Cytological diagnosis and telomerase activity of cells in effusions of body cavities.

Telomerase is a ribonucleoprotein that synthesizes telometric DNA on chromosome ends, and may be related to the aging and immortality of cells. Recently, a telometric repeat amplification protocol (TRAP) assay for telomerase activity, using the polymerase chain reaction, was developed. We examined the limitations of TRAP assay by applying it to a cultured colon cancer cell line (COLO320) and 58 human cytological materials from body cavity effusions, and obtained the following results; i) The limits of the TRAP assay were 20-50 cells for the COLO320 cell line; ii) One COLO320 cell per 100 normal blood white cells was detectable; iii) Seventeen of 58 samples were positive for telomerase activity in this study. The sensitivity was 69% (9/13) and the specificity was 87.5% (28/32) between cytological diagnosis and telomerase activity; iv) Among 29 malignant cases, 15 were positive for telomerase activity, while there were 11 cytologically positive cases. The positive cases detected by the combination of cytology and telomerase activity accounted for 21 of the total 29 cases (72.4%). These results suggest that the measurement of telomerase activity in body cavity effusions may be useful as an adjunctive tool for cytological and clinicopathological diagnosis and that this technique is potentially applicable to remnant cytological materials.

Chemopreventive effects of ferulic acid on oral and rice germ on large bowel carcinogenesis.

Effects of ferulic acid (FA) known as a major constituent of rice bran or germ on 4-nitroquinoline-1-oxide (4NQO)-induced oral carcinogenesis were examined in 4 groups of male rats. The incidences of tongue carcinomas and preneoplastic lesions (severe dysplasia) in rats of the group given FA in the diet at a dose of 500 ppm after exposure to 4NQO for 5 weeks in drinking water at a dose of 20 ppm, was significantly lower on termination of the experiment (32 weeks) in the group with the carcinogen alone (P < 0.005 and P < 0.001 respectively). The results suggest chemopreventive activity for this phenolic compound on oral cancer. Modifying effects of rice germ on azoxymethane (AOM)-induced formation of aberrant crypt foci (ACF) were also examined, in 8 groups of male F344 rats. Numbers of ACF/colon, ACF/cm2 and aberrant crypts/colon in the group treated with AOM (15 mg/kg s.c. once a week for 3 weeks) and gamma-amino butylic acid (GABA)-enriched defatted rice germ (2.5% in diet) or the group with AOM and rice germ (2.5% in diet) were smaller than those of the group with AOM alone (P < 0.005). Exposure to defatted rice germ or rice germ during the initiation phase or the post-initiation phase also decreased incidences of AOM-induced large bowel neoplasms (P < 0.05). These results suggest chemopreventive potential for rice germ on colonic neoplasia.

Cytologic appearance of carcinosarcoma (malignant ameloblastoma and fibrosarcoma) of the maxilla. A case report.

The cytologic appearance of a carcinosarcoma (malignant ameloblastoma and fibrosarcoma) of the maxilla in a 63-year-old man is described. On his first admission the diagnosis of malignant ameloblastoma was made on biopsy. After five surgical excisions, radiotherapy and chemotherapy, the diagnosis was changed to carcinosarcoma because the stromal cells of the tumor had become malignant. Aspiration biopsy cytology of the tumor, with a cystic lesion found at the left suborbital area, revealed malignant epithelial cells, indicating malignant ameloblastoma. Imprint smears of both surgically resected and autopsy material showed two types of malignant neoplastic cells, of epithelial and mesenchymal origin.

Ki-1-positive large cell anaplastic lymphoma diagnosed by urinary cytology. A case report.

Cytomorphologic and immunocytochemical findings in urinary cytology in a case of Ki-1-positive large cell anaplastic lymphoma in the left cervical region in a 42-year-old man are reported. Smears stained with Papanicolaou or Wright-Giemsa stain showed a cytomorphologic spectrum that varied from immunoblastlike cells to large neoplastic cells with pleomorphic nuclei and vacuolated cytoplasm. Neoplastic cells with multilobulated, bizarre nuclei and mitosis were also observed. Immunocytochemistry on urinary sediments showed that the tumor cells were of a "null" phenotype and strongly expressed the Ki-1 antigen and "activation" markers of HLA-DR, Tac (CD25) and OKT9 (CD71). Histology and immunohistochemistry confirmed these findings.
