Phytosterols: applications and recovery methods.

Phytosterols, or plant sterols, are compounds that occur naturally and bear close structural resemblance to cholesterol, but have different side-chain configurations. Phytosterols are relevant in pharmaceuticals (production of therapeutic steroids), nutrition (anti-cholesterol additives in functional foods, anti-cancer properties), and cosmetics (creams, lipstick). Phytosterols can be obtained from vegetable oils or from industrial wastes, which gives an added value to the latter. Considerable efforts have been recently dedicated to the development of efficient processes for phytosterol isolation from natural sources. The present work aims to summarize information on the applications of phytosterols and to review recent approaches, mainly from the industry, for the large-scale recovery of phytosterols.

Applied microbiology and biotechnology in the conservation of stone cultural heritage materials.

The contribution of applied microbiology and biotechnology for the preservation and restoration of culturally relevant stoneworks has been used only to a minor extent. Until recently it only involved the identification of the living organisms accountable for the deterioration of those materials by classic phenotypic identification methods. This seems to be changing, given the amount of work recently published that focuses in the introduction of molecular-based techniques for the detection of microorganisms in historic stone. Such techniques complement and expand the information up till now gathered by conventional identification methods. Along with this, efforts are being made to develop and implement bio-based methodologies that may actively contribute to the bioremediation of weathered historic stoneworks. The present mini-review aims to provide an overview of recent findings on these matters.

Age differences in central and peripheral intraocular pressure using a rebound tonometer.

AIM: To evaluate the influence of age on the measurements and relationships among central and peripheral intraocular pressure (IOP) readings taken with a rebound tonometer. METHODS: The IOPs were measured using the ICare rebound tonometer on the right eyes of 217 patients (88 men and 129 women) aged 18-85 years (mean 45.9 (SD 19.8) years), at the centre and at 2 mm from the nasal and temporal limbus along the horizontal meridian. Three age groups were established: young (< or =30 years old; n = 75), middle aged (31-60 years old; n = 77) and old patients (>60 years old; n = 65). RESULTS: A high correlation was found between the central and peripheral IOP readings, with the central readings being higher than the peripheral ones. Higher IOP values for the central location were found in the younger patients. Older patients had significantly lower temporal IOP readings than those for the remaining two groups (p<0.001), whereas no significant differences were found among groups when IOP was measured at the central and nasal locations. A significant decrease was observed in the nasal and temporal IOP readings as the age increased (p = 0.011 and 0.006, respectively). CONCLUSION: Older patients had lower IOP values than the middle-aged and younger patients in the temporal peripheral location. A negative correlation was found between age and IOP by rebound tonometry in the corneal periphery but not in its centre.

Extension of the resonant scattering technique to liquid crystals without resonant element.

We report X-ray resonant scattering experiments performed on the prototype liquid-crystalline compound MHPOBC doped with a chemical probe containing a resonant atom (selenium). We determined directly for the first time the microscopic 3- and 4-layer structure of the ferrielectric subphases (SmC(FI1)* and SmC(FI2)*) present in MHPOBC. Despite the low fraction of the selenium probe, the resonant signal is strong enough to allow an unambiguous determination of the basic structure of the ferrielectric subphases. These experiments demonstrate that the resonant scattering technique can be extended to liquid crystalline materials without resonant element and may stimulate new studies. A non-resonant Bragg reflection was also found in the SmC(FI1)* phase in pure MHPOBC, consistent with the 3-layer distorted model, but never detected before.

Scanning electron microscopy investigations on bis(2-ethylhexyl)phthalate treated Mycobacterium cells.

Comparative investigation of steroid transforming activity and ultrastructural changes of bis(2-ethylhexyl)phthalate (BEHP, phthalate) treated Mycobacterium sp. NRRL B-3805 cells was carried out. Transformation of beta-sitosterol into androstenedione (AD) and androstadienedione (ADD) was performed in phthalate medium by resting cells preincubated in the organic solvent for a period from 3 to 24 h. It was observed that a preincubation greater than 12 h leads to the development of dense formations on the cells surface, reduction in the cell turgor, disruption in the cell walls, and formation of zones with reduced electron density. The preincubation for 24 h causes deeper changes in the cell ultrastructure but the treated cells retain their steroid transforming activity, allowing up to 80% of the substrate to be converted into AD and ADD. A preincubation of the resting Mycobacterium cells in BEHP for 6 h might be recommended as it leads to an achievement of stoichiometrical transformation of the substrate into AD and ADD and slightly higher initial rate of the reaction performed.

