Characterization of Eimeria species. I. Isolation and study of pathogenicity of a pure strain of Eimeria perforans (Leuckart, 1879; Sluiter and Swellengrebel, 1912).

Eimeria perforans was isolated in pure strain by utilizing specific biological characters and specific-pathogen free rabbits. The purity of this strain was demonstrated by measurements of oocysts, derived from a line, started by infection from a single oocyst. The parasite, when inoculated in large numbers, causes a mild disease response for only about two days.

Changes in hydromineral metabolism in diarrhoeic rabbits. 2. Study of the modifications of electrolyte metabolism.

The study of the equilibrium of the main electrolytes (Na+, K+, Cl-), in plasma and intestinal content and the evolution of the partial plasma (Na+, K+, Cl-) in the young rabbit suffering from diarrhoea revealed disturbances of the mineral metabolism. Mineral losses were lower in sick animals than in controls. When the reduction of ingested matter was taken into account, the ion balance showed a deficit. The apparent coefficient of digestive utlization (aCDU) of the electrolytes Na+ and K+ dropped, although the reabsorption of these ions in the last segments of the digestive tract was generally good. The pH remained normal and the proteinaemia fell, suggesting a haemodilution. This was backed up by the presence of a hyponatraemia and a hypochloraemia, but did not account for the large drop in kaliemia. On the other hand, the plasma osmotic pressure increased, very probably in association with a high uraemia. It would therefore seem that the pathogenesis of diarrhoea in the young rabbit differed from that of other known examples, such as diarrhoea in the calf and in the infant. In the young rabbit, the pathogenesis of diarrhoea resulting from coccidiosis would seem to be, as much a result of a nutritional deficiency in particular of potassium, as of a malfunctioning of the digestive tract.

[Isolation and description of a new species of Eimeria in rabbits (Orychtolagus cuniculus) : Eimeria pellerdyi P. Coudert, 1977 (Sporozoa, Coccidia)].

Several pure isolates of Coccidia have been obtained in coccidia-free Rabbits by methods taking advantage of differences in the duration of their life-cycle and their site of development, as well as by the inoculation of experimentally immunized animals. Among these isolates one has been shown to represent a new species for which we propose the name: Eimeria pellerdyi P. Coudert, 1977. The oocysts of this species are of medium size (21 x 30 mu), without oocystic residual body. The endogenous life-cycle is completed in 10 days. Schizogony takes place in the superficial epithelial cells of the caecum and the colon, and gamogony in the depth of the Lieberkühn's crypts of the same organs. E. perllerdyi seems to be one of the most pathogenic coccidia species in Rabbits.

[Intestinal coccidiosis of the rabbitt: comparison of the pathogenic power of Eimeria intestinalis with 3 other Eimeria].

The study of the specific pathogenicity of Eimeria sp. was impossible in the absence of coccidium free Rabbits and in consequence of pure strains of Coccidia. Recently, we could realise these two conditions and started the comparative study of E. magna, E. media, E. perforans and E. intestinalis infection. E. intestinalis is characterised by a higher pathogenicity: symptoms and mortality are more important and complete recovery is rare.

A comparison of the osmotic pressures along the digestive tract of the domestic fowl and the rabbit.

1. The osmotic pressures of the contents of the alimentary tract from the adult hen and cockerel and rabbit were measured. 2. In the rabbit osmolality of the fluids from the different segments of the digestive tract was similar (331 &/- 12 mOm) and slightly hypertonic to blood plasma (297 &/- 3-12 mOsm). 3. In the hen osmotic pressures were: crop 537, gizzard 312, duodenum 571, proximal jejunum 650, distal jejunum 573, proximal ileum 514 and distal ileum 451 mOsm. Only osmolality of the crop contents was influenced by egg formation. 4. In the cockerel osmotic pressure in the alimentary tract followed the same pattern as the hen and was modified by the time of day. 5. Plasma osmolality in the fowl was about 320 mOsm. Absorption of water across the proximal jejunum is achieved therefore against a gradient of about 330 mOsm in the female and 420 mOsm in the male fowl.

Immediate postnatal sucking in the rabbit: its influence on pup survival and growth.

This study was aimed at characterising the impact of immediate postnatal sucking on pup survival and development. The interactive effects of postnatal success with the day 0 weight of pups, the nest-access regimen (controlled or free) or parity of females was investigated. Pups (n = 900) were categorised according to their initial ingestion of colostrum. In primiparous does: (1) pup mortality between d0-d10 was higher for unsuccessful than for successful early suckers; (2) lighter d0-weight reduced survival for unsuccessful but not for successful pups; (3) free nest-access of females annihilated the survival advantage fostered by the initial sucking success. In secondiparous does, these impacts waned. Finally, whatever the does' parity, only d0-weight influenced pup weight-gain between d0-21. Thus, pup survival seemed to depend (at least in primiparae) on their ability to suck right after birth, and to display a pattern of energy saving without being disturbed by the females' nest entries.

[Synthesis and evaluation of the aldose reductase inhibitory activity of new diaryl pyridazine-3-ones].

It has been possible to prepare from 4,6-diaryl pyridazinones a series of derivatives substituted in the 2-position by chains of various lengths bearing a carboxylic acid function. Pig lens aldose reductase inhibitory activity was evaluated for all compounds. N-acetic acid derivative 3c with a chlorine atom on the phenyl nucleus at the 6-position on the pyridazin ring was the most active pyridazinone with an IC50 value of 1.2 x 10(-5) M. Furthermore, it has been shown that lipophilicity and spatial configuration of the synthesized compounds took a prominent part on enzymatic activity.

Synthesis of 6-[(4-imidazol-1-yl)-phenyl]-4-phenyl 4,5-dihydro-2H-pyridazin-3-one: a potential blood platelet aggregation inhibitor.

A new 4,5 diaryl pyridazin-3-one, an analogue of imazodan and Cl-930 was prepared starting from conveniently available chalcone. The resulting derivative was tested in order to determine the area of pharmacological activity. This compound was devoid of positive inotropic effects but showed an important inhibition of PAF-acether induced blood platelet aggregation in vitro with a Cl50 value of 3,4 microM. Ex vivo, anti-aggregating effect against PAF-acether was less important with a DE50 value of 63 mg/kg per os.

[4-Arylpiperazine derivatives: a seedbed of drugs for various therapeutic indications].

The main focus of this review is the pharmacology and the therapeutic use of chemical related agents, the arylpiperazine derivatives. These compounds produce a variety of behavioural responses and pharmacological effects which directly and principally result from activation of serotonin systems. However, minor modifications in the chemical structure of these products involve important changes in affinity and selectivity for 5-HT receptors since it can also display significant affinity for dopaminergic, adrenergic or histaminergic receptors. The different arylpiperazine drugs therapeutically used are described as well as some compounds presently under investigation.
