Guidelines for immunoassay data processing.

These guidelines outline the minimum requirements for a data-processing package to be used in the immunoassay laboratory. They include recommendations on hardware, software, and program design. We outline the statistical analyses that should be performed to obtain the analyte concentrations of unknown specimens and to ensure adequate monitoring of within- and between-assay errors of measurement.

Length of myosin rod and its proteolytic fragments determined by electron microscopy.

The mean length of rabbit myosin chymotryptic rod in electron micrographs of unidirectionally shadowed preparations was 154 nm. Tryptic light meromyosin prepared from this rod had a mean length of 86 nm whereas long and short S2 measured 62 and 43 nm, respectively. The combined length of light meromyosin and long S2 appeared to be less than that of rod, which indicated that possibly some material was lost on digestion. Molecular lengths agreed with sedimentation equilibrium Mr values but were lower than those indicated by gel electrophoresis.

Teratogenicity of anticonvulsant drugs. II: A prospective study.

To 468 women with epilepsy enrolled in the study, 171 children were born and evaluated prospectively. The mothers in this group were characterized by low socioeconomic status, high frequency of tonic-clonic generalized seizures, and use of combinations of anticonvulsants. Approximately 30% of infants exposed to diphenylhydantoin in utero had minor craniofacial and digital changes. The infants did not show an increased rate of growth retardation, mental retardation, or major malformations. It is suggested that the nature of the association of birth defects and maternal epilepsy is complex and that the teratogenicity of anticonvulsant drugs plays only a minor role in this association. The sample size necessary to demonstrate a two- to threefold increase in malformations is beyond the capability of a single center.

Teratogenicity of anticonvulsant drugs. IV: The association of clefting and epilepsy.

Cleft lip with or without cleft palate (CL/P) is the most common congenital malformation reported among infants of epileptic mothers. This study sought to examine the relative roles of anticonvulsant teratogenicity and other factors responsible for this association. Among 175 families with a proband with isolated CL/P, there were 13 parents with epilepsy and a high frequency of other family members with CL/P and epilepsy. Evaluation of the 13 cases suggested that teratogenicity of anticonvulsant drugs was not the primary factor responsible for the observed association of maternal epilepsy and clefting. Among 140 families with a proband with clefting other than CL/P, there were no instances of parental epilepsy observed.

The use of immunohistochemical probes in the study of benign and malignant breast disease.

A review is presented of the use of immunohistochemical probes in the study of the cellular pathology of the breast. Using a combination of monoclonal and polyclonal antisera it has been possible to investigate the relationship between myoepithelial cells and basement membrane components in benign breast disease and the disturbance of this relationship in early invasive and infiltrating ductal carcinomas. The use of immunohistochemical techniques at the ultrastructural level provides the methodology for future detailed analyses. The heterogeneous expression of antigenic determinants characteristic of epithelial cells in the normal breast is considered in conjunction with its relevance to the heterogeneity seen in the invasive tumours.

The dexamethasone suppression test as a predictor of sleep deprivation antidepressant effect.

An abnormal dexamethasone suppression test (DST) result, a sensitive and specific marker for endogenous depression, was found to be associated with an antidepressant response to sleep deprivation in patients who met DSM-III criteria for Major Depressive Episode regardless of whether they met criteria for melancholia or psychotic subtypes of this disorder. These findings support previous reports of an association between an abnormal DST result and antidepressant effects of sleep deprivation in depressed patients. Our results extend the positive association between an abnormal DST result and the antidepressant response to sleep deprivation to include depressed patients who are clinically nonmelancholic during their current episode but who have an abnormal DST result.

Benign carotenaemia in children.

Six children presented with yellow pigmentation of the skin as a result of carotenaemia, and 5 were aged less than 1 year when this was first noted. Serum carotene levels depend on carotene intake, and absorption is promoted if food is of a fine texture. Five of the children were fed a partly homogenised diet. Pigmentation resolved when the carotene intake was reduced. This condition should be suspected, especially in young children, if the skin is yellow while the sclera remains a normal colour.
