[The production and reactivity of the monoclonal antibody (MCA-97) to endometrial adenocarcinoma].

Spleen cells from BALB/c mouse immunized with the human endometrial adenocarcinoma cell line (ISHIKAWA) were fused with mouse myeloma cell line (NS-1) in the presence of polyethylene glycol (Mr 1000). One monoclonal antibody, MCA-97 (IgM subclass), which showed reactivity with the ISHIKAWA-cell line, was obtained by a limiting dilution technique. In a cellular enzyme-linked immunospecific assay, the MCA-97 antibody reacted with all of 7 adenocarcinoma cell lines tested. In immunoperoxidase testing of formalin-fixed paraffin-embedded sections, the MCA-97 antibody reacted with most endometrial adenocarcinomas and endometrioid adenocarcinomas of the ovary, but did not react with squamous cell carcinomas or ovarian serous adenocarcinomas. In addition, it reacted with the glandular epithelium of normal tissues, such as proliferative endometrium, fallopian tube, uterine cervix and gastrointestinal tract. The reversed passive hemagglutination method demonstrated the antigen defined by MCA-97 in the sera of 5 out of 11 patients with endometrial carcinomas, and 3 out of 4 of those with ovarian endometrioid adenocarcinomas. It was not demonstrable in sera of most normal female volunteers, or any patients with cervical squamous cell carcinomas or ovarian serous adenocarcinomas. Thus, MCA-97 is of potential clinical application in diagnostic serology and pathology.

Dexamethasone suppresses estrogen action at the pituitary level without modulating estrogen receptor dynamics.

The administration of glucocorticoid combined with antiestrogen such as clomiphene has been shown to be effective for the induction of ovulation in patients with anovulation. The present study was undertaken to examine the effects of glucocorticoid on estrogen-induced changes in the pituitary gland. A single intraperitoneal (i.p.) administration of 10 micrograms estradiol-17 beta (E2) in ovariectomized and adrenalectomized rats resulted in a significant stimulation of pituitaries with regard to wet tissue weight and progesterone receptor content. An i.p. administration of 1 mg dexamethasone in these animals had no effects on both the values. However, the E2-induced increases in pituitary weight and progesterone receptor content were significantly inhibited by pretreatment with 1 mg of dexamethasone. The pretreatment with dexamethasone, on the other hand, had no significant effect on the dynamics of pituitary estrogen receptor induced by the injection of E2, i.e. the degree of nuclear translocation, occupancy and cytoplasmic receptor replenishment. The inhibitory effect of dexamethasone, therefore, does not seem to be mediated through estrogen receptor system in the pituitary. These results suggest that dexamethasone acts directly on the pituitary gland to suppress the action of E2, and which may be involved in the process of induction of ovulation by glucocorticoid-clomiphene treatment.

Oxytocin in the cerebrospinal fluid and plasma of pregnant and nonpregnant subjects.

The oxytocin concentration in the cerebrospinal fluid (CSF) and plasma of pregnant women at term with and without labor pain were measured by radioimmunoassay and compared with those of non-pregnant women of matched age. The oxytocin concentrations in the CSF were 4.9 +/- 4.1 microU/ml (mean +/- S.D.) in pregnant women with labor pain, 4.1 +/- 2.4 microU/ml in those without labor pain and 4.0 +/- 2.8 microU/ml in nonpregnant women, and the oxytocin concentrations in the plasma of these subjects were 45.2 +/- 19.6, 17.1 +/- 22.2 and 7.0 +/- 5.3 microU/ml, respectively. Thus the oxytocin level in the CSF did not change appreciably even when the level in the plasma was raised in the pregnant women with labor pain. These findings suggest that oxytocin does not penetrate the blood-brain barrier, and that oxytocin in the CFS has little or no central role in parturition in women.

A human sperm coating antigen isolated from sperm cell membrane.

A human sperm cell membrane antigen that is highly specific to sperm and seminal plasma was isolated from plasma membrane fraction of spermatozoa using rabbit antiserum against human seminal plasma. In addition to the high specificity to sperm and seminal plasma, the isolated antigen showed the following characteristics: (1) It is a glycoprotein of approximately 12,000 daltons that has an affinity to lentil lectin; (2) it is distributed in human milk other than in sperm and seminal plasma, but is not found in any other organs and tissues including testis; (3) seminal plasma contains the largest amount of the antigen activity, 60-fold greater than spermatozoa and 900-fold greater than milk, suggesting that this antigen could be a sperm-coating seminal plasma antigen.