Non-contact tonometry synchronized with cardiac rhythm and its relationship with blood pressure.

PURPOSE: The main objectives of this study were to determine the differences between non-synchronized intraocular pressure (IOP_N) and intraocular pressure readings synchronized with cardiac pulse and try to determine if these parameters are related to blood pressure values. METHODS: One hundred and sixty-five right eyes from 165 volunteers (107 females, 58 males) aged from 19 to 73 years (mean +/- S.D., 29.93 +/- 11.17) were examined with the Nidek NT-4000, a new non-contact tonometer that allows the measurement of IOP synchronized with the cardiac rhythm. IOP measurements in the four different modes of synchronization were taken in a randomized order. Three measures of each parameter were taken and then averaged. The blood pressure was determined three times with a portable manometer and mean values of systolic and diastolic pressure and the pulse rate were computed. Mean arterial pressure (MAP) was determined as being 1/3 of systolic plus 2/3 of diastolic blood pressure. RESULTS: The mean +/- S.D. values for the standard intraocular pressure (IOP_N: 14.76 +/- 2.86), intraocular pressure in the systolic instant or peak (IOP_P: 14.99 +/- 2.85), intraocular pressure in the middle instant between heartbeats or middle (IOP_M: 14.68 +/- 2.76), and intraocular pressure in the diastolic instant or bottom (IOP_B: 13.86 +/- 2.61) were obtained. The IOP_P was higher than the remaining values. A significant difference in mean IOP existed between IOP_B and the remaining modes of measuring (p < 0.05). Differences were statistically significant for all pair comparisons involving IOP_B. Arterial blood pressure values were systolic 125.5 +/- 14.22, diastolic 77.7 +/- 8.38 and MAP 93.64 +/- 9.44 mmHg. The pulse rate was 77.3 +/- 12.6 beats per minute. Except for the MAP (p = 0.025) there was no significant correlation between different IOP values and systolic or diastolic blood pressure, or pulse rate. CONCLUSIONS: NT-4000 is able to differentiate IOP values when synchronized with the cardiac rhythm and those differences are expected to be within a range of +/-2.5 to +/- 3.0 mmHg. IOP_B seems to be the parameter whose value differs from the non-synchronized and the remaining synchronized parameters in a significant way. Other than a weak association with MAP, no significant correlation between IOP and BP was found. The measurements of IOP readings for the three modes are consistent with timings during the cardiac cycle and IOP pulse cycle.

The influence of cycloplegia in objective refraction.

The purpose of this study was to compare refractions measured with an autorefractor and by retinoscopy with and without cycloplegia. The objective refractions were performed in 199 right eyes from 199 healthy young adults with a mean age of 21.6 +/- 2.66 years. The measurements were performed first without cycloplegia and repeated 30 min later with cycloplegia. Data were analysed using Fourier decomposition of the power profile. More negative values of component M and J(0) were given by non-cycloplegic autorefraction compared with cycloplegic autorefraction (p < 0.0001). However more positive values for the J(45) vector were given by non-cycloplegic autorefraction, although this difference was not statistically significant (p = 0.233). By retinoscopy, more negative values of component M were obtained with non-cycloplegic retinoscopy (p < 0.0001); for the cylindrical vectors J(0) and J(45) the retinoscopy without cycloplegia yields more negative values (p = 0.234; p = 0.112, respectively). Accepting that differences between cycloplegic and non-cycloplegic retinoscopy are only due to the accommodative response, the present results confirm that when performed by an experienced clinician, retinoscopy is a more reliable method to obtain the objective starting point for refraction under non-cycloplegic conditions.

Comparison of the ICare rebound tonometer with the Goldmann tonometer in a normal population.

