The rice tapetum degeneration retardation gene is required for tapetum degradation and anther development.

In flowering plants, tapetum degeneration is proposed to be triggered by a programmed cell death (PCD) process during late stages of pollen development; the PCD is thought to provide cellular contents supporting pollen wall formation and to allow the subsequent pollen release. However, the molecular basis regulating tapetum PCD in plants remains poorly understood. We report the isolation and characterization of a rice (Oryza sativa) male sterile mutant tapetum degeneration retardation (tdr), which exhibits degeneration retardation of the tapetum and middle layer as well as collapse of microspores. The TDR gene is preferentially expressed in the tapetum and encodes a putative basic helix-loop-helix protein, which is likely localized to the nucleus. More importantly, two genes, Os CP1 and Os c6, encoding a Cys protease and a protease inhibitor, respectively, were shown to be the likely direct targets of TDR through chromatin immunoprecipitation analyses and the electrophoretic mobility shift assay. These results indicate that TDR is a key component of the molecular network regulating rice tapetum development and degeneration.

The immunosuppressant cyclosporin A antagonizes human formyl peptide receptor through inhibition of cognate ligand binding.

Cyclosporin A (CsA) is a fungus-derived cyclic undecapeptide with potent immunosuppressive activity. Its analog, cyclosporin H (CsH), lacks immunosuppressive function but can act as an antagonist for the human formyl peptide receptor (FPR). More recent studies have shown that CsA also inhibits fMLF-induced degranulation in differentiated HL-60 promyelocytic leukemia cells. However, it is unclear whether CsA interferes with ligand-receptor interaction, G protein activation, or other downstream signaling events. In this study we used human neutrophils, differentiated HL-60 cells, and rat basophilic leukemia (RBL)-2H3 cells expressing human FPR (RBL-FPR) to identify the action site of CsA. In functional assays, CsA inhibited fMLF-stimulated degranulation, chemotaxis, calcium mobilization, and phosphorylation of the MAPKs ERK 1/2 and the serine/threonine protein kinase Akt. CsA also blocked Trp-Lys-Tyr-Met-Val-D-Met (WKYMVm)-induced functions in RBL-FPR cells. Concentrations for half-maximal inhibition with CsA are generally 6- to 50-fold higher than that of CsH. CsA was compared with another immunosuppressant, ascomycin, relative to the inhibitory effects on FPR-mediated chemotaxis, calcium mobilization, and degranulation. In these experiments, ascomycin produced no inhibitory effects at low micromolar concentrations (1-4 microM), whereas the inhibitory effects of CsA were prominent at comparable concentrations. Finally, CsA dose-dependently inhibited the uptake of fNle-Leu-Phe-Nle-Tyr-Lys-fluoresceine and [3H]fMLF or [125I]WKYMVm binding to FPR. However, CsA and CsH did not show any obvious inhibitory effect on FPR-like 1-mediated cellular functions. These results demonstrate that CsA is a selective antagonist of FPR and that its inhibition of fMLF-stimulated leukocyte activation is at the level of cognate ligand binding.

Impact of waterborne copper on the structure of gills and hepatopancreas and its impact on the content of metallothionein in juvenile giant freshwater prawn Macrobrachium rosenbergii (Crustacea: Decapoda).

This study aims to elucidate both the impact of copper on the structure of the gills and hepatopancreas and the induction of metallothionein (MT) during waterborne copper exposure in juvenile Macrobrachium rosenbergii. Structural observations were performed with light microscopy, and the MT titrations were performed with the cadmium saturation assay. The structural changes that occurred in the gills and hepatopancreas appeared to result from copper accumulation, and the degree of damage observed in both tissues was relevant to the elevated waterborne copper concentration. Exposure to copper concentrations ranging from 0.01 mg/L to 0.4 mg/L for 7 days resulted in profound structural changes including the accumulation of hemocytes in the hemocoelic space; swelling and fusion of the lamellae; abnormal gill tips; and hyperplastic, necrotic, and clavate-globate lamellae in the gills. Similarly, hemocytic infiltration in the interstitial sinuses, an increased number of hemocytes, thickening and ruptures of the basal laminae, and necrosis of the tubules were observed in the hepatopancreas. The MT measurements showed no significant differences in MT contents between the control group and the group treated with 0.01 mg/L waterborne copper. The maximum MT content was observed at the level of 0.4 mg/L waterborne copper.

