MET oncogene aberrant expression in canine osteosarcoma.

The objective of this study was to investigate the role of the MET oncogene in canine osteosarcoma. Seven large-breed dogs affected by spontaneous skeletal osteosarcoma underwent en bloc tumor excision. Total RNA was extracted from frozen tumor samples and assessed for expression of the MET oncogene by Northern blot analysis. Five of seven biopsy samples expressed high levels of the MET oncogene; its expression in the primary tumors was comparable with that previously identified in primary osteosarcomas in humans. A lung metastasis from one of the dogs expressed MET at a higher level than did its primary tumor. Spontaneously arising osteosarcoma in dogs clinically and pathologically mimics the corresponding disease in humans. We previously demonstrated that the MET oncogene was aberrantly expressed in a high percentage of human osteosarcomas. The results of the current study also provide a molecular parallel between the tumors in dogs and humans. This in vivo model may be helpful in evaluating new strategies for therapy against osteosarcoma.

Fast procedures for coincidence-summing correction in gamma-ray spectrometry

Simplified and fast procedures for coincidence-summing correction in gamma-ray spectrometry were investigated. These procedures are based on the usual theoretical expressions of the correction factors, but differ in the determination of the total efficiency curve based on the following approximations: (a) replacement, below the knee efficiency value, of the total efficiency by the full-energy peak efficiency; and (b) use of linear interpolations (in log-log plot) between only two experimental points above the knee efficiency value; or (c) assumption of a peak-to-total efficiency ratio independent on the counting geometry; or (d) assumption of a constant relation between the peak-to-total efficiency ratios and the photoelectric-to-total cross section ratios. The above approximations were separately assumed for determination of the coincidence-summing correction factors for nuclides with complex decay scheme (133Ba, 134Cs, 152Eu) and for 60Co and 88Y measured on a 15% relative efficiency p-type coaxial HPGe detector, for three source-detector geometries: point source placed on top of and at 10 cm from the detector window, and 1 l Marinelli beaker filled with aqueous solution. The results were compared with those based on more accurate experimental determinations of the total efficiency curve from measurements of standard sources of eight different single-gamma-ray emitters. The usefulness of each simplified procedure is evaluated with respect to its accuracy and to the reduction of the number of standard sources and measurement time.

HGF/scatter factor selectively promotes cell invasion by increasing integrin avidity.

Hepatocyte growth factor/scatter factor (HGF/SF) controls a genetic program known as 'invasive growth', which involves as critical steps cell adhesion, migration, and trespassing of basement membranes. We show here that in MDA-MB-231 carcinoma cells, these steps are elicited by HGF/SF but not by epidermal growth factor (EGF). Neither factor substantially alters the production or activity of extracellular matrix proteases. HGF/SF, but not EGF, selectively promotes cell adhesion on laminins 1 and 5, fibronectin, and vitronectin through a PI3-K-dependent mechanism. Increased adhesion is followed by enhanced invasiveness through isolated matrix proteins as well as through reconstituted basement membranes. Inhibition assays using function-blocking antibodies show that this phenomenon is mediated by multiple integrins including beta1, beta3, beta4, and beta5. HGF/SF triggers clustering of all these integrins at actin-rich adhesive sites and lamellipodia but does not quantitatively modify their membrane expression. These data suggest that HGF/SF promotes cell adhesion and invasiveness by increasing the avidity of integrins for their specific ligands.

Use of conventional taxonomy, electrophoretic karyotyping and DNA-DNA hybridization for the classification of fermentative apiculate yeasts.

A taxonomic study was conducted that considered strains of the genera Hanseniaspora/Kloeckera held in the Industrial Yeasts Collection (DBVPG) of the Dipartimento di Biologia Vegetale of the Università di Perugia, Italy. Standard phenotypic as well as molecular criteria were considered in a effort to revisit the classification of these strains, some of which have been in the collection for about 50 years. Results of salient physiological tests showed that some of the DBVPG and type strains could not be identified by current taxonomic keys. Electrophoretic karyotypes were identical for some species, with the type strains of the seven accepted species showing only five distinct chromosomal patterns. DNA-DNA hybridization analyses, using a non-radioactive dot-blot technique, allowed for the distinction of taxa. The taxonomic implications of these results are discussed.

