Expression of herpes simplex virus common surface antigens and malignancy by clonal cells of a herpes simplex virus type 2-transformed line.

Herpes simplex virus (HSV) type common surface antigens (CSA) were examined by indirect immunofluorescence with rabbit antiserum to HSV type 1 in a clonal hamster cell line 155-4 transformed by HSV type 2. The tumor formation was examined in hamsters transplanted with various transformed and tumor cells. The examination of subclones derived from 1554-cell line gave the following results. (1) Thirty subclones were isolated and classified into three phenotypes as to CSA expression: (i) in CSA-positive type (20% of the clones isolated), the number of CSA-positive cells increased soon (5 hr) after seeding at 37 degree; (ii) in CSA-inducible type (33% of the clones), the number of CSA-positive cells increased after treatment with actinomycin D (ACT-D, 2 micrograms/ml), but not without ACT-D; (iii) CSA-negative type (47% of the clones), the number of CSA-positive cells did not increase after seeding or after ACT-D treatment. (2) In tumor cell lines derived from the parent line and from three representative clones, CSA expressions were similar to that by CSA-negative type. (3) Transformed cells expressing CSA after seeding or after ACT-D treatment formed tumors and metastases less efficiently than cells expressing little CSA in transplanted hamsters. (4) Tumor cell lines formed tumors and metastases more efficiently than transformed cells expressing little CSA in transplanted hamsters.

Neurochemical studies of an analgesic, 1,3-diphenyl-5-(2-dimethylaminopropionamide)-pyrazole [difenamizole].

For the purpose of clarifying the action mechanism of the analgesic agent, difenamizole (DFZ), chemically known as 1,3-diphenyl-5-(2-dimethylaminopropionamide)-pyrazole, the following properties were investigated: 1) monoamine metabolizing enzymes, 2) biosynthesis of dopamine (DA), 3) binding of DA on the synaptic membrane of mice, 4) uptake of catecholamine (CA) by synaptosome in discrete brain areas and 5) DA release from the striatal slices of mice. Pertinent results obtained are delineated below: 1) DFZ at 10(-4) M inhibited the monoamine oxidase (MAO) activity to some extent although it had no activity on catechol-O-methyltransferase (COMT) at 10(-6) -10(-4) M. 2) DFZ significantly enhanced the DA accumulating activity of pargyline in the striatum. 3) DFZ inhibited the binding of 3H-DA to synaptic membrane by 25% at 10(-4) M in the striatum. 4) DFZ inhibited the DA uptake by 50% at 10(-4) M though the activity was somewhat weaker than imipramine or cocaine on the striatum. 5) DFZ inhibited the DA release due to high K+ concentration in striatal slices at 10(-4) -5x10(-4) M. From the above data, it was concluded that DFZ may exert its analgesic effect by inhibiting the release of DA and preventing the binding of DA with the receptors.

The protective antigen of malarial sporozoites (Plasmodium berghei) is a differentiation antigen.

Pb44, the protective antigen of rodent malaria sporozoite (Plasmodium berghei) covers the entire surface of mature, salivary gland sporozoites. This antigen is undetectable in approximately 50% of the immature, i.e., oocyst sporozoites. On the surface of the remaining oocyst sporozoites, Pb44 is found in patches. Pb44 is present in early exoerythrocytic liver stages of P. berghei but is present in early exoerythrocytic liver stages of P. berghei but becomes undetectable after 30 hr of intrahepatocytic development. It seems likely that Pb44 is a differentiation antigen associated with an unique function of the sporozoites, perhaps penetration in the target host cells.

Affinity chromatography of Streptomyces erythreus trypsin-like enzyme on Japanese quail ovomucoid.