Simple and environmental friendly procedure for the gas chromatographic-mass spectrometric determination of caffeine in beverages.

A simple, quick and organic solvent saving procedure has been developed for the GC/MS determination of caffeine in beverages. The procedure involves the mixing of 25 microL sample with 1 mL ethyl acetate, and a following simple desiccation procedure in a 1.5 mL autosampler vial. A linear calibration curve was generated with caffeine concentration ranging from 0.005 mg/L to 30.0mg/L. The procedure developed provides a 0.001 mg/L detection limit of caffeine in the final solution by injection of 1 microL solution and the relative standard deviation (RSD) was less than 2% for independent measurement. The total amount of organic solvent used for individual detection is 1 mL of nontoxic ethyl acetate. The developed method was repeatable and could be applied to determine trace amounts of caffeine in popular commercial beverages.

Recombinant M. smegmatis vaccine targeted delivering IL-12/GLS into macrophages can induce specific cellular immunity against M. tuberculosis in BALB/c mice.

In the present study, we constructed a viable therapeutic vaccine of recombinant M. smegmatis mediated IL-12/GLS (granulysin) gene transfer into murine macrophages to exert the immunotherapy effects on the Mycobacterium tuberculosis infection. We tested this recombinant therapeutic vaccine in an in vivo study to determine its capability of stimulating host specific immune responses against M. tuberculosis. BALB/c mice intranasally immunized with the therapeutic vaccine developed an efficient Th1 protective immune response against M. tuberculosis which was equal to that of the BCG strain. Inoculation intranasally with this viable vaccine induced high level of serum IFN-gamma, IL-12 and IgG2a. The viable vaccine was capable of inducing purified protein derivative (PPD) antigen-specific splenocytes proliferation and IFN-gamma production from T cells in spleens of the immunized mice. In addition, intranasally inoculation with the viable vaccine can induce PPD antigen-specific sIgA production in the broncho-alveolar lavage fluid (BALF) of the immunized mice. No change of IL-4 level was found in all groups. The therapeutic mechanism of this viable vaccine against M. tuberculosis infection observed here appeared to be a result of the specific Th1 immune response activated by mycobacterium antigen from M. smegmatis and the expression of sIL-12/GLS in alveolar macrophages via the M. smegmatis-mediated gene transfer method. This research demonstrates that the therapeutic gene can be introduced into a host by viable mycobacteria works to induce the host specific immune response against M. tuberculosis infection in vivo. Since this therapeutic vaccine can strongly induce specific Th1 responses against M. tuberculosis in BALB/c mice and has no obviously harmfulness to the host simultaneously, the recombinant vaccine might be a potential candidate therapeutic vaccine against tuberculosis.

[Effect of lung stress index on titration of positive end-expiratory pressure at post-recruitment in three canine acute respiratory distress syndrome models].

OBJECTIVE: To investigate the relationship of lung stress index and positive end-expiratory pressure (PEEP) at post-recruitment in different canine acute respiratory distress syndrome (ARDS) models. METHODS: The ARDS models were induced by intravenous oleic acid, saline lavage and hydrochloric acid aspiration in anesthetized dogs. During volume control ventilation with constant inspiratory flow, PEEP was set to obtain a b (stress index) value between 0.9 and 1.1 (b = 1) before and post recruitment maneuver (RM). PEEP was changed to obtain b < 1 (0.6 < b < 0.8) and b > 1 (1.1 < b < 1.3). Meanwhile, the recruited volume (RV) was measured and pulmonary mechanics and gas exchange were observed. RESULTS: At b = 1 after RM, PEEP were (10.8 +/- 2.3), (12.8 +/- 1.8) and (9.2 +/- 1.8) cm H2O in the oleic acid, saline-lavaged and hydrochloric acid aspiration groups, respectively. PEEP in saline-lavaged group was higher than that in hydrochloric acid aspiration group (P < 0.05). The ratio of partial arterial oxygen tension and fraction of inspiratory oxygen (PaO(2)/FiO(2)) at b = 1 without RM was lower than those post-RM in all three groups (P < 0.05). In oleic acid group, PaO(2)/FiO(2) at b = 1 post-RM was (399 +/- 61) mm Hg, which was higher than that at b < 1 [(307 +/- 71) mm Hg], but there was no difference between those at b = 1 and b > 1. At b = 1 after RM, PaO(2)/FiO(2) in the saline-lavaged group was higher than that in acid aspiration group, but no difference between saline-lavaged group and oleic acid group was found. At b = 1 post-RM, RV were higher than that at b = 1 before RM in all three groups (P < 0.01), but there was no significant difference among three groups. At b = 1 post-RM in three groups, pulmonary compliance were higher than those at b > 1, but airway plateau pressure were lower than those at b > 1. CONCLUSIONS: Lung stress index could be a good indicator for PEEP titration at post-RM.

