A highly sensitive and specific sandwich enzyme immunoassay for water soluble A and B substances.

A highly sensitive and specific sandwich enzyme immunoassay for water soluble A and B substances is described. An affinity-purified rabbit anti-A IgG-coated polystyrene ball was incubated with water soluble A substance and subsequently with affinity-purified rabbit anti-A IgG-horseradish peroxidase conjugate. In addition, an affinity-purified goat anti-B IgG-coated polystyrene ball was incubated with water soluble B substance and subsequently with affinity-purified goat anti-B IgG-horseradish peroxidase conjugate. Peroxidase activity bound to the polystyrene balls was assayed by fluorimetry. The detection limits of artificial A and B antigens (synthetic A and B trisaccharides coupled to bovine serum albumin) were both 0.1 fmol/tube, which were 30- and 300-fold lower, respectively, than those by the hemagglutination inhibition method. In addition, A and B substances in secretor saliva could also be measured 3- to 10-fold and 100- to 1,000-fold more sensitively, respectively, by the present method than by the hemagglutination inhibition method.

Capillary tube isotachophoresis of proteins in early postmortem serum, cerebrospinal fluid and pericardial fluid.

Serum, cerebrospinal fluid (CSF) and pericardial fluid (PDF) obtained from 22 early cadavers were subjected to capillary tube isotachophoresis. Separated proteins were grouped into three main fractions, namely, Fr. 1, Fr. 2 and Fr. 3, in order of their decreasing effective (net) mobilities. The percentages of main fractions to total content were calculated by UV absorbance integration. It was found that Fr. 1 decreased and Fr. 2 increased in all three kinds of specimens with an increase in time following death. In serum and also in CSF, the degree of these changes between 6 and 58 hours following death seemed large enough (about 30%) to be used for crude estimations of early postmortem intervals. Changes in PDF proceeded more gradually than in serum and or in CSF, with only 5-20% changes during this period.

A highly specific and sensitive sandwich enzyme immunoassay for human hemoglobin A.

A highly specific and sensitive sandwich enzyme immunoassay for human hemoglobin A (Hb A) is described. A rabbit anti-human Hb A IgG-coated polystyrene ball was incubated with human Hb A and subsequently with affinity-purified anti-human Hb A Fab'-horseradish peroxidase conjugates, which had been prepared before and after absorption with Japanese monkey Hb-Sepharose 4B and dog Hb-Sepharose 4B. Bound peroxidase activity was measured by fluorimetry using 3-(4-hydroxyphenyl)propionic acid as a substrate. The detection limits of human Hb A using the conjugate before and after the absorption were 0.65 pg/tube (3 X 10(10)-fold dilution of whole blood) and 2.0 pg/tube (1 X 10(10)-fold dilution of whole blood), respectively. Human Hb A could be discriminated from Hb of animals such as Japanese monkey, dog, cat, pig, horse, sheep, chicken and cow by measuring bound peroxidase activity in the presence and absence of the conjugates prepared before and after the absorption. Human Hb A in bloodstains on cotton gauze could be discriminated from Hb of animals described above even after seven washings. Human Hb A in 220,000-fold diluted bloodstains on cotton gauze could also be discriminated from Hb of animals described above.

Peroxidase labeling of IgMs fragment of ABO blood group specific mouse monoclonal IgM.

A method for peroxidase labeling of the monomeric subunit (IgMs) of ABO blood group specific mouse monoclonal IgM is described. IgM was purified from a commercial monoclonal anti-B blood grouping reagent by a combination of salt precipitation, euglobulin precipitation, and gel filtration. IgM was mildly reduced with L-cysteine to yield SH-bearing IgMs. Finally, IgMs was conjugated to horseradish peroxidase, into which SH-reacting maleimide groups had been introduced using N-succinimidyl 6-maleimidohexanoate, through the selective reaction between SH of IgMs and maleimide groups of peroxidase.

A case of homicidal poisoning involving several drugs.

Accidental or suicidal poisonings due to benzodiazepines have been previously reported. A case of fatal, homicidal poisoning with benzodiazepines, antipyretic analgesics (anti-inflammatory drugs), and beer is described here. In this homicidal poisoning, the drugs and beer were given to the decedent by his wife. Autopsy findings showed no clinically significant macroscopic findings except for slight postmortem change. Capillary gas chromatography coupled to mass spectrometry was employed to quantitate the drugs in biological fluids and stomach contents. Six drugs (brotizolam, triazolam, ibuprofen, dihydrocodeine, phenylpropanolamine, and chlorpheniramine) were identified and quantitated in blood, urine, and stomach contents. Although each drug was present in a very small quantity, the cause of death was determined to be the combination of these drugs and alcohol poisoning.

A 20-kDa protein in human seminal plasma that is identical to gross cystic disease fluid protein 15 and prolactin-inducible protein.

A new method is described for isolating the 20-kDa protein, which used to be purified using an actin-immobilized column from human seminal plasma. This method employed ion-exchange column chromatography and isoelectric focusing separation. The level of seminal protein was determined by sandwich ELISA to be 1.06 +/- 0.27 mg/mL (mean +/- SD, n = 8). Its cDNA was cloned from a human salivary gland cDNA library by immunoscreening. The 553-nucleotide sequence included the 5' untranslated region and extended to the poly(A) tail. It encoded a protein of 118 amino acid residues in addition to a signal sequence of 28 residues. This sequence was identical to those of gross cystic disease fluid protein 15 and prolactin-inducible protein cDNAs. Northern blotting revealed the common expression in the submandibular gland and seminal vesicle. An immunohistochemical study in paraffin-embedded tissues from human male sex organs also indicated that the 20-kDa protein is mainly produced in the seminal vesicle.
