Subacute nephrotoxicity and induction of renal cell carcinoma in mice treated with ferric nitrilotriacetate.

We investigated the induction of renal tumors by the ferric complex of nitrilotriacetic acid (Fe-NTA) in male and female A/J mice. Fifty-three male and 21 female mice received i.p. injections of Fe-NTA, 1.8 to 2.7 mg of iron/kg of body weight/day, 6 days a wk for 12 wk, at the longest. Ten male and ten female mice received nitrilotriacetic acid (NTA) i.p. at the dose equivalent to the NTA portion of Fe-NTA for the same period of time. Twenty male and 20 female mice left untreated served as the controls. Twenty-eight of the 53 Fe-NTA-treated male mice died within 14 days of the treatment. Renal proximal tubular cell necrosis was the major autopsy finding in these mice. On the other hand, all the Fe-NTA-treated female mice and NTA-treated male and female mice survived the 12 wk of treatment. Renal tubular cell carcinoma had developed in 15 of the 25 male mice and in one of the 21 female mice by the 420th day after the start of the experiment. The NTA-treated and control mice did not develop any tumors. In conclusion there is no species specificity in rats or mice in the induction of the renal carcinoma by Fe-NTA, but male mice are far more susceptible to both the acute or subacute toxicity and carcinogenic effect of Fe-NTA than are female mice.

A model for tumorigenicity and metastatic potential: growth in 1.0% agar cultures.

We developed a method to determine the amount of work performed by cells through cell division in 1.0% agar cultures. There was no correlation between the cloning efficiencies of 1.0 and 0.3% agar cultures. Growth in 1.0% agar cultures correlated well with such malignant properties as tumorigenicity and the invasive and metastatic potentials. Our method revealed that metastatic MC and F cell lines possess different means of taking advantage of energy to proliferate against an environmental pressure from those possessed by nontumorigenic (ME and T-C3H) cell strain/line or nonmetastatic but tumorigenic (L,MR, and magc1) cell lines.

An improved DAPI-DNA cytofluorometry by hypotonic cytolysis. Its application to tumor cell heterogeneity.

The effect of cell cytolysis on DAPI-DNA cytofluorometry was studied in order to eliminate nonspecific cytoplasmic hindrances in DAPI staining. A great improvement in the reproducibility of DNA determination was attained when cells were treated with hypotonic KCl (0.075 M) solution. The DNA content per cell of the mgC5 clonal line (normal diploid cell containing 2C DNA units) after hypotonic treatment was 3.07 +/- 0.06, while that of mgC5 cells without hypotonic treatment was 3.50 +/- 0.26, indicating a remarkable decrease from 7.4% to 2.0% in the coefficient of variation. In detecting tumor cell heterogeneity, our cytolytic method was superior to the method of counting the mode of chromosome number. The modal chromosome numbers of nontumorigenic m and its derivatives, mgC4 and mgC5, were 69, 68 and 67 respectively, while those of their tumorigenic counterparts, magc, mgC4V1 and mgC5V1 were 68, 66 and 67. The DNA contents of nontumorigenic m, mgC4 and mgC5 were 3.00 +/- 0.10 (mean +/- SD), 3.07 +/- 0.06 and 3.07 +/- 0.12, while those of their tumorigenic counterparts, magc, mgC4V1 and mgC5V1 were 3.67 +/- 0.15, 3.23 +/- 0.06, 3.27 +/- 0.06 respectively. A statistically significant difference in DNA content between the tumorigenic and nontumorigenic cells was observed at least at the p less than 0.05 level (t-test) when DNA content was employed for comparison, but not when the modal chromosome number was employed.(ABSTRACT TRUNCATED AT 250 WORDS)

Nephrotoxicity and renal cell carcinoma after use of iron- and aluminum-nitrilotriacetate complexes in rats.