Possible site of negative and positive feedback action of oestrogen on gonadotrophin secretion in normal women.

For determination of the site of action of oestrogen (E) during the negative and positive feedback phases of gonadotrophin secretions, studies were made on the pituitary response to a small amount of LRH and the pulsatility of gonadotrophins after E administration in normal cycling women in the mid-follicular phase. The pituitary responses to an iv bolus of 2.5 micrograms of synthetic LRH were evaluated by measuring serum LH and FSH 2 h before and 8 h after administration of 20 mg of conjugated E (Premarin). In the next cycle, the pituitary responses to a same dose of LHRH were also observed 2 h before and 56 h after E injection. The mean levels of serum LH and FSH and the peak responses to LRH were significantly (P less than 0.05) decreased 8 h after E injection, but were significantly (P less than 0.05) increased 56 h after E administration. In the third cycle, the pulsatility of gonadotrophins was evaluated by measuring serum LH and FSH every 15 min for 180 min before and 8 h and 56 h after E injection. The pulse frequencies of gonadotrophins were not significantly different before and 8 h and 56 h after E injection. The amplitudes of pulses 56 h after Premarin injection were significantly higher than those before the injection. These findings suggest that the negative and positive feedback effects of E on gonadotrophin secretion may be caused, in part, by its direct action on the pituitary response to LRH.

A clinicopathologic study of endometrial carcinoma with special reference to new histological variants.

The clinical and pathological features of 229 patients with endometrial carcinoma were analyzed with special reference to new histological variants. Histologically, 187 endometrial carcinomas were endometrioid form of adenocarcinoma, 10 mucinous carcinoma, 8 papillary serous carcinoma, 5 clear cell carcinoma, 1 secretory carcinoma, and 18 adenocarcinoma with squamous differentiation. Of these, papillary serous carcinoma was found to be a highly malignant form of endometrial carcinoma. Clear cell carcinoma was also associated with a poor prognosis. No fatal cases were observed in patients with mucinous carcinoma, secretory carcinoma, and adenocarcinoma with squamous differentiation. Patients with endometrioid form of adenocarcinoma were prognostically in between. Analysis of the patients with poor prognosis revealed that the length of time between onset of symptoms and surgery was not related to surviving periods and other prognostic factors such as clinical stage, histological grade, and myometrial invasion.

[Stimulatory effect of a traditional herbal medicine, Unkeito on LH-RH release].

In order to study the effect of Unkeito (Chinese name, Wen-Jan-Tang), a traditional Chinese herbal medicine and its components on luteinizing hormone-releasing hormone (LH-RH) and LH release, the mediobasal hypothalamus (MBH) alone or the pituitary alone or the pituitary in sequence with the MBH from normal female rats in diestrus was perifused in a sequential double-chamber perifusion system. LH-RH release from MBH increased significantly (p less than 0.05) by 50-100% of the basal level 30-90 min after the beginning of Unkeito administration. Unkeito at 5 micrograms/ml induced significant LH release (60-95% increase) from the pituitary in series with the MBH, but had no effect on LH release from the pituitary perifused alone. One of Unkeito's components Botanpi induced significant LH release, although other five components had no effect on LH release. These data suggest that Unkeito induces LH release from the pituitary through hypothalamic LH-RH, and can be used for the treatment of patients with hypothalamic amenorrhea.

[Tissue polypeptide antigen as a tumor marker for gynecologic malignancies].

Tissue polypeptide antigen (TPA) was measured by radioimmunoassay in sera from 413 patients with various gynecologic tumors. The distribution of positive values (greater than 102 U/l) in the patients with gynecologic malignancies was such that the percentage of patients with elevated values appeared to increase with advancing stages of disease. Among the cervical cancer patients, elevated TPA values were observed in 11% of preinvasive, 35% of stage I and 67% of advanced cases. Similarly, the TPA values were elevated in 35% of the endometrial cancer patients. Among the patients with ovarian malignancies, serum TPA was elevated in 33% of borderline, 47% of stage I and 86% of advanced cases. However, serum TPA was elevated in 21% of patients with uterine myomas and in 12% of those with ovarian benign tumors. The serial measurements of TPA in sera of the patients with gynecologic malignancies showed that serum TPA levels correlated with the effect of treatment and the clinical courses. The present observations demonstrate that the lack of tumor specificity of TPA limits its diagnostic value in gynecologic malignancies but that serial measurements of this antigen appear to be useful for monitoring of patients.
