Microscopic lesions of spontaneous ocular blastomycosis in dogs.

Ocular tissues from 13 dogs with naturally-occurring blastomycosis were examined histologically. Choroiditis and separation of the sensory retina from its epithelium were the most prominent lesions, although all three coats of the eye were always involved. Severe panophthalmitis was associated with a marked exudative reaction within the ocular tissues. Whereas inflammation of the anterior ocular tissues was often intense, it was rarely associated with the presence of B. dermatitidis. Sequelae to the ocular inflammation included cataract, synechiae, rubeosis iridis, and closure of the drainage angle. The last of these resulted in glaucoma and resulting degeneration of the optic nerve.

Ocular angiosarcoma in four horses.

In 4 horses with ocular angiosarcomas, the common characteristics were that all horses were aged, and the neoplasms developed initially on conjunctival surfaces, grew slowly, and metastasized despite excision and radiation treatment. Histopathologically, the neoplasms were of 2 types: a capillary pattern of scant stroma and endothelial-lined vascular channels, and a solid pattern composed of sheets of pleomorphic cells with hyperchromatic nuclei and poorly defined cytoplasmic borders. Definitive diagnosis was made by use of factor VIII:RAg, which stained the tissues intensely.

Cytotoxic murine monoclonal antibody recognizing an ovine lymphocyte subpopulation similar to the human OKT4-positive set.

A cytotoxic murine immunoglobulin G2b monoclonal antibody was produced from immunization with ovine thymocytes. It reacts with a monomorphic determinant on ovine lymphocytes. This antibody 11.2 G11 does not react with B cells, lyses 50 to 60% of peripheral blood T cells, and precipitates a single chain protein with an apparent m.w. of 57,000. Its effect on mitogen- and antigen-driven lymphocyte proliferation supports its similarity in the sheep to the OKT4 antibody in humans.

Congenital feline portosystemic shunts.

Five young cats with portosystemic communications, 2 with single intrahepatic and 3 with single extrahepatic portosystemic communications, were managed surgically. One cat with a ductus venosus was treated successfully by surgery. Ptyalism and behavioral changes were similar in all 5 cats. Biochemical abnormalities included low BUN values, increased blood ammonia values, and increased sulfobromophthalein retention. None of the cats had portal hypertension at the time of surgery. Seemingly, single portosystemic shunts should be considered a surgical disease in the cat.

Development and characterization of a monoclonal antibody to class II MHC antigens in rhesus macaques.

The characterization of a monoclonal antibody with specificity for a monomorphic determinant on rhesus macaque class II antigens is described. This antibody, designated 2D16, is an IgG2b immunoglobulin which also displayed useful cross-reactivity with lymphoreticular cells and cell lines of other species including man, bonnet and stumptail macaques, sheep, dog and horse. Limited polymorphism of the 2D16 epitope was observed in the dog.

Malignant histiocytosis of Bernese mountain dogs.

A second histiocytic proliferative disorder, which resembled malignant histiocytosis of man, was identified in 13 Bernese mountain dogs. Malignant histiocytosis was clearly distinct from systemic histiocytosis, which was reported earlier in this breed. Eleven cases involved male dogs. Ten dogs occurred in the same family line as the dogs afflicted with systemic histiocytosis. Clinical or radiological evidence of pulmonary involvement was present in nine dogs. Neurological disturbances were present in five dogs. Anemia was observed in five dogs and was associated with prominent erythrophagocytosis in two instances. The clinical course was rapidly progressive. Necropsy examinations revealed that infiltrates were especially frequent in the lungs and hilar lymph nodes. Other lymph nodes, liver, spleen, and central nervous system were also frequently involved. Evidence for primary pulmonary involvement was present in seven dogs. The original diagnosis in seven cases was large cell anaplastic carcinoma of the lung by light microscopy only. The infiltrates were composed of large, pleomorphic, phagocytic mononuclear cells and multinucleated giant cells which also manifested marked cytological atypia and numerous, frequently bizarre, mitotic figures. Ultrastructural studies and the immunohistochemical demonstration of lysozyme and alpha 1-antitrypsin in the tumor cells in the majority of cases were consistent with a macrophage origin.

Hepatic lesions associated with intrahepatic arterioportal fistulae in dogs.

Seven cases of hepatic arterioportal fistulae in young dogs (mean age 6 months) are described. All cases were presumed to be congenital in origin. The onset of clinical signs, which frequently included gastrointestinal and neurological disturbances, was usually sudden. All dogs had clinical evidence of portal hypertension in the form of ascites, and all developed multiple extrahepatic portacaval venous shunts consequential to portal hypertension. The neurological disturbances were likely the result of portacaval shunting. The arterial and venous vessels involved in the fistulae had markedly altered wall structure. Hepatic regions adjacent to the fistulae frequently evidenced marked bile duct proliferation. Hepatic parenchymal atrophy, relative collapse of distributing portal veins, dilatation of hepatic arterial branches and proliferation of hepatic arterioles were seen throughout the liver; these changes closely resembled those present with portacaval shunting in the absence of hepatic arterioportal fistulae. The importance of recognizing that hepatic arterioportal fistulae and multiple extrahepatic portacaval shunts usually coexist and separately influence the morphological appearance of the liver is stressed.