The aim of this study was to evaluate the accuracy of measurement of intraocular pressure (IOP) using a new induction/impact rebound tonometer (ICare) in comparison with the Goldmann applanation tonometer (AT). The left eyes of 46 university students were assessed with the two tonometers, with induction tonometry being performed first. The ICare was handled by an optometrist and the Goldmann tonometer by an ophthalmologist. In this study, statistically significant differences were found when comparing the ICare rebound tonometer with applanation tonometry (AT) (p < 0.05). The mean difference between the two tonometers was 1.34 +/- 2.03 mmHg (mean +/- S.D.) and the 95% limits of agreement were +/-3.98 mmHg. A frequency distribution of the differences demonstrated that in more than 80% of cases the IOP readings differed by <3 mmHg between the ICare and the AT. In the present population the ICare overestimates the IOP value by 1.34 mmHg on average when compared with Goldmann tonometer. Nevertheless, the ICare tonometer may be helpful as a screening tool when Goldmann applanation tonometry is not applicable or not recommended, as it is able to estimate IOP within a range of +/-3.00 mmHg in more than 80% of the population.

Reduction of sludge production from WWTP using thermal pretreatment and enhanced anaerobic methanisation.

The objective of the study presented here was to investigate the performance of an enhanced two-step anaerobic process for the treatment of WWTP sludge. This process was developed to answer the urgent need currently faced by WWTP operators to reduce the production of biosolids, for which disposal pathways are facing increasing difficulties. A pilot plant was operated on a full-scale WWTP (2,500 p.e.) over a period of 4 months. It consisted of a thermal pre-treatment of excess sludge at 175 degrees C and 40 min, followed by dewatering and methanisation of the centrate in a fixed-film reactor. The thermal lysis had a two-fold enhancing effect on sludge reduction efficiency: firstly, it allowed a decrease of the HRT in the methaniser to 2.9 days and secondly, it yielded biosolids with a high dewaterability. This contributed to further reductions in the final volume of sludge to be disposed of. The two-step process achieved a sludge reduction efficiency of 65% as TSS, thus giving an interesting treatment option for WWTP facing sludge disposal problems.

Minimisation of excess sludge production in a WWTP by coupling thermal hydrolysis and rapid anaerobic digestion.

In many cases, reducing sludge production could be the solution for wastewater treatment plants (WWTP) that here difficulty evacuating the residuals of wastewater treatment. The aim of this study was to test the possibility of minimising the excess sludge production by coupling a thermal hydrolysis stage and an anaerobic digestion with a very short HRT. The tests were carried out on a 2,500 p.e. pilot plant installed on a recycling loop between the clarifier and the actived sludge basin. The line equipped with the full scale pilot plant produced 38% TSS less than the control line during a 10 week period. Moreover, the rapid anaerobic digestion removed, on average, more than 50% of the total COD load with a hydraulic retention time (HRT) of 3 days. Lastly, the dryness of the remaining excess sludge, sanitised by the thermal hydrolysis, was more than 35% with an industrial centrifuge. This combination of thermal hydrolysis and rapid anaerobic digestion equally permits a significant gain of compactness compared to traditional anaerobic digesters.

Analysis of the prescribed burning practice in the pine forest of northwestern Portugal.

The ignition of low-intensity fires in the dormant season in the pine stands of north-western Portugal seeks to reduce the existing fuel hazard without compromising site quality. The purpose of this study is to characterise this practice and assess its effectiveness, based on information resulting from the normal monitoring process at the management level, and using operational guidelines, fire behaviour models and a newly developed method to classify prescribed fire severity. Although the region's humid climate strongly constrains the activity of prescribed fire, 87% of the fires analysed were undertaken under acceptable meteorological and fuel moisture conditions. In fact, most operations achieved satisfactory results. On average, prescribed fire reduces by 96% the potential intensity of a wildfire occurring under extreme weather conditions, but 36% of the treated sites would still require heavy fire fighting resources to suppress such fire, and 17% would still carry it in the tree canopy. Only 10% of the prescribed burns have an excessive impact on trees or the forest floor, while 89% (normal fire weather) or 59% (extreme fire weather) comply with both ecological integrity maintenance and wildfire protection needs. Improved planning and monitoring procedures are recommended in order to overcome the current deficiencies.

Evaluation of the automated phoenix system for potential routine use in the clinical microbiology laboratory.

