[Role of ceruloplasmin, transferrin and lipid peroxidation in bacterial infections of the CNS].

Ceruloplasmin and transferrin blood serum levels were measured during bacterial infection of the CNS. A decrease in ceruloplasmin level and a gradual increase in transferrin level were observed on the 5th day of the disease. The minimum ceruloplasmin level was observed simultaneously with the maximum MDA and GOT levels. There was a close correlation between the time course of ceruloplasmin, transferrin, MDA and GOT changes and the clinical condition of the patient.

Alterations of surface charges of plasma lipoproteins in ischemic heart disease.

For a charged and an uncharged long chain spin probe the partition between the aqueous phase and the lipoproteins LDL, HDL2 and HDL3 was measured by use of ESR spectroscopy. The partition coefficients were compared for lipoproteins from normal donors and lipoproteins from patients with ischemic heart disease. The partition coefficients of the uncharged spin probe are not different. However, the charged spin probe has a significantly different partition for LDL and HDL3. This difference results from changes in the surface charge. Patients with ischemic heart disease have LDL which is more electropositively charged and HDL3 is more electronegatively charged compared to the corresponding lipoproteins of normal subjects. The surface charge of HDL2 is not changed. The results are discussed in the light of current concepts of the pathogenesis of atherosclerosis.

[Lipid peroxidation and hyperfermentemia in bacterial lesions of the CNS].

It was found that the increase in the serum level of lipid peroxidation products in patients with meningococcal meningitis correlated with the severity of the disease. The dependence between the level of lipid peroxidation products and the activity of cell enzymes in the serum has been investigated. During the first day of the disease GOT activity was strongly increased, while the level of lipid peroxidation products was 1560 +/- 237 mumol/l, which may be of a diagnostic value.

[Lipid peroxidation and free Fe2+ during ischemia and reoxygenation of the liver].

Ischemia and reoxygenation were experimentally induced in thin liver sections. It has been shown that free iron decompartmentalization takes place 30 min after the induction of ischemia, with no lipid peroxidation activation observed. In reoxygenation, activation of lipid peroxidation and decrease in free iron concentration take place in the liver cells. It is suggested that free iron accumulation in the tissues during ischemia causes lipid peroxidation activation during reoxygenation.

[The ceruloplasmin-transferrin antioxidant system during hyperbaric oxygenation in rats].

Antioxidant system ceruloplasmin-transferrin (Cp-Tr) and malonic dialdehyde (MDA) concentration changes were studied in the rat serum after the exposure to hyperbaric oxygenation (HBO) at 4 atm for 25 min. 5 sessions of HBO led to an increase in serum MDA concentration. 5 HBO sessions were followed by the activation of Cp-Tr system. Afterwards MDA concentration began to decrease and by the 9th session even reached the initial levels. It is suggested that antioxidant system Cp-Tr takes part in the protection of the organism from toxic oxygen action.

[Changes in the surface charge of plasma lipoproteins during their auto-oxidation].

Low density lipoproteins (LDL) and high density lipoproteins (HDL) surface potential (charge) changes were studied upon autooxidation, using positively charged spin probes. Lipid peroxidation product accumulation in LDL and HDL suspensions was found to be accompanied by a significant reduction in their surface area associated with a decreased negative surface charge, and probably, deposition of lipid peroxidation polar products and/or surface charge redistribution as a result of lipoprotein autooxidative modification.

[Role of the cholesterol of platelet membranes in disorders of their structuro-functional condition in angina pectoris patients].

A study of 80 anginal patients showed their platelet molar cholesterol/phospholipid ratios, platelet membrane microviscosity, membrane release of calcium and fatty acids, and platelet aggregation capacity to be elevated. It is assumed that excessive platelet cholesterol has a significant effect on platelet aggregation, that is mediated by the intraplatelet processes examined, in anginal patients.

[Changes in the surface potential of plasma lipoproteins in ischemic heart disease. Studied with spin probes].

Changes in lipoprotein surface potentials were studied by a positively charged analog as a spin probe. Low density lipoproteins (LDL) and high density lipoproteins (subfractions HDL2 and HDL3) of patients with coronary heart disease (CHD) were studied. CHD patients have revealed a significant decrease (by 14.4 +/- 0.3 mV) in LDL and an increase (by 6.3 +/- 2.0 mV) in HDL3 negative surface potential, as compared to the control. The increase in HDL2 surface potential in CHD patients was insignificant (1.9 +/- +/- 2.5 mV). The possible role of LDL and HDL3 surface potential changes in the mechanism of interaction of these types of lipoproteins with vascular wall and blood cellular membranes and in pathogenesis of CHD and atherosclerosis is discussed.