For simple, rapid purification of Streptomyces erythreus trypsin-like enzyme (TLE), we examined affinity chromatography with quail ovomucoid (QO) as a ligand of an affinity matrix. We prepared two affinity matrices by the cyanogen bromide method and the oxirane coupling method, and compared their binding efficiencies for TLE using frontal affinity chromatography. The affinity matrix in which QO was immobilized by the cyanogen bromide method showed 30% binding efficiency. However, another matrix, in which QO was immobilized by the oxirane coupling method with the lysine residue of the reactive site blocked by citraconylation, showed 92% binding efficiency. The results suggest that some biologically inactive QO was formed during the coupling reaction by the cyanogen bromide method. When we purified TLE from S. erythreus culture broth by affinity chromatography on QO-Sepharose, TLE was purified about 1,100-fold by affinity chromatography, and was further purified by DEAE-Sephadex A-25 column chromatography to homogeneity. The overall yield of TLE activity was higher than 90%. Thus, we were able to greatly improve previous purification procedures.

Preferential action of liposome-entrapped 1-(2-chloroethyl)-3-(4-methylcyclohexyl)-1-nitrosourea on lung metastasis of Lewis lung carcinoma as compared with the free drug.

Lipid vesicles entrapping a lipophilic antitumor agent, 1-(2-chloroethyl)-3-(4-methylcyclohexyl)-1-nitrosourea (MeCCNU), within the membrane phase were prepared and their antimetastatic activity was compared with that of free MeCCNU using intravenously inoculated Lewis lung carcinoma. It was found that the liposome preparation exhibited more potent inhibitory activity than the free drug on colony formation in the lung, when administered intravenously as well as intraperitoneally. Superior life-prolongation effect was also observed with liposome preparations as compared with the free drug in this system. However, the two forms of MeCCNU showed almost the same activity against not only Lewis lung carcinoma but also P388 leukemia inoculated subcutaneously and intraperitoneally, respectively. These results suggest that the superior effect of liposome-entrapped MeCCNU on lung metastasis might be due, at least in part, to preferential distribution of liposomes to the lung as compared with the free drug.

Monovalent fragments (Fab) of monoclonal antibodies to a sporozoite surface antigen (Pb44) protect mice against malarial infection.

Monoclonal antibodies (IG1, k) directed against a surface component of Plasmodium berghei sporozoites (Pb-44) confer complete protection to mice against a lethal inoculum of parasites. The degree of protection is a function of the number of parasites used in the challenge and of the antibody concentration in serum. Passive transfer of 10 micrograms of antibody per mouse abolished or profoundly diminished the infectivity of 10(3) sporozoites, but much higher amounts of antibody were required for complete protection against challenge with 10(4) parasites. Fab fragments of the monoclonal antibodies were as effective as the intact antibodies in mediating protection as determined by the neutralizing assay. This observation suggests that the antibodies interfere with a parasite function necessary for its infectivity, such as, for example, the ability to penetrate into the target cell or to multiply in the hepatocytes. When sporozoites are incubated with the intact monoclonal antibodies at 37 degrees C, a long filament appears at its posterior end (circumsporzoite precipitation [CSP] reaction). Fab fragments are ineffective at high concentrations. However, if after treatment with Fab, the sporozoites are incubated with rabbit antibodies to mouse k-chains, a strong CSP reaction is observed. We conclude that the CSP reaction can result from the cross-linking of Pb44 and that it has the characteristics of a capping reaction followed by the shedding of the immune complexes.

Hybridoma produces protective antibodies directed against the sporozoite stage of malaria parasite.

Hybrid cells secreting antibodies against sporozoites of Plasmodium berghei were obtained by fusion of plasmacytoma cells with immune murine spleen cells. The monoclonal antibodies bound to a protein with an apparent molecular weight of 44,000 (Pb44), which envelopes the surface membrane of sporozoites. Incubation of sporozoites in vitro with antibodies to Pb44 abolished their infectivity.

Immunohistochemical localization of cyclic nucleotides in guinea pig lip.