A comparative study was designed to evaluate the identification (ID) and antimicrobial susceptibility testing (AST) performances of the BD Phoenix Automated Microbiology System (Becton Dickinson Diagnostic Systems [BD], Pont de Claix, France). A total of 305 single clinical isolates were collected, and comparisons were made with routine manual methods in use in our microbiology laboratories. The percentages of correct IDs were 93.3, 89.4, 91.8, and 85.7% for enterobacteria, nonfermenting gram-negative bacilli, staphylococci, and streptococci-enterococci, respectively. The median ID time was 3 h, and the median time for AST was 10 h 30 min. AST results showed variable percentages of errors for the different antibiotics. None of the enterobacteria and 0.3% of Pseudomonas aeruginosa isolates showed a very major error (VME). Only one strain of Staphylococcus aureus showed a VME with oxacillin. We demonstrate here the efficiency of the Phoenix system, which can be used for the majority of strains encountered in a university-based laboratory, for ID and AST.

Behaviour of Mycobacterium sp. NRRL B-3805 whole cells in aqueous, organic-aqueous and organic media studied by fluorescence microscopy.

The present work aimed at quantifying the viability and morphological changes occurring during the time course of the side-chain cleavage of beta-sitosterol, in aqueous, two-phase organic-aqueous and organic media by free resting cells of Mycobacterium sp. NRRL B-3805. The solvent used was bis(2-ethylhexyl) phthalate (BEHP). A 66.3% reduction in cell viability was observed after 24 h when the cells were incubated in phosphate buffer only, but the percentage of viable cells was constant thereafter. In biphasic systems with BEHP, cell viability was maintained at higher values in the first 48 h, during which complete degradation of substrate was achieved. The availability of oxygen, which should be higher in the biphasic system than in the aqueous system, and of a carbon and energy source, thus seem important for the cells to retain their viability. In biphasic systems, cells tended to shrink and decrease their surface roughness, i.e. to decrease their surface area, possibly as a way to protect themselves from mechanical stress due to the presence of organic-aqueous interfacial forces, which resulted in disaggregation of cell clusters. A method used to visualise BEHP droplets with a standard optical microscope showed that the cells adhered to the surface of the solvent droplets, but no cells were observed inside these. In pure BEHP medium, cells retained their viability level for at least 150 h, independently of a pre-incubation period, which did not seem to induce any adaptation effect. Solvent biocompatibility, higher oxygen availability and reduced interfacial stress could have contributed to this maintenance of viability.

Isolation of a biodegradable sterol-rich fraction from industrial wastes.

Several industrial waste materials were screened for their sterol content. The possibility of using these industrial by-products as sterol sources for the microbiological production of 4-androsten-3,17-dione (AD) and 1,4-androsta-diene-3,17-dione (ADD) was investigated. Two methods of obtaining the sterol fraction from wastes were developed. Sterol-rich (96-98%) fractions were isolated in a yield above 70%, from a tall-oil effluent of a paper pulp industry and from edible-oil deodorizates. These fractions were subsequently used as a substrate for microbial degradation by a Mycobacterium sp. strain and proved to be easily converted to AD and ADD.

Timed artificial insemination in beef cattle using GnRH agonist, PGF2alpha and estradiol benzoate (EB).

The present work evaluated low-cost protocols for timed artificial insemination (TAI) in beef cattle. In Experiment 1, cycling nonlactating Nelore cows (Bos indicus, n=98) were assigned to the following groups: GnRH-PGF (GP) and GnRH-PGF-GnRH (GPG), whereas cycling (n=328, Experiment 2) or anestrus (n = 225, Experiment 3) lactating (L) cows were divided into 3 groups: GP-L, GPG-L and GnRH-PGF-Estradiol benzoate (GPE-L). In Experiment 4, lactating cows (n=201) were separated into 3 groups: GP-L, GPE-L and G/2PE-L. Animals from Experiment 1, 3 and 4 were treated (Day 0), at random stages of the estrous cycle, with 8 microg of buserelin acetate (GnRH agonist) intramuscularly (i.m.), whereas in Experiment 2 half of the cows received 8 and the other half 12 microg of GnRH (i.m.). Seven days later (D 7) all animals were treated with 25 mg of dinoprost trometamine (PGF2alpha, i.m.) except those cows from the G/2PE-L group which received only 1/2 dose of PGF2alpha (12.5 mg) via intravulvo-submucosa (i.v.s.m.). After PGF2alpha injection the animals from the control groups (GP and GP-L) were observed twice daily to detect estrus and AI was performed 12 h afterwards. The cows from the other groups received a second GnRH injection (D 8 in GPG-L and d9 in GPG groups) or one injection of estradiol benzoate (EB, 1.0 mg, D 8 in GPE-L group). All cows from GPG and GPG-L or GPE-L groups were AI 20 to 24 or 30 to 34 h, respectively, after the last hormonal injection. Pregnancy was determined by ultrasonography or rectal palpation 30 to 50 days after AI. In the control groups (GP and GP-L) percentage of animals detected in heat (44.5 to 70.3%) and pregnancy rate (20 to 42%) varied according to the number of animals with corpus luteum (CL) at the beginning of treatment. The administration of a second dose of GnRH either 24 (Experiment 2) or 48 h (Experiment 1) after PGF2alpha resulted in 47.7 and 44.9% pregnancy rates, respectively, after TAI in cycling animals. However, in anestrus cows the GPG treatment induced a much lower pregnancy rate (14.9%) after TAI. The replacement of the second dose of GnRH by EB (GPE-L) resulted in a pregnancy rate (43.3%) comparable to that obtained after GnRH treatment (GPG-L, 47.7%, Experiment 2). Furthermore, the use of 1/2 dose of PGF2alpha (12.5 mg i.v.m.s., Experiment 4) resulted in pregnancy rate (43.5%) similar to that observed with the full dose (i.m.). Both protocols GPG and GPE were effective in synchronizing ovulation in cycling Nelore cows and allowed approximately a 45% pregnancy rate after TAI. Additionally, the GPE treatment is a promising alternative to the use of GPG in timed AI of beef cattle, due to the low cost of EB when compared to GnRH agonists.

