Cloning of 87 kDa outer membrane protein gene of Pasteurella multocida P52.

Pasteurella multocida serotype B:2 is the causative agent of haemorrhagic septicaemia, a fatal disease of cattle and buffaloes. Formalin inactivated whole cell bacterin is used to prepare vaccines in India. However, outer membrane proteins (OMPS) of P. multocida were reported to be potential immunogens. The 87-kDa OMP of P. multocida P52, serotype B:2 has been identified as one of the major antigens because this protein reacted with serum of vaccinated animals. The gene omp87, encoding an 87-kDa OMP was amplified and cloned into pBluescript SK(-) vector. This gene was found to localise in a 9.0 kb Hind III fragment of P. multocida genome.

The role of neoadjuvant and adjuvant chemotherapy regimens consisting of different combinations of drugs in the treatment of advanced oral cancer.

We performed a retrospective analysis on the effect of neoadjuvant chemotherapy with three cycles of methotrexate (100 mg/m2 on day 1), cisplatin (90 mg/m2 on day 1) and bleomycin (20mg/m2 on day 1-5) with 21 d gap between each cycle in 44 patients with advanced squamous cell carcinoma of the cheek, lip and tongue followed by surgery and adjuvant chemotherapy consisting of cisplatin (90 mg/m2 on day 1), Mitomycin C (6 mg/m2 on day 1) and 5-fluorouracil (1000 mg/m2 120 h continuous infusion from day 1) repeated every 3 weeks for three cycles. Following induction chemotherapy, complete response was observed in 11 out of 44 patients (25%), and a partial response in a further 28 patients (64%). The overall median survival of all patients was 29 months and those in stage III and stage IV were 30 and 15 months respectively (P< 0.001). The median duration of the time to relapse in patients who responded to adjuvant chemotherapy was 28 months. The main toxic effect was vomiting followed by hematological toxicity. No treatment-related deaths occurred. The regimen showed a significant response, encouraging median survival and a good tolerability profile.

Effects of kappa receptor agonists on D1 and D2 dopamine agonist and antagonist-induced behaviors.

Striatal dynorphin-containing neurons receive dopaminergic inputs from the substantia nigra pars compacta and project primarily to the substantia nigra pars reticulata and entopeduncular nucleus. These neurons mainly express dopamine (DA) D1 receptors and thus dynorphin system stimulation might be expected largely to influence D1 receptor agonist or antagonist effects on motor function. It is well known the interaction existing between DA D1 and D2 drugs in the induction of behavioral effects. However, the effects of dynorphin on selective D1 and D2 DA agonist and antagonist-induced behaviors have not yet been investigated. Administration of the kappa agonists spiradoline (0.5, 1 and 5 mg/kg) or U50,488H (1, 10 and 25 mg/kg) decreased non-stereotyped grooming induced by the selective D1 agonist SKF38393. This effect was inhibited by the non-selective opioid receptor antagonist naloxone (20 mg/kg) and by the selective kappa antagonist nor-binaltorphimine (nor-BNI, 20 mg/kg). Stereotypies induced by the selective D2 agonist quinpirole were decreased by spiradoline (1 and 5 mg/kg) and by U50,488H (1, 10 and 25 mg/kg), while jerking movements of a type associated with increased D2 receptor and decreased D1 receptor stimulation emerged. Kappa agonist effects were inhibited by the prior administration of SKF38393 (10 mg/kg); these inhibitory effects were blocked by prior administration of the D1 antagonist SCH23390 (5 mg/kg). Naloxone reversed the effects of both kappa agonists on quinpirole-induced stereotypies. Kappa agonists increased D1 antagonist-induced catalepsy, but had no effect on D2 antagonist-induced catalepsy. Naloxone and nor-BNI inhibited this effect. These results suggest that the motoric effects of D1 receptor antagonists in part reflect stimulation of striatal dynorphin containing efferents.

Cloning of 2.4 kb bovine herpesvirus-1 DNA fragment containing glycoprotein III gene into pUC18 plasmid vector.

A recombinant pBR322 plasmid containing bovine herpesvirus-1 HindIII 'I' fragment was analysed using EcoRI and BamHI restriction endonucleases. This recombinant plasmid was labelled with [alpha 32P]dATP and hybridized with southern blot of HindIII digested BHV-1 DNA fragments. A 2.4 kb double digested EcoRI-BamHI fragment of HindIII 'I' was subcloned into pUC18 plasmid to get complete gIII gene. The recombinant pUC18 plasmid was analysed for 2.4 kb BHV-1 DNA insert by restriction digestion with EcoRI and BamHI. Southern blot of restriction digested plasmid was hybridized with [alpha 32P]dATP labelled BHV-1 DNA probe.

Cloning of HindIII digested bovine herpesvirus-1 DNA fragments from an Indian respiratory isolate.

The DNA from an Indian isolate of Bovine Herpesvirus-1 was isolated and analysed with restriction endonucleases. On shot gun cloning seven HindIII digested BHV-1 DNA fragments could be cloned in pBR322 vector. Recombinant clones with viral DNA insert was identified by insertional inactivation of the selection marker and restriction analysis of recombinant plasmids with HindIII. Further, recombinant plasmids were analysed with HindIII, EcoRI and BamHI restriction endonucleases to identify the different viral DNA inserts. The restriction site map of recombinant plasmids were correlated with the map reported earlier. The southern blot of restriction digested recombinant plasmids was hybridized with radio-labelled BHV-1 DNA as probe.

Idiopathic long q-t syndrome: brief case report and discussion.

