Excreted factors and membrane-associated carbohydrates of Indian leishmaniae.

Antigenic identity between the cell surface carbohydrate ligands and their excreted factor (EF) in all forms of Indian leishmaniasis were assessed by 11 carbohydrate-specific lectins. Our results demonstrated that 3 lectins, viz. PNA, SBA and WFA showed unique selectivity towards skin-dwelling parasites. The EFs of visceral leishmaniasis (VL), post-kala-azar dermal leishmaniasis (PKDL) and cutaneous leishmaniasis (CL) formed band against ConA at 1000, and 500 micrograms/ml concentration, respectively. RCA-120 at 3 mg/ml concentration was positive to both Leishmania tropica and L. major, whereas SBA was specific to L. tropica, L. major and PKDL strains at the same concentration. Antisera directed against the EFs (EF-As) induced agglutination to homologous promastigotes suspension. Cross-reactivity of agglutination was observed in different strains but highest was found among PKDL and CL strains. Although PKDL cases are normally found in patients with the history of visceral leishmaniasis (kala-azar), surprisingly in this study, the surface saccharides of PKDL strains had close affinity to CL type instead of VL.

A comparison of five selective media for the primary isolation of Leishmania strains in India.

Leishmania parasites of the three distinct pathogenic forms of Indian origin (Cutaneous, visceral and post Kala-azar dermal leishmaniasis) were assessed with five selective media for their rate of primary isolation. Leishmania donovani strains showed the higher rate of isolation in modified Tobie's medium, and Leishmania tropica in Tobie's with FCS. However, strains of post Kala-azar dermal leishmaniasis (PKDL) showed 100% isolation in Schneider's with FCS medium. But as a whole modified Tobie's medium showed a higher rate of parasitic isolation for all the three pathogenic forms. In cutaneous leishmaniasis, the dry type and early lesion are also considered significant in the successful isolation of parasite. The growth study of L. donovani strain in the five selective media showed the modified Tobie's medium was best for the multiplication of the parasite.

Improved method of producing amoebic liver abscesses in hamsters for screening of systemically active amoebicides.

An improved method of producing hepatic amoebiasis in hamsters has been developed. A serially liver-passaged strain of Entamoeba histolytica from experimentally-infected hamsters was used for infection. Small pieces from the infected liver were examined for the presence of amoebae and contained approximately 20,000 active trophozoites per piece. Such liver pieces, when used as an inoculum, produced infection in all of the animals. The method has been successfully employed for screening of known amoebicides such as emetine, dehydroemetine, metronidazole, tinidazole, chloroquine, mepacrine, amodiaquine and ambilhar.

Inhibitory action of aminoglutethimide on DMBA-induced mammary carcinogenesis.

The present communication reports the inhibitory action of aminoglutethimide on 9,10-dimethyl-1,2-benzanthracene (DMBA) induced mammary carcinogenesis in virgin female Holtzman rats. When 20 mg of DMBA is given to the rats and maintained on normal diet for 32 weeks, approximately 70% of the animals develop mammary tumors. When animals similarly treated with DMBA are put on diets containing 0.025%, 0.05%, and 0.1% of aminoglutethimide for 32 weeks, there is a decline in the number of tumor-bearing animals as well as in the number of tumors per tumor-bearing animal, especially at higher diet doses of the drug. This inhibitory action on the induction of mammary tumors by DMBA could be mainly due to the suppressive action of the drug on endocrine functions.

Multilayer-enzyme linked immunosorbent assay (ML-ELISA) for detection of Entamoeba histolytica trophozoite coproantigen.

Multilayer ELISA (ML-ELISA) method for Entamoeba histolytica trophozoite coproantigen was developed for diagnosis of amoebiasis and its usefulness vis-a-vis microscopic examination analysed for paired specimens from 87 patients from Lucknow and Hyderabad, India. All the stool specimens positive for trophozoites were also found to be positive in ML-ELISA. Out of a total 32 samples positive for cysts in microscopy 21 (65.62%) gave positive reactions for coproantigen. Results on 26 samples drawn from otherwise healthy subjects with negative microscopical picture, 4 were positive for coproantigen. The results of the present study indicate that ML-ELISA test is suitable for early diagnosis of acute amoebic patients for timely treatment.

Improved preservation medium for Campylobacter jejuni.

An egg-based medium was found to be superior to the conventional Wang transport medium and the recently developed biphasic medium for the preservation of Campylobacter jejuni in the laboratory. Strains of C. jejuni preserved in egg-based medium maintained at 4 degrees C were viable for over 3 months. The survival of C. jejuni in egg-based medium held at room temperature (27 +/- 2 degrees C) was also relatively longer than in Wang transport medium and biphasic medium.

Visual and auditory evoked responses in patients with Parkinson's disease.

Visual evoked responses were obtained in 47 Parkinsonian patients and 26 age-matched controls. The stimulation to binocular and uniocular latency of the major positive peak was longer in the Parkinsonian patients and its amplitude was smaller than in the control subjects. There was a large interocular difference suggesting that at least part of the delay occurs at prechiasmatic level. The latency and amplitude varied markedly following ingestion of levodopa as Sinemet. Auditory evoked potentials were obtained in 16 Parkinsonian patients and 11 age-matched controls. The latency of the NV wave was prolonged in the former although the amplitude was the same, suggesting that the abnormality may be a widespread one.