A Semliki Forest virus replicon vectored DNA vaccine expressing the E2 glycoprotein of classical swine fever virus protects pigs from lethal challenge.

Classical swine fever virus (CSFV) causes significant losses in pig industry in many countries in Asia and Europe. The E2 glycoprotein of CSFV is the main target for neutralizing antibodies. Recently, the replicon of alphaviruses, such as Semliki Forest virus (SFV), has been developed as replicative expression vectors for gene delivery. In this study, we constructed a plasmid DNA based on SFV replicon encoding the E2 glycoprotein of CSFV and evaluated its efficacy in rabbits and pigs. The results showed that the animals immunized with the DNA vaccine developed CSFV-specific neutralizing antibodies and were protected from virulent or lethal challenge. This demonstrates that the SFV replicon-derived DNA vaccine can be a potential marker vaccine against CSFV infections.

Toll-like receptor 3 is an essential component of the innate stress response in virus-induced cardiac injury.

Enterovirus-induced myocardial injury can lead to severe heart failure. To date, little is known about the early innate stress response that contributes to host defense in the heart. Toll-like receptor 3 (TLR3) is important in the initiation of the innate antiviral response. We investigated the involvement of TLR3, which recognizes viral double-stranded RNA, on encephalomyocarditis virus (EMCV) infection. To examine the contribution of TLR3 in protection from EMCV infection, we infected mice deficient in TLR3 with 50 plaque-forming units of EMCV. TLR3-deficient (TLR3(-/-)) mice were more susceptible to EMCV infection and had a significantly higher viral load in the heart compared with TLR3(+/+) mice. Histopathological examination showed that the inflammatory changes of the myocardium were less marked in TLR3(-/-) than in TLR3(+/+)mice. TLR3(-/-) mice had impaired proinflammatory cytokine and chemokine expression in the heart following EMCV infection. However, the expression of interferon-beta was not impaired in EMCV-infected TLR3(-/-) mice. EMCV infection leads to a TLR3-dependent innate stress response, which is involved in mediating protection against virus-induced myocardial injury.

Shape-specific detection based on fluorescence resonance energy transfer using a flexible water-soluble conjugated polymer.

We present the detection of the shape-specific conformation of DNA based on the fluorescence resonance energy transfer (FRET) by using a novel flexible water-soluble cationic conjugated polymer (CCP). The flexible backbone of CCP has more conformational freedom with the potential to be responsive to analyte shape by electrostatic interaction between flexible CCP and negatively charged DNA. The analyte shape dependent recognition is accomplished by structural changes that compressed or extended the flexible CCP. The morphology-dependent spectral properties of the novel flexible polymer related to the analyte shapes are investigated in detail, where two types of chromophores, referred to as "isolated" segment and "packed" segment aggregates, within the flexible polymer are identified by means of ensemble and single molecule measurements upon binding with different geometric DNA. The change in fluorescence intensity upon binding with shape-specific DNA without obvious color shifts makes this novel flexible polymer a suitable CCP donor for FRET measurements. The results provide insights for understanding the spectral properties of flexible water-soluble CCP and CCP/DNA interaction related to the geometry of target analyte.