The cellular localization of cAMP and cGMP in the guinea pig lip was investigated with an indirect immunofluorescence technique. cAMP was distributed both in cell membranes and cytoplasmic elements with coarse and granular intranuclear fluorescence, while cGMP was localized in nuclear sites including nuclear membranes as well as cell membranes and cytoplasm. In the basal portion, both cAMP and cGMP fluorescence was weaker in cytoplasm and cell membranes than seen in the upper portion.

Steroid 16 alpha-hydroxylase from human fetal liver; inhibition by steroids.

The 16 alpha-hydroxylase system in fetal liver which used dehydroepiandrosterone (DHA) or pregnenolone as substrate, was apparently inhibited by various endogenous and synthetic steriods: DHA, pregnenolone, their sulfates, androstenediol, androstenetriol, estrone, estradiol-17 beta, ethynylestradiol and chlormadinone-acetate. The inhibition constants (Ki) towards DHA were as follows; pregnenolone 22 muM, DHA-sulfate 13 muM, pregnenolone-sulfate 21 muM, androstenediol 16 muM, androstenetriol 53 muM estrone 32 muM, estradiol-17 beta 74 muM, ethynylestradiol 22 muM and chlormadinone-acetate 27 muM. The Ki values towards pregnenolone were DHA 6.3 muM, DHA-sulfate 8.3 muM, pregnenolone-sulfate 3.9 muM, androstenediol 8.7 muM, androstenetriol 14.7 muM, estradiol-17 beta 15.4 muM and ethynylestradiol 16.0 muM, respectively. The reaction products, 16 alpha OH-DHA and 16 alpha OH-pregnenolone, showed little inhibitory effect upon the 16 alpha-hydroxylase.

Sucralfate, a basic aluminum salt of sucrose sulfate. IV. Interaction with enzyme pepsin.

The interactions of sucralfate in various physical states, i.e., suspension, paste, and dissociated sucrose sulfate ion, with the enzyme pepsin were studied. The suspended form of sucralfate strongly adsorbs pepsin. The paste form also adsorbs pepsin but to a limited extent due to the reduced effective surface area. In view of other studies indicating selective coating of the ulcer lesion with sucralfate, this property supports the concept that it provides a potential protective barrier against the peptic attack of the lesion. By contrast, the interaction of sucrose sulfate ions with pepsin was found almost nil, giving additional evidence to the previous conclusion that the inhibition of peptic hydrolysis by sucrose sulfate is due to the interaction with the substrate rather than pepsin.

Induction and regulation of immunoglobulin expression in a murine pre-B cell line, 70Z/3. I. Cell cycle-associated induction of sIgM expression and kappa-chain synthesis in 70Z/3 cells by LPS stimulation.

Stimulation of a murine pre-B cell line, 70Z/3, with LPS induced sIgM expression and an increase of L kappa-chain mRNA within 12 hr. In synchronized 70Z/3 cells, the cell division cycle was 10 to 11 hr and LSP-stimulation did not affect the cell division cycle. LPS-signals given at G1/S boundary induced sIgM expression in the G2 to M-phase. On the other hand, LPS-signals given after M-phase did not induce sIgM expression. Inhibition of cell division with demecorcin did not affect sIgM expression in the M-phase. Induction of intracellular kappa-chain synthesis was also observed in the G2 to M-phase when the LPS-stimulation was provided at G1/S boundary, but LPS-signals given after the M-phase did not induce de novo synthesis of kappa-chain. These results showed that LPS-induced sIgM expression and synthesis of kappa-chain were cell cycle-related events and sIgM expression was associated wih de novo synthesis of kappa-chain.

Sucralfate, a basic aluminum salt of sucrose sulfate. III. Inhibition of peptic hydrolysis of fibrinogen by sucrose sulfate.