Ocular angiosarcoma in the horse: morphological and immunohistochemical studies.

Angiosarcomas arising in ocular tissues of four aging horses are described. Tumors were locally invasive and eventually metastasized via the mandibular and cervical lymph nodes. Pathologically, the tumors contained well-differentiated regions, in which vascular channels were lined by pleomorphic endothelial cells, as well as poorly-differentiated regions, in which vascular channels were either rudimentary or absent. Red blood cells were scare in vascular structures formed by the tumors. Factor VIII related antigen (VIII:RAg), a blood vascular endothelial marker, was demonstrated by immunohistochemistry in tumor cells in both the well-differentiated and poorly-differentiated regions. The results suggest that ocular tumors arose in blood vascular endothelium and represented true hemangiosarcomas although a lymphatic endothelial origin could not be excluded. Immunohistochemical demonstration of VIII:RAg is a valuable adjunct to conventional stains in the diagnosis of equine vascular neoplasia.

Utilization of cytoplasmic lysozyme immunoreactivity as a histiocytic marker in canine histiocytic disorders.

Immunoreactive lysozyme was readily detectable in canine histiocytic disorders including systemic histiocytosis, malignant histiocytosis and granulomatous panniculitis. Lysozyme was less reliable as a histiocytic marker in cutaneous histiocytoma; forty percent of these tumors were negative for lysozyme expression. The marked heterogeneity in lysozyme expression in cutaneous histiocytoma may indicate that a proportion of these tumors show relatively primitive histiocytic differentiation and do not express lysozyme. Alternatively, this same proportion may exhibit a phenotype akin to cutaneous Langerhans cells which do not contain lysozyme. Lysozyme was not detectable in the tumor cells in lymphomatoid granulomatosis, atypical cutaneous histiocytoma, and histiocytic lymphosarcoma. Other evidence that these three disorders do not represent true histiocytic proliferative disorders is discussed.

Characterization of cytoplasmic lysozyme immunoreactivity as a histiocytic marker in normal canine tissues.

The immunoreactive lysozyme distribution in normal canine tissues was determined to assess the value of lysozyme as a marker of histiocytic differentiation. Lysozyme was largely confined to monocytes, neutrophils, macrophages, serous cells in mucosal associated exocrine glands, and renal proximal tubular epithelium. Macrophages in the majority of tissues stained intensely for lysozyme, although in lymphoid tissues not all acid phosphatase and nonspecific esterase positive cells contained lysozyme. In particular, dendritic antigen presenting cells, including Langerhans cells, follicular dendritic cells, and interdigitating reticulum cells, lacked immunoreactive lysozyme; hence lysozyme appeared to represent a discriminatory marker with respect to these cells. Also, a small proportion of non-dendritic macrophages appeared to lack lysozyme. It was concluded that the demonstration of immunoreactive lysozyme was a useful adjunct to conventional morphological techniques for the identification of tissue macrophages provided that due caution was exercised in the interpretation of the results of lysozyme staining.

Immunoglobulin containing cells in the tracheobronchial tree of cattle: relationship to age.

The cells of the pulmonary immune system of cattle were examined using an immunoperoxidase technique to identify cytoplasmic immunoglobulins. Cells reacting immunohistochemically with bovine IgG1, IgG2, IgA and IgM antisera were quantified in formalin-fixed paraffin embedded tissues from systematically selected sites within the tracheobronchial tree. Non-pneumonic lungs were selected for study from five groups of cattle which ranged in age from neonatal calves to aged adult cattle. Age associated differences were found. Neonatal calves lacked immunoglobulin containing cells (ICC). The numbers of ICC in 18-month-old cattle were significantly greater than in other age groups. The relative frequency of each immunoglobulin class among the cells of the airway mucosa was IgA greater than IgG1 greater than IgG2 greater than IgM for all cattle four months and older (n = 20) with an IgA to IgG1 ratio of 3.46. All airway categories from trachea to bronchiole had similar proportions of cells in each immunoglobulin class although the cell density declined in the distal airways.

Bronchus associated lymphoid tissue in the lungs of cattle: relationship to age.