Elevation of serum l-lactate dehydrogenase B correlated with the clinical stage of lung cancer.

To identify potential biomarkers related with lung cancer metastasis, conditional media (CM) proteins collected from a primary non-small cell lung cancer (NSCLC) cell line NCI-H226 and its brain metastatic subline H226Br were analyzed by one-dimensional electrophoresis (1-D PAGE) and matrix-assisted laser desorption/time of flight mass spectrometry (MALDI-TOF-MS). Twelve biomarkers were identified, of which l-lactate dehydrogenase B (LDHB) chain was significantly up-regulated in the CM of H226Br cell and was further validated in 105 lung cancer, 93 non-lung cancer, 41 benign lung disease, as well as 65 healthy individuals sera using enzyme-linked immunosorbent assay (ELISA). It was found that the levels of LDHB were specifically elevated in NSCLC sera compared with other groups and were progressively increased with the clinical stage. At the cutoff point 0.260 (OD value) on the receiver operating characteristic (ROC) curve, LDHB could comparatively discriminate lung cancer from benign lung disease and healthy control groups with sensitivity 81%, specificity 70% and total accuracy 76%. These findings demonstrated that secretome could open up a possibility to find, identify, and characterize novel biomarkers related with invasion and metastasis.

Single nucleotide polymorphism C677T in the methylenetetrahydrofolate reductase gene might be a genetic risk factor for infertility for Chinese men with azoospermia or severe oligozoospermia.

AIM: To analyze the distribution of the single nucleotide polymorphism (SNP) C677T in the methylenetetrahydrofolate reductase (MTHFR) gene in 355 infertile Chinese patients with idiopathic azoospermia or severe oligozoospermia and 252 fertile Chinese men as controls to explore the possible association of the SNP and male infertility. METHODS: Using the polymerase chain reaction (PCR)-restriction fragment length polymorphism technique, the allele and genotype distribution of SNP C677T in the MTHFR gene were investigated in both patients and controls. RESULTS: The frequencies of allele T (40.9% vs 30.4%, P = 0.002, odds ration [OR] = 1.58, 95% confidence interval [CI]: 1.24-2.02) and mutant homozygote (TT) (18.3% vs. 11.5%, P=0.023, OR=1.72, 95% CI: 1.07-2.76) as well as carrier with allele (TT + CT) (63.4% vs. 49.2%, P = 0.0005, OR = 1.79, 95% CI: 1.29-2.48) in infertile patients were significantly higher than those in controls. After patient stratification, the significant differences in distribution of the SNP between each patient subgroup and control group still remained. CONCLUSION: Our findings indicate that there is an association of SNP C677T in the MTHFR gene with male infertility, suggesting that this polymorphism might be a genetic risk factor for male infertility in Chinese men.

Inclusion complexes of beta-cyclodextrin with ionic liquid surfactants.

The three kinds of ionic liquid (IL) surfactants, 1-dodecyl-3-methylimidazolium hexafluorophosphate (C(12)mimPF(6)), 1-tetradecyl-3-methylimidazolium hexafluorophosphate (C(14)mimPF(6)), and 1-hexadecyl-3-methylimidazolium hexafluorophosphate (C(16)mimPF(6)), were used to form the inclusion complexes (ICs) with beta-cyclodextrin (beta-CD). The surface tension measurements revealed that there were two kinds of inclusion formations, 1:1 and 1:2 (beta-CD/IL) stoichiometry for beta-CD-C(12)mimPF(6) and beta-CD-C(14)mimPF(6) ICs, and only 1:1 stoichiometry for beta-CD-C(16)mimPF(6) ICs. These inclusion compounds were further characterized by XRD, (13)C CP/MAS NMR, (1)H NMR, rotating frame nuclear Overhauser effect spectroscopy (ROESY), and thermogravimetry (TGA). The results showed that these ICs were fine crystalline powder. The host-guest system presented a channel-type structure, and each glucose unit of beta-CD was in a similar environment. It was suggested that hydrophobicity played a crucial role in supporting the formation of ICs. The decomposition temperature of these ICs was lower than those of their precursors. Furthermore, the possible inclusion structures were also speculated. These inclusion behaviors are likely to be used to recover ILs in the process of their preparation.