Wistar male rats were tested for nephrotoxicity and carcinogenicity after administration of ferric nitrilotriacetate [(NTA) CAS: 139-13-9] (Fe-NTA) [No. of rats (n) = 24] and Al-NTA (n = 24). The control rats were given AlCl3 (n = 10), NTA (n = 10), and saline (n = 10). Sublethal doses of Fe-NTA [5-7 mg Fe/kg (body wt)] and Al-NTA [1.5-2.0 mg Al/kg (body wt)] were chosen and injected ip for 3 months. AlCl3 and NTA were given in equivalent doses and saline was given in equivalent volumes. All the rats given Fe-NTA or Al-NTA had a depressed weight gain, polyuria, and glucosuria from the 1st week. Histologically, acute tubular necrosis and regenerating epithelial cells were observed. Regenerative atypical epithelial cells in the renal cortex were seen at the termination of Fe-NTA or Al-NTA administration. Control rats had no remarkable changes. After 1 year, primary renal cell carcinoma and metastases to liver, lung, and peritoneum were observed only in Fe-NTA-treated rats (14 of 18 surviving rats). On the contrary, there were no tumors in Al-NTA-treated rats (none of 12 surviving rats). The results suggest that nephrotoxicity and renal cell carcinoma are two independent phenomena from Al-NTA treatment and that a long-term sublethal dose of Al-NTA is not related to renal carcinogenicity.

[Effect of PGF2 alpha on ovulation and collagenolytic activities in the rabbit treated by indomethacin].

To clarify a regulatory role of prostaglandins in collagenolytic enzyme activities of the follicular wall at ovulation, we studied whether the ovulation-blocking effect of indomethacin (IM) on hCG-stimulated rabbits could be reversed by an intravenous infusion of PGF2 alpha. The activities of BANA hydrolase and DNP peptidase were also measured by using synthetic substrates N-benzoyl-DL-arginine-2-naphthylamide HCl (BANA) and DNP-Pro-Gln-Gly-Ile-Ala-Gly-Gln-D-Arg-OH (DNP peptide), respectively, every hour during the specific preovulatory period. The ovulation-blocking effect of IM (4mg/kg) was reversed by an intravenous infusion of PGF2 alpha (1.5 mg/kg) starting at 3 hr after the hCG injection. In the control rabbits treated with hCG(100 iu) alone, these enzymatic activities were increased toward ovulation with the peak level 7-9 hr after a hCG injection and decreased significantly at 10 hr. The preovulatory increase in these enzymatic activities disappeared following the ovulation-blocking doses of indomethacin. By the addition of the PGF2 alpha infusion, the IM-induced changes in BANA hydrolase and DNP peptidase activities were returned close to the control. It is suggested that the preovulatory increases in BANA hydrolase and DNP peptidase are mandatory for ovulation and PGF2 alpha facilitates ovulation through its regulatory action on these enzymes.

Nephrotoxicity of ferric nitrilotriacetate. An electron-microscopic and metabolic study.

Repeated intraperitoneal injections of ferric nitrilotriacetate (Fe-NTA) induce nephrotoxic features such as proximal tubular necrosis and renal failure, an unexpected phenomenon for a ferric compound. The mechanism of Fe-NTA toxicity was investigated by electron microscopy and respiration studies of renal cortical mitochondria in rats. Four hours after a single intraperitoneal injection of Fe-NTA, 5 mg iron/kg body wt, loss of microvilli, increased number of cytoplasmic vacuoles, electron-dense cytoplasmic deposits, mitochondrial swelling, karyorrhexis, and rupture of cytoplasmic membrane were observed in proximal tubular epithelia. At 24 hours, an increased number of cells had become necrotic. Polarographic studies of mitochondria from renal cortex 4 hours after Fe-NTA treatment showed a significant decrease in State 3 respiration and DNP-uncoupled respiration, whereas little change was observed in State 4 respiration and ADP/O.

Biosynthesis of various types of collagen by human hepatoma cells in vitro.

