[Neuropathological study of Menkes' kinky hair disease: on the mechanism of development of cerebrovascular lesions (author's transl)].

In order to elucidate the mechanism of development of cerebrovascular lesions in Menkes' kinky hair disease (MKHD), the authors examined the brain and its vasculature of an autopsied case of this disease in light and electron microscopy. The gross and light microscopic examinations revealed that focal degeneration including pseudolaminar necrosis of cerebral cortices roughly concentrated on the gyri in and around the Sylvian fissures where tortuous vessels with congestion and dilatation were most prominently present. Despite their conspicuous irregularity in shape of the lumens and in thickness of the walls, the vessels were devoid of significant changes in elastic lamina and intima. In contrast, mitochondrial disease manifested by enlargement and/or rounding of mitochondria with tubular cristae and dense body as well as vacuolar degeneration were noted in various sizes of vessels in endothelial cells, pericytes and medial muscle cells, which exhibited various degrees of degeneration. On the other hand the preliminary observation had disclosed the mitochondrial changes of the same character in the neurons in various structures of this brain, such as Purkinje cells, granule cells, and neurons in cerebral cortex, thalamus and globus pallidus, and even in glial cells. Therefore it seemed reasonable to consider that the mitochondrial disease in MKHD might be ubiquituous in the brain including its vasculature. The observations of various degrees of degeneration of vessel wall cells due to mitochondrial disease and of irregular proliferation of reticulin fibrils in the spaces among degenerated muscle cells of the tunica media may be the evidences responsible for the tortuosity, dilatation and congestion of vessels, which eventually give rise to the vascular lesions in the brain parenchyme in MKHD. The findings of increased numbers of enlarged specific granules in the endothelium of dilated vessels, and dense material accumulation in their perivascular spaces with proliferation of basal lamina may have something to do with dilatation of small vessels and alteration of their blood brain barrier.

Establishment and characterization of human T hybrid cells secreting immunoregulatory molecules.

Hybridization of human T cells with an azaguanine-resistant human T cell line gave rise to T hybrid cell lines secreting several immunoregulatory molecules. Analyses of karyotypes, HLA phenotypes, and other surface phenotypes, such as T-cell-specific antigens or receptors for sheep erythrocytes and the patterns of mitogen responsiveness confirmed that the hypoxanthine/aminopterin/thymidine-resistant cell lines were human T-T hybridomas. One of the established hybrid clones (24-A) secreted human interleukin-2 (IL-2). The culture supernatants induced the proliferation of concanavalin A-stimulated murine T cells and supported the proliferation of an IL 2-dependent human cytotoxic T-cell line. In a clone (38-B) that did not show any IL-2 activity in culture supernatants, the addition of macrophages induced IL-2 production in the presence of phytohemagglutinin, suggesting that interleukin-1 induced IL-2 production in T hybrid cells. Hybrid cells secreting killer helper factor were also established. The culture supernatants from this clone, 55-A, helped the induction of cytotoxic T cells against UV-treated human B-blastoid cells but did not show any IL-2 activity.

Does intravenous fat spare nitrogen in the injured rat?

The role of glucose versus fat as a source of energy in parenteral nutrition is controversial, particularly when administered after trauma, injury or in sepsis. In the present series of experiments, rats undergoing laparotomy and jugular vein cannulation were infused with hypertonic glucose or Intralipid with and without amino acids. The results demonstrate that the combination of amino acids and glucose has a better nitrogen-conserving ability than amino acids and fat, lasting at least 4 days after injury. However, when infused without amino acids, fat had the same nitrogen-conserving quality as glucose. The authors attribute the improved nitrogen-conserving quality of amino acids and glucose to the specific carbohydrate influence on amino acid metabolism, an action which is specific in both protein-free and protein-containing meals but has only very limited effect in the absence of simultaneously infused or fed amino acids. In the moderately to severely injured rat, a glucose calorie is significantly more effective in sparing nitrogen than a fat calorie.

Lymphoproliferative response to membrane extracts of nasopharyngeal carcinoma.

The tumor-associated antigen (TAA) was extracted from cultured cells obtained from nasopharyngeal carcinoma (NPC), and the specific immune reaction was studied by utilizing the proliferation reaction of lymphocytes from NPC patients. The following results were obtained. (1) The stimulation of lymphocytes by membrane soluble antigen (MS-Ag) was significantly different in the group of patients with NPC and the control groups (patients with malignant tumors of the head and neck and healthy individuals). Lymphocytes from the NPC patients gave a significantly higher stimulation index (SI) value with NPC MS-Ag, but a lower value with PHA, as compared with those of normal controls. (2) No definite correlation was observed in anti-Epstein-Barr virus (EBV) antibody titers. (3) As for the correlation with various histological types, apparently enhanced reaction was noted irrespective of the histological type of the cancer. (4) In connection with stage classification, the reaction was always negative in patients in stage IV. A particularly close correlation was noted between the degree of the response and the N classification. Some inhibitory factor affecting the LP reaction might appear with the progress of N and M.