The presenilin-2 loop peptide perturbs intracellular Ca2+ homeostasis and accelerates apoptosis.

In cells undergoing apoptosis, a 22-amino-acid presenilin-2-loop peptide (PS2-LP, amino acids 308-329 in presenilin-2) is generated through cleavage of the carboxyl-terminal fragment of presenilin-2 by caspase-3. The impact of PS2-LP on the progression of apoptosis, however, is not known. Here we show that PS2-LP is a potent inducer of the mitochondrial-dependent cell death pathway when transduced as a fusion protein with HIV-TAT. Biochemical and functional studies demonstrate that TAT-PS2-LP can interact with the inositol 1,4,5-trisphosphate receptor and activate Ca(2+) release from the endoplasmic reticulum. These results indicate that PS2-LP-mediated alteration of intracellular Ca(2+) homeostasis may be linked to the acceleration of apoptosis. Therefore, targeting the function of PS2-LP could provide a useful therapeutic tool for the treatment of cancer and degenerative diseases.

Autogenous coronoid process as a new donor source for reconstruction of mandibular condyle: an experimental study on goats.

OBJECTIVE: To evaluate the feasibility and effectiveness of autogenous coronoid process grafts for reconstruction of the mandibular condyle. STUDY DESIGN: Two groups of 9 goats each were used. Each animal underwent unilateral total condyle and disk removal and was treated by grafting an autogeneous coronoid process. The animals in group A were sacrificed at 4, 12, and 24 weeks after surgical procedure, and their grafted coronoid processes were harvested for histological observation. All animals in group B received 3-dimensional CT scanning examination at different times and were sacrificed at 24 weeks after surgical procedure. The items of ramus height (RH), transverse dimension of the condyle (CT), and posteroanterior dimension of the condyle (CP) were measured for comparison with the operated and nonoperated sides. RESULTS: Despite the differences in the measurements of RH, CT, and CP between the operated and nonoperated sides, the grafted coronoid process had a similar shape and histological structures to the normal condyle at 24 weeks after surgical procedure. The head of the neocondyle was covered by a cap of fibrous tissue, which might play the role of the articular disc. CONCLUSIONS: Under the mechanical stimuli of the temporomandibular joint site, the grafted coronoid process could gradually remodel to a neocondyle with functional shape and structure in goats. This animal study suggests that autogenous coronoid process could be considered as a new donor source for reconstruction of the mandibular condyle.

Studies on the stability of the chloramphenicol in the microemulsion free of alcohols.

Microemulsion composed of Span20 + Tween20 isopropyl myristate (IPM) + H2O were investigated as potential drug delivery systems for eye drops. The system is important in that all its components are food grade so that the microemulsion is almost free of toxicity and irritation. The phase transition was investigated using the electrical conductivity measurements. The chloramphenicol is used to treat the eye diseases such as trachoma and keratitis. However, this drug in the common eye drops hydrolyzes easily. The main product of the hydrolysis is glycol. Here, the chloramphenicol was trapped into the oil-in-water (o/w) microemulsions free of alcohols. Its stability was investigated by the high performance liquid chromatography (HPLC) assays in the accelerated experiments of 3 months. The location of the chloramphenicol molecules in the microemulsion formulations was determined by means of dynamic light scattering (DLS) and 1H NMR spectroscopy. The results of HPLC revealed that the content of the glycols in the microemulsion formulation was much lower than that in the commercial eye drops at the end of the accelerated experiments. It implied that the stability of the chloramphenicol in the microemulsion formulations was increased remarkably. The results of DLS and NMR confirmed that the chloramphenicol molecules should be trapped into the hydrophilic shells of the microemulsion drops, which were composed of many oxyethylene groups. The benzene rings of the chloramphenicol molecules were near the group of alpha2-CH2 and the oxyethylene groups of the surfactant molecules. It was this reason that enabled the chloramphenicol molecules in the microemulsions to be screened from the bulk water and its stability to be increased remarkably.
