[The results of two years of artificial insemination (author's transl)].

Artificial insemination donor (AID) is the solution for the sterility of many couples where the man is irreversibly sterile. We present the results of AID using frozen sperm in the two first years of work at the Centre d'Etudes et de Conservation du Sperme (C.E.C.O.S.), which is situated in the Midi-Pyrénées region of France. The results that have been obtained are satisfactory and prove the value of this method, but the means at present at the disposal of the Center do not always meet the demands made on it.

[Analysis of 830 case histories of male sterility of which 282 are usable. Results of our diagnostic investigations and therapeutic procedures].

Overall results are given of the investigation and treatment of 282 cases of male sterility: 245 cases of secretory and 37 of excretory sterility; 126 cases in which treatment produced no change, 110 cases showing biological improvement and 45 cases in which pregnancy occurred; 128 of the 282 men had varicoceles. There were three groups of patients; 1) Those with azoospermia: 59 cases, predominantly with excretory problems, bilateral atrophy and chromosomal anomalies: of these, eight showed an improvement, of which two resulted in pregnancy; 2) Those with oligo-asthenospermia: 110 cases, with predominantly solitary varicoceles, intoxications and unknown causes: 66 per 100 showed improvement, and in 23 of these cases pregnancy resulted; 3) Those with varicoceles associated with another cause, predominantly unilateral atrophy and intoxications: 62.8 percent showed biological improvement, and 12 of these cases resulted in pregnancy. Also mentioned is the group of patients with solitary asthenospermia; solitary oligospermia and hyperzoospermia of more than 200 million per ml. Varicocele is the predominant pathology in these three groups.

[Plasma concentrations and urinary excretion of gonadotropins in azoo and oligospermia. Relationship with spermatogenesis and plasma concentration of androgens (author's transl)].

Plasma concentrations and urinary excretions rate of gonadotropins, testosterone, delta 4-androstenedione, dehydroepiandrosterone and 17 and 17 alpha-hydroxyprogesterone plasma concentrations have been studied in 69 patients with oligo or azoospermia and compared with results obtained in 30 normal control subjects. A sperm count and a caryotype have been done in all patients and a testicular biopsy in 35. Analysis of the histo-hormonal correlations does not show a particular stage of spermatogenesis associated with a modification in FSH production. There is a quantitative relationship between spermatogenesis and FSH production. Plasma testosterone is low and LH production normal in excretory azoospermia. In the other patients plasma testosterone is also low but associated with an increased production of LH. Finally there is a direct relationship between urinary excretion of LH and FSH in both normal subjects and patients, suggesting a modulation by specific factors of a common regulation of the secretion of both gonadotropins.

The human EGF receptor gene: structure of the 110 kb locus and identification of sequences regulating its transcription.

The human epidermal growth factor receptor (EGFR) proto-oncogene is shown to span 110 kb of DNA divided into 26 exons. Analysis of sequences surrounding exon 1 reveals a highly CG rich region which promotes transcription. The activity of the EGF receptor promoter can be modulated by E1A protein and receptor RNA levels increased by stimulation with phorbol ester or fetal calf serum. Promoter activity is assayed by linkage to the chloramphenicol acetyl transferase gene and transfection in three cell lines, with quantitation of plasmid DNA uptake by isolation of a Hirt supernatent from each transfection. Deletion analysis of the CG rich promoter region of the gene and construction of chimeric EGFR/SV40 promoters are used to demonstrate positive transcription elements located both within exon 1 and 5' to the start of transcription. Negative regulation of transcription by sequences within a -140 to +80 region is suggested. Cotransfection experiments suggest a requirement for the interaction of DNA binding protein Sp1 for maximal activity. Finally, derepression of a positive regulatory sequence located in exon 1 during cotransfection experiments is shown. Results are discussed in reference to the multilevel regulation of EGF receptor expression.