The occurrence and morphological features of bronchus associated lymphoid tissue (BALT) were examined by light and electron microscopy in the non-pneumonic left lungs of five age groups of cattle from neonate to aged adult. The amount of BALT was correlated with age. It was absent in neonatal lungs and increased progressively with age until it declined in aged adult cattle. Within age groups the occurrence and morphology of BALT was variable. Widely scattered lymphoid aggregates were the predominant morphological type of BALT in the bovine lung. Follicular development in BALT foci was less frequent and quite variable within individual lungs. Morphologically distinct lymphoepithelium overlying BALT foci could not be demonstrated in non-pneumonic bovine lungs.

Systemic histiocytosis of Bernese mountain dogs.

A histiocytic proliferative disorder was identified in six closely related Bernese mountain dogs. Clinical signs included anorexia, weight loss, stertorous respiration, and conjunctivitis with marked chemosis. Multiple cutaneous nodules were distributed over the entire body but were especially prevalent in the scrotum, nasal apex, nasal planum, and eyelids. Lesions consisted of perivascular infiltrates of large histiocytes as well as minor populations of lymphocytes, neutrophils, and eosinophils. Histiocytes were further characterized by enzyme histochemistry and electron microscopy. Necropsy examinations of four dogs revealed that the histiocytic infiltrates were widespread and involved skin, lung, liver, bone marrow, spleen, lymph nodes, kidneys, testes, orbital tissues, and others. However, skin and peripheral lymph nodes were more consistently involved. The disease course was punctuated by remissions and relapses not clearly influenced by conventional therapeutic measures. Preliminary results of an experimental therapeutic regimen involving administration of bovine thymic extracts in two dogs are present. The relationship of the disorder to other human and canine histiocytic proliferative disorders is discussed.

Comparative pharmacology and clinical efficacy of newer agents in treatment of heart failure.

The animal and human pharmacology of several new drugs (prazosin, trimazosin, pirbuterol, and carbazeran) useful in the treatment of congestive heart failure (CHF) is delineated in relation to the pharmacology of other agents employed for CHF management. Prazosin and trimazosin are selective alpha 1-blockers that cause a balanced increase in cardiac output (CO) and reduction in left ventricular filling pressure (LVFP); the reduction in diastolic blood pressure with these drugs is significantly related to increase in treadmill exercise, fall in LVFP, and increase in CO. Pirbuterol is a relatively selective beta 2-agonist with somewhat greater effects on CO than on LVFP. Early promise in CHF therapy is being shown by a novel series of cyclic adenosine monophosphate (cAMP) phosphodiesterase inhibitors with combined direct inotropic and vasodilator effects. Double-blind long-term studies demonstrate persistent efficacy of prazosin and trimazosin in CHF as measured by improvement in New York Heart Association functional class, treadmill exercise performance, and noninvasive measures of cardiac function; these data are supported by studies in which repeat cardiac catheterization has been performed after several months of therapy. Double-blind studies of other CHF drugs are in progress.

Structure--activity relationships in a series of novel 3,4-dihydro-4-oxopyrimido[4,5-b]quinoline-2-carboxylic acid antiallergy agents.

A series of more than 50 new 3,4-dihydro-4-oxopyrimido[4,5-b]quinoline-2-carboxylic acid derivatives and related compounds with substituent variations at the 2, 3, and 5--9 positions was prepared and evaluated for antiallergy activity using the rat PCA assay. These compounds were obtained by the condensation of the appropriately substituted 2-aminoquinoline-3-carboxamides with dialkyl oxalates, followed by further chemical transformations. More than two-thirds of the compounds prepared exhibited intravenous activity ranging from 1 to 400 times disodium cromoglycate (DSCG). Structure--activity data suggest that the presence of a carboxylic acid moiety at the 2 position affords optimal potency and that esters are preferred for good oral absorption. Best oral activity, with ED50 values ranging from 0.3 to 3.0 mg/kg, was displayed by ethyl esters with methoxy and/or ethoxy groups at the 7 and 8 positions.

Development of ethyl 3,4-dihydro-4-oxopyrimido[4,5-b]quinoline-2-carboxylate, a new prototype with oral antiallergy activity.

Structural modification of 3,4-dihydro-4-oxoquinazoline-2-carboxylic acid leading to ethyl 3,4-dihydro-4-oxopyrimido[4,5-b]quinoline-2-carboxylate, a new prototype with oral antiallergy activity of the disodium cromoglycate type, is described. This prototype is 10 times more potent than disodium cromoglycate in the rat passive cutaneous anaphylaxis test. Structure-activity studies indicate that a carboxylic acid moiety directly attached to the 2 position of the pyrimidine ring is most favorable for intravenous activity while esters of this acid are preferred for oral activity. The oral activity of ethyl 3,4-dihydro-4-oxopyrimido[4,5-b]quinoline-2-carboxylate (ED50 = 3 mg/kg) places this ester among the more potent orally active antiallergy agents reported to date.