A cloned human hepatoma cell line (HH2-1) produced and formed collagen fibers in vitro. The relative rate of collagen synthesis by the cells was increased with an enhancement of the cell density. An analysis of the components of the collagen using sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed that the cells synthesized interstitial collagen, types I and III, and other collagenous proteins. Thus, human hepatoma cells may play an important role in the formation of stromal collagen in the tumor.

Acute renal failure and glucosuria induced by ferric nitrilotriacetate in rats.

Nitrilotriacetate (NTA), an effective metal-chelating agent, has been used as a substitute for polyphosphates in household laundry detergents. Nephrotoxicity and renal tumorigenicity have been reported in experimental animals that received high doses of NTA po for 4 weeks to 2 years. Since NTA exists in water as a variety of NTA-metal complexes, it was important to investigate the biological effects of NTA in a complexed form. In this study, acute and subchronic toxicity of a ferric iron chelate of NTA (Fe-NTA) was investigated in rats. When Fe-NTA was given ip, acute tubular necrosis and renal failure occurred following a single injection of 15 mg iron/kg. Repeated injections of sublethal doses produced degeneration and necrosis of the proximal tubular epithelium and was associated with polyuria, glucosuria, aminoaciduria, and azotemia. After 9 days of treatment, regeneration of the tubular epithelium with atypical cells was observed. Except for a parenchymal iron deposit, no marked changes were observed in other organs. None of these effects were observed in animals given noncomplexed NTA. In conclusion, the toxicity observed following high doses of NTA given po may be the result of an absorbed metal-NTA chelate.

Mathematical models of cell variation seen in a heterogeneous malignant cell population.

We established mathematical models for the cell variation seen in a heterogeneous malignant cell population, with the supposition that it occurs as the result of the competition between two types of cells, (A) and (B), leading to a change of stem cells. Models I and II: In the case of differences in the ability of (A) and (B) cells to adapt themselves to an environment, the proportion of cells which are less adaptable to the environment decreases exponentially and eventually disappears. Model III supposes that under certain environmental conditions, the two types of cells exist simultaneously in fixed proportions, and transformations of (B) cell to (A) cell and of (A) cell to (B) cell occur at a certain rate but are independent of each other. This process is considered to follow the Markov's chain theory. Based on this supposition, we established Model III and introduced the concept of "coefficient of cell variation". We found that Model III fits the process of cell variation seen in m cell line and we calculated the coefficients of cell variation seen in this cell line in different environments. The possible mechanism of the cell variation of this cell line is discussed.

Liver, kidney, and central nervous system toxicity of aluminum given intraperitoneally to rats: a multiple-dose subchronic study using aluminum nitrilotriacetate.

In the first test, aluminum nitrilotriacetate (Al-NTA), aluminum chloride, aluminum potassium sulfate, or saline was injected ip, employing male Wistar rats. Each group consisted of ten rats. Al was given in a dose of 5 mg Al/kg body wt/day, for 14 days. Only those rats given Al-NTA showed morphological damage of the liver and kidney. Damages included diffuse midzonal coagulation necrosis of hepatocytes and acute proximal tubular necrosis of the kidney at Day 4. Seven of ten rats given Al-NTA died within 5 days. The second test was performed in metabolic cages. Al-NTA, in a dose of 1.5 to 2.0 mg Al/kg body wt/day, and NTA, of an equivalent dose, were injected ip for 54 days. Midzonal coagulation necrosis and some regenerative changes were observed in the hepatic parenchyma at Day 8. At the end of the study, complete regeneration occurred in the liver. Biochemical tests at Days 6, 13, and 28 showed high amounts of GOT, GPT, LDH, gamma-GTP, and ALP. Necrosis of proximal tubular cells of the kidney and some regeneration was noted at Day 8. Metabolic acidosis was demonstrated at Days 6, 13, and 28. Moreover, from Day 38 on, atrophy of the nerve cells of the cerebrum and demyelination of the brain stem were observed. Control rats given NTA did not exhibit any organ damage. It is concluded that a relatively small dose of Al can produce toxicosis when given with certain metal chelators.