Percutaneous treatment of life-threatening congenital arteriovenous malformations with the Wallgraft endoprosthesis.

PURPOSE: To report the percutaneous endovascular repair of massive congenital arteriovenous malformations (AVMs) of the right arm complicated by tissue ischemia, severe edema, and life-threatening recurrent hemorrhagic episodes. METHODS AND RESULTS: A 25-year-old man with a 3-year history of symptomatic upper right arm AVMs had been treated unsuccessfully with surgical ligation and coil embolization. The arm had become massively enlarged, disfigured, severely painful, and unusable, with extension of swelling to the upper chest. Under compassionate use as part of an investigational protocol, the AVMs were closed with 6 overlapping Wallgraft endoluminal prostheses deployed in the right brachial, axillary, and subclavian arteries, preserving the vertebral and distal circulations. Unfortunately, the patient suffered numerous pulmonary emboli and died 2 days after the procedure. CONCLUSIONS: Percutaneous treatment of massive AVMs appears feasible with the Wallgraft endoprosthesis, but optimal postprocedural management of such large anomalies should include immediate venous filter placement and aggressive anticoagulation therapy with intravenous heparin.

Thrombolytics and left-sided prosthetic valve thrombosis. A case report.

Prosthetic valve thrombosis is a well-known condition. Because surgical treatment of prosthetic valve thrombosis is associated with a high mortality rate, the use of thrombolysis as therapy for this condition has gained popularity in recent years. In this article, we discuss the cases of 3 patients who presented to our institution with left-sided prosthetic valve thrombosis between 1994 and 1997. All 3 patients presented with New York Heart Association functional class III or IV symptoms, and all were successfully treated with urokinase. The use of thrombolytic therapy for left-sided prosthetic valve thrombosis is associated with low mortality rates, and therefore is an attractive alternative to valve replacement or thrombectomy. However, the risk of embolic and hemorrhagic complications precludes the use of thrombolysis in patients with large thrombi and in those with New York Heart Association functional class I and II symptoms; for these patients, the risk associated with thrombolysis exceeds surgical mortality rates.

[The use of HIV-1 protease inhibitors. Their effects on AIDS-correlated outpatient and hospitalization activities. A catchment area study].

OBJECTIVE: To evaluate the effects of the use of protease inhibitors (IP) on the out-patient department's activities and hospitalizations of HIV-1 infected subjects. METHODS: From the December 1996 IP was available in our hospital, so the years 1996 and 1997 were studied. RESULTS: In the year 1997, 336 patients were visited with an increase of the 41% compared with the year 1996. In the 1997 the 45.2% of hospitalizations were related to AIDS, while in the year 1996 the 65.4% of them were related to AIDS. CONCLUSIONS: The use of IP had increased the activities of the out-patient department and certainly had a role in reducing the hospitalizations related to AIDS.

Neurocardiogenic syncope and Prinzmetal's angina associated with bronchogenic carcinoma.

A clinical case is presented illustrating a previously unreported association of (1) neurocardiogenic syncope of new onset in a 57-year-old man, (2) Prinzmetal's angina, and (3) bronchogenic carcinoma of the lung. Initiation of aggressive chemotherapy resulted in immediate suppression of both cardiac manifestations. This newly described paraneoplastic syndrome is discussed.

[Classification problems in iatrogenic hepatic traumas. 10 years of personal experience: 17 cases].

The authors propose modifying Moore's classification of hepatic trauma in an attempt to classify and emphasise iatrogenic hepatic lesions. Although fortunately rare, iatrogenic hepatic trauma are particularly severe since they occur in patients with liver pathologies or precarious general conditions and are therefore characterised by a high mortality rate: 29% in a series of 17 cases.