Idiopathic long q-t syndrome (LQTS) is an infrequently occurring familial disorder in which affected family members have an abnormally prolonged q-tc interval with syncope, ventricular arrythmias and sudden death. In this article, we present the case of a 54-year-old female admitted for syncope, who was on no medications except for insulin. Her electrocardiogram on admission had prolonged q-tc interval (0.50 ms.). She had repeated episodes of torsades de pointes during her hospitalization, which were later controlled by beta-blockers. Electrocardiograms of her mother and daughter showed asymptomatic prolonged q-tc interval. This syndrome has an autosomal dominant pattern of transmission and it was first described by Romano and Ward in a patient with normal hearing. Our case is unusual because this condition presented so late in life.

Identification of heterogeneity in human isolates of Giardia lamblia by isoenzyme studies.

Electrophoretic mobility patterns of six enzymes, viz. alkaline phosphatase E.C. 3.1.3.1., acid phosphatase E.C. 3.1.3.2., malic enzyme E.C. 1.1.1.40., phosphoglucomutase E.C. 2.7.5.1., isocitrate dehydrogenase E.C. 1.1.1.42., glucose-6-phosphate dehydrogenase E.C. 1.1.1.49 of two axenically cultured human Giardia lamblia isolated from India (PD-1 and PD-2) and one strain from Portland, Oregon, USA (P-1) were compared using polyacrylamide gel electrophoresis (PAGE). Based on the difference in the mobility patterns of the enzymes phosphoglucomutase, isocitrate dehydrogenase and malic enzyme, the PD-1 and PD-2 isolates appeared to be quite different from P-1. In the present study, the isocitrate dehydrogenase and alkaline phosphatase enzymes were used for the first time for differentiation of Giardia isolates. In the case of PD-1, two alkaline phosphatase bands could be seen whereas only one band was observed in PD-2 and P-1. Thus, the three strains could be grouped into three different zymodemes. These findings reveal the significant heterogeneity in G. lamblia isolates both from widely separated areas and within a single region. Heterogeneity among G. lamblia strains may explain the variable clinical manifestations, host response and treatment efficacy characteristic of human giardiasis.

Prevalence of Chlamydia trachomatis in women seeking termination of pregnancy.

Fifty consecutive unmarried women seeking termination of pregnancy in Toa Payoh Hospital were analysed to determine the prevalence of Chlamydia trachomatis infection as compared to gonococcal infection. Cases harbouring chlamydial infection were followed up to exclude postabortal pelvic infection. Incidence of positive cultures were also compared among the various ethnic groups in the population. Results from the data analysed showed that chlamydial infection was significantly more prevalent than gonorrhoea in our local women seeking therapeutic abortion. However, further research to define the exact size of the problem is indicated.

Obstetric outcome of the unwed adolescents.

Two hundred and fifty-six unmarried primipara were analysed retrospectively over a five year period in the Department of Obstetrics and Gynaecology, Toa Payoh Hospital. Thirteen were omitted because of insufficient data. The patients were subdivided into an adolescent group, and those more than 18 years of age at time of booking. Maternal complications and neonatal outcome were compared between the two groups of patients. The mode of delivery, in particular the Cesarean section rate, were also compared. Cross comparison was also made among the various ethnic groups in the two categories of patients. Conclusions drawn from these data did not indicate that the adolescent unwed mothers were at higher risk than older women of similar marital status.

Chlamydia trachomatis infection in unmarried women seeking abortions.

Fifty consecutive unmarried women seeking termination of pregnancy in a state run general hospital in Singapore were screened for cervical Chlamydia trachomatis infection before abortion. Chlamydial infection was diagnosed by taking a cervical swab, culturing the organism in tissue culture media, and identifying the inclusion bodies by dark ground fluorescent microscopy. Chlamydia trachomatis was recovered in as many as 14% of cases. None of the patients gave any history suggestive of promiscuity. Compared with gonorrhoea in the non-prostitutes sexually active women of the population studied, the incidence of infection with Chlamydia trachomatis was found to be high. Patients with positive cultures often defaulted from follow up, thus posing a genuine risk of the spread of the disease by vertical and horizontal transmission.

Brain tumors and pregnancy. Presentation of a case and a review of the literature.

A case of brain tumor complicating a full-term pregnancy is reported. The literature is reviewed to show the effect of pregnancy on these tumors, the method of diagnosis, and management. Pregnancy often unmasks the existence of an intracranial neoplasm. The diagnosis can easily be missed, as the symptoms such as headache, vomiting, visual disturbance etc. are often encountered in pregnancy with or without pre-eclampsia. A high index of suspicion on the part of the obstetrician is a key to timely diagnosis. Computerized axial tomography is extremely useful in confirming or refuting the diagnosis of brain tumor. Generally speaking, neurosurgical intervention is best deferred until after delivery. In most cases, pregnancy may be allowed to continue under close supervision until the baby is reasonably mature. Labor may be induced in suitable cases, and the baby should be delivered by elective forceps as soon as the second stage of labor is reached to cut down maternal bearing-down efforts.

A double-blind controlled clinical trial of carnidazole and tinidazole in the treatment of vaginal trichomoniasis.

A double-blind controlled clinical trial of the new, not yet commercially available drug, carnidazole and tinidazole, was carried out in 77 women with a proven vaginal trichomoniasis. 39 patients and their sexual partners received a single dose of 2 g of carnidazole and the other 38 couples were treated in the same manner with tinidazole. The cure rate was 100% and 95% respectively. The side-effects were higher in the carnidazole group, although they were mild and well-tolerated by the patients and their partners. The history of the disease and its treatment are also briefly reviewed.