[Lipid peroxidation and the functioning of the Ca pump of the sarcoplasmic reticulum of skeletal muscle in hypercholesterolemia].

During prolonged hypercholesterolemia (HCh), there was a reduction in the efficacy of Ca-pump function of the sarcoplasmic reticulum (SR) of rabbit skeletal muscles, an increase in cholesterol content, and intensification of lipid peroxidation in SR membranes. Supplementation of the cholesterol-rich diet with alpha-tocopherol effectively prevented dysfunction of the SR Ca-pump and reduced the enhanced LPO level in SR membranes without having any effect on cholesterol content in blood serum and SR membranes. The increase in the LPO level seems likely to be primarily responsible for abnormalities in the SR Ca-pump during HCh.

[Role of endogenous free iron in activating lipid peroxidation in ischemia].

Ischemia was simulated in rat liver perfused by physiological solution. The concentration of free iron and lipid peroxidation (LPO) products was measured 1, 2, 3, 4 and 5 hours after ischemia onset. The ESR method was used to measure free iron concentration. The LPO intensity was evaluated by the TBA test and by optical density at 232 nm. The content of free iron in cytoplasm increased in the course of ischemia with an increase in the concentration of LPO products. The content of free iron in the membranes remained unchanged. It is supposed that activation of LPO in ischemia may be caused by the appearance in the cytoplasm of a large amount of free iron. This iron can be liberated from ferritin in conditions of low oxygen concentration.

[Structural changes in erythrocyte membranes in nephropathy].

Spin probes were used to study alteration of red cell membranes in nephropathy of varying degree of gravity. Iminoxyl radicals of the lipid nature were applied as spin probes, in particular 2-(3-carboxypropyl)-4,4-dimethyl-2-tridecyl-3-oxazolidinyl (probe I). It was shown that in nephropathy, the orderliness parameter increases and the hydrophoby of probe I localized in a red cell suspension of nephropathy patients diminishes as compared with analogous parameters in healthy pregnant women. This attests to both immobilization of the fatty acid chains of phospholipids and to an increase in the polarity of the lipid bilayer in the area of probe localization. It was established that diminution of probe I hydrophoby is in a satisfactory agreement with the disease gravity and the degree of edema in patients. It was noted that alterations discovered in red cell membranes in nephropathy are similar to those seen during activation of lipid peroxidation in membranes. The possibility of lipid peroxidation involvement into the pathogenesis of nephropathy is discussed.

[Structural changes in membrane lipids of the sarcoplasmic reticulum of skeletal muscles during hypercholesterolemia].

Membrane lipid phase has been studied in the sarcoplasmic reticulum (SR) of skeletal muscles, using spin probes. Hypercholesterolemia was found to increase rotation and decrease hydrophobicity of water-soluble probe medium located inside SR vesicles. This is probably indicative of the reduction in SR vesicle membranes. At the same time reduced rotation and increased hydrophobicity and regularity of fat-acid probe micro-environment are observed in hypercholesterolemia, with the differences disappearing nearer to the centre of the membrane. It is suggested that POL activation and cholesterol accumulation in SR membranes in hypercholesterolemia lead to greater density of phospholipid molecules in the membrane.

[Role of lipid peroxidation in changes in the structure of Ca-ATPase in skeletal muscle sarcoplasmic reticulum during hypercholesterolemia].

Using the method of spin labels it was shown that in hypercholesterolemia (HCh), the following parameters decreased: the velocity of maleimide spin label binding to sarcoplasmic reticulum (SR) Ca-ATPase of rabbit skeletal muscles, the accessibility of spin-labeled thiol groups of the enzyme to potassium ferricyanide and sodium ascorbate, and the mobility of the Ca-ATPase molecule fragment to which the spin label was attached. In addition, intensification of lipid peroxidation was demonstrated in SR membranes. Supplementation of the high-cholesterol diet with alpha-tocopherol resulted in the decreased rates of lipid peroxidation in SR membranes and increased values of the above parameters relative to the values found under HCh. It is concluded that the effect of alpha-tocopherol in vivo on the structure of the Ca-ATPase proteolipid complex in HCh is due mainly to antioxidant properties of the diet-supplementing substance.