Effects of prolyl hydroxylase inhibition on arterial collagen synthesis and blood pressure in hypertensive rats.

Changes in arterial collagen synthesis and the effects of P-1894B, a potent inhibitor of prolyl hydroxylase, were investigated on rat hypertension, induced by deoxycorticosterone acetate (DOCA) and salt. In DOCA-salt hypertensive rats, prolyl hydroxylase activity increased significantly in the abdominal aorta and in the mesenteric artery. Incorporation of 14C-proline into the thoracic aorta, abdominal aorta and mesenteric artery was higher and the hydroxyproline content of the abdominal aorta and mesenteric artery was higher than in the control rats. Treatment with P-1894B significantly inhibited prolyl hydroxylase activity, reduced arterial collagen synthesis, but did not prevent or reverse hypertension.

[Collagen synthetic activity in rabbit ovary during ovulation and its blockage by indomethacin].

In order to investigate the possibility of the participation of prostaglandins (PGs) in the collagen synthetic activity during the ovulatory process, we measured prolyl hydroxylase (PH) activity and the hydroxyproline (Hyp) content of ovarian follicles in rabbits after the administration of hCG and indomethacin (IM). The minimum dose of IM (4mg/kg) effective in blocking ovulation was given simultaneously with hCG to mature female Japanese white rabbits. PH activity in the ovary reached its peak level at 13hr after the hCG injection, immediately after ovulation. This increase was not observed in other tissues such as skin, aorta or ear obtained from ovulating rabbits. In the IM-treated rabbits in which ovulation was blocked, PH activity was rather suppressed and did not show any prominent peaks, although IM did not directly suppress PH activity in the in vitro assay system. The amount of Hyp did not show any significant changes during the ovulatory process or during ovulation blockage by IM. It is suggested that PGs play an important role in controlling collagen synthetic activity of the ovary through the ovulatory process.

[Effect of indomethacin on collagenolytic enzyme activities in rabbit ovary].

To investigate the possible participation of prostaglandins in the activation of collagenolytic enzymes of the follicular wall at ovulation, we measured the activities of neutral and acid collagenase in the rabbit ovaries at various stages of follicular development by using synthetic substrate DNP-Pro-Gln-Ile-Ala-Gly-Gln-D-Arg OH (DNP peptide) with its optimal pH 7.6 and alpha-N-benzoyl-DL-arginine-2-naphthylamide HCl (BANA) with pH 6.0, and the effect of ovulation-blocking doses of indomethacin (4 mg/kg) on DNP peptidase and BANA hydrolase activities were investigated. DNP peptidase and BANA hydrolase activities were increased toward ovulation with the highest level 7 to 9hrs after the hCG injection and then decreased significantly at 10hr. At 11hr, around the time of ovulation, the activity stayed at its low level, then rose by 13hr. Following the concomitant administration of IM with hCG, ovulation was blocked and the preovulatory increases in DNP peptidase and BANA hydrolase activities were not observed and their activities stayed at the low level until 20hr. It is suggested that collagenolytic activity for the ovulatory process started to intensiby 6hrs and ended 3hrs prior to ovulation and PGs are necessary for the enzymatic activation of DNP peptidase and BANA hydrolase.

Variations in 5'-nucleotidase activity in established mesenchymal cell lines from syngeneic A/J mice.

5'-Nucleotidase activity was analyzed in four different mesenchymal cell lines (F, m, e and SP) established from syngeneic A/J mice. The 5'-nucleotidase activity of fibroblasts was lower in transformed cells (F and m) than in nontransformed cells (e). An increase in cell contact during confluence or during high cell density increased 5'-nucleotidase activity, and a decrease in cell contact caused a decrease in 5'-nucleotidase activity in both fibroblastic (F, m and e) and reticulum (SP) cell lines. These results are evidence that 5'-nucleotidase activity in mesenchymal cells is influenced by intercellular contact as well as transformation.
