and brain DNA and also morphological changes of several organs. Yet, it looks like major effects are produced by hypoproteic diet since it causes, apart from general damage, an irreversible delay in growing process which is not reset by normal feeding.

[Relation between serologic verification and anamnestic data on the principal virus diseases of infancy].

Serologic status of about 300 subjects (age: 2-15 years) for measles, rubella, varicella and mumps was compared with the anamnestic recall of their mothers. Measles, rubella and varicella antibodies were determined by ELISA methods. Mumps antibodies were titrated by hemagglutination-inhibition test. Serology agreed with the clinical history of the subject in 83% of the cases for measles, in 69% for rubella, in 87% for varicella and in 82% for mumps. The frequency of seronegative subjects who were reported to have had the disease was low (less than 8%) for each of the four viruses. The opposite situation was found in the 8% of the subjects for measles, in 23% for rubella, in 10% for varicella and in 13% for mumps. In general there was no significant difference in the rate of disagreements between serology and history of the disease in relation to mothers' schooling and occupation (house wives or mothers who work outside the home). Geometric mean titers of hemagglutination-inhibiting antibody to mumps virus sharply declined 3-4 years after the disease and stabilized thereafter. Prevalence of antibodies according to age shows that measles, rubella and varicella antibodies are acquired early in life, while 29% of the subjects 14-15 years of age still lacked mumps antibodies.

[HIV infection: a study of a population of non-European students].

In this study serum samples from foreign students, temporally living in Turin, coming from Asia, Central and South America and Africa, have been examined by ELISA test, to detect HIV antibodies; positive samples were confirmed by Western blot and indirect immunofluorescence assay. Particular care has been dedicated to the analysis of anamnestic data and risk factors of African students.