As an extension from a previous study with serum albumins, the interaction of sucrose sulfate with bovine plasma fibrinogen was studied with respect to peptic inhibition, complexation and pH-dependence. While such features unique to this protein were found as a small degree of dependences on substrate concentration in peptic inhibition and in compositional ratios of insoluble complexes, the interaction of sucrose sulfate with fibrinogen at acid pH proved essentially similar to that with serum albumin. Both interactions are electrostatic in nature and show extensive binding that results in the formation of soluble as well as insoluble complexes resistant to peptic hydrolysis. The concentration ratio is more important than the absolute concentrations of the substrate and the inhibitor determining the degree of peptic inhibition. A sucrose sulfate to fibrinogen molar ratio of 50 represented the saturation point and was almost identical to the ratio found in the interaction with serum albumin expressed in mol of sucrose sulfate bound by a unit weight of protein. The principles found with serum albumin and fibrinogen will also apply in the interaction with other proteins at acid pH.

Antitumor activities of newly synthesized 5-carbamoyl-1 H-imidazol-4yl 1-adamantanecarboxylate and 5-carbamoyl-1H-imidazol-4yl piperonylate.

In synthetic studies on the chemical modification of the nucleoside antibiotic bredinin, two new derivatives, 5-carbamoyl-1H-imidazol-4-yl 1-adamantanecarboxylate and 5-carbamoyl-1H-imidazol-4-yl piperonylate, were found to possess a potent antitumor activity in several experimental tumor systems, even though bredinin itself shows only in vitro cytotoxicity and thus lacks therapeutic effectiveness. These two derivatives of bredinin exhibited antitumor activity against a wide variety of tumors, including leukemias L1210 and P388, Lewis lung carcinoma, B16 melanoma, Colon 26 and 38 adenocarcinomas. Ehrlich carcinoma, and Sarcoma 180. It is noteworthy that these agents showed good therapeutic effects not only against ascitic types of tumors but also against a number of slow-growing solid tumor lines, particularly the ascitic and solid forms of Ehrlich carcinoma. At their optimal doses, both compounds effected a complete cure of all or most of the mice treated. Although the mechanisms of action of these compounds remain unknown, they are able to suppress in vivo tumor growth, presumably by being slowly anabolized in vivo to an active form and inhibiting purine de novo synthesis as bredinin does.

Adrenergic innervation in the Eustachian tube of guinea pigs.

We have succeeded in demonstrating for the first time adrenergic innervation in the Eustachian tube of guinea pigs. The approach was histochemical using the fluorescence method with glyoxylic acid. There was an apparent regional variation with a greater number of axon terminals in the pharyngeal area of the Eustachian tube as compared to the tympanic area.

Steroid specificity of human placental 5-ene-3 beta-hydroxysteroid oxidoreductase.

The properties of 5-ene-3 beta hydroxysteroid oxidoreductase (3 beta-HSD) from human placental homogenates were studied in vitro. The apparent Michaelis constants for 3 beta-HSD with the substrates pregnenolone (delta 5P) and dehydroepiandrosterone (DHA) were 170 nM and nM respectively. The optimal pH for both these substrates was between 10 and 12. With NAD as the substrate, the Km for the pregnenolone was 20 microM and for DHA, 17 microM. The activity of 3 beta-HSD was inhibited by various steroids. Competitive inhibitors (pregnenolone substrate) included: ethynylestradiol (inhibition constant Ki=7.3 nM), DHA (Ki=46 nM), estradiol-17 beta (Ki=46 nM), cholesterol (Ki=0.68 microM) and 16 alpha-hydroxydehydroepiandrosterone (16 alphaOHDHA) (Ki=2.2 microM). When the substrate was DHA, competitive inhibition occurred with the following steroids: ethynylestradiol (Ki=6.4 nM), estradiol-17 beta (Ki=69 nM), pregnenolone (Ki=91 nM), cholesterol (Ki=1.3 microM) and 16 alphaOHDHA (Ki=1.9 microM). 4-Ene-3-ketosteroids such as androstenedione, progesterone (delta 4P), norethindrone and chlormadinone acetate acted as noncompetitive inhibitors towards both substrates.