Use of danaparoid sodium (Orgaran) as an alternative to heparin sodium during cardiopulmonary bypass: a clinical evaluation of six cases.

Heparin-induced thrombocytopenia (HIT) has become more prevalent in today's cardiac setting and has resulted in the need for alternative anticoagulant therapies. Danaparoid sodium, one alternative to heparin, has been used in six cardiopulmonary bypass procedures in this hospital. This clinical experience has resulted in the progressive refinement of a protocol for the 'safe' clinical use of danaparoid sodium. Although there were six positive outcomes with the use of danaparoid sodium, alternatives must be explored in order to find the optimal anticoagulant for the treatment of HIT.

Synchronization of ovulation in crossbred dairy heifers using gonadotrophin-releasing hormone agonist, prostaglandin F2alpha and human chorionic gonadotrophin or estradiol benzoate.

Girolando (Gir x Holstein) is a very common dairy breed in Brazil because it combines the rusticity of Gir (Bos indicus) with the high milk yield of Holstein (Bos taurus). The ovarian follicular dynamics and hormonal treatments for synchronization of ovulation and timed artificial insemination were studied in Girolando heifers. The injection of a gonadotrophin-releasing hormone (GnRH) agonist was followed 6 or 7 days (d) later by prostaglandin F2alpha (PGF2alpha). Twenty-four hours after PGF2alpha injection either human chorionic gonadotropin (hCG, GPh-d6 and GPh-d7 groups) or estradiol benzoate (EB, GPE-d6 and GPE-d7 groups) was administered to synchronize ovulation and consequently allow timed artificial insemination (AI) 24 and 30 h after hCG and EB injection, respectively. Follicular dynamics in Girolando heifers was characterized by the predominance of three follicular waves (71.4%) with sizes of dominant follicles (10-13 mm) and corpus luteum (approximately 20 mm) similar to those for Bos indicus cattle. In the GnRH-PGF-hCG protocol, hCG administration induced earlier ovulation (67.4 h, P<0.01) compared to the control group (GnRH-PGF) and a better synchronization of ovulation, since most of it occurred within a period of 12 to 17 h. Pregnancy rate after timed AI was 42.8 (3/7, GPh-d6) to 50% (7/14, GPh-d7). In contrast, estradiol benzoate (GnRH-PGF-EB protocol) synchronized ovulation of only 5 of 11 heifers from the GPE-d7 group and of none (0/7) from the GPE-d6 group, which led to low pregnancy rates after timed AI (27.3 and 0%, respectively). However, since a small number of Girolando heifers was used to determine pregnancy rates in the present study, pregnancy rates should be confirmed with a larger number of animals.

Gelastic epilepsy possibly following antirabies vaccine.

A case of child with gelastic epilepsy following administration of antirabies vaccine is presented. No associated structural brain could be identified in the patient. Possibility of relationship between antirables vaccine and gelastic epilepsy is discussed.
