Establishment of three rat soft tissue tumor cell lines with different degrees of myogenic differentiation.

Soft tissue malignancies often show divergent differentiation, including myogenic lineage. Five rat tumors induced with 20-hydroxymethylcholanthrene (20-OH-MCA), were cultured in vitro, and three cell lines (YMC-1, YMC-2, YMC-3) were established from them. YMC-2 and -3 cells were spindle-shaped, and YMS-1 cells round and epithelioid. In confluency, YMC-3 cells formed myotubes. The nude mouse xenotransplants showed morphological features consistent with their myogenic phenotypes. Muscle-specific enzyme activities were highest in YMC-3 cells. These lines may be useful in the investigation of the myogenic differentiation of undifferentiated mesenchymal cells.

A B cell lymphoma line, BALL-1 stimulates T cells to produce a unique eosinophil chemotactic factor.

Human mononuclear leukocytes (MNL), probably OKT4-positive T cells, produced an eosinophil chemotactic factor (ECF) when they were cocultured with irradiated BALL-1, a B cell lymphoma line. Treatment of MNL, with anti-IL-2 antibody failed to suppress BALL-1-induced ECF production. Periodate-lysine-paraformaldehyde-fixed but not acetone- and ethanol-fixed BCLL induced evident ECF production. These results suggested that some cell surface molecules play a role in the induction of ECF production. Isoelectric point of BALL-1-induced ECF was around pH7, whereas that of IL-2-induced ECF was around pH 5. The molecular weight of BALL-1-induced ECF was between 10 and 30 kD. Although a combination of MoAb against IL-3, IL-5, and GM, CSF suppressed the activity of IL-2-induced ECF, it failed to suppress that of BALL-1-induced ECF. Furthermore, BALL-1-induced ECF suppressed fMLP-induced respiratory bursts of eosinophils, while IL-2-induced ECF failed. We propose that at least one reason for eosinophil infiltrate into the stroma of tumors is that the tumor cells stimulate T cells to produce BALL-1-induced ECF, and the eosinophils attracted by the ECF exhibit different functions from those by other ECF.

Heterogeneous eosinophils of allergic rhinitis in chemotactic response.

Chemotactic responses of eosinophils from patients with allergic rhinitis to 5 STO-2-derived eosinophil chemotactic factors (ECF), IL-3, IL-5 and GM-CSF were examined. The patients were divided into two groups: patients with perennial allergic rhinitis sensitive to Dermatophagoides farinae and those with seasonal allergic rhinitis sensitive to Japanese cedar pollen. There was no essential difference between chemotactic response of eosinophils from the former to STO-2-derived ECF and that from healthy individuals. However, eosinophils from patients with seasonal allergic rhinitis failed to respond to one of the STO-2-derived ECFs, ECF-P19. Before and after nasal antigen provocation, no change occurred in the chemotactic profiles. Furthermore, we found that eosinophils from both types of patients with allergic rhinitis responded not only to IL-3 and GM-CSF but also to IL-5 unlike those of healthy individuals.

Synthesis and biological activities of the optical isomers of (+/-)-4-amino-5-chloro-2-ethoxy-N-[[4-(4-fluorobenzyl)-2- morpholinyl]methyl]benzamide (mosapride).

The enantiomers, (S)-(-)-1 and (R)-(+)-1, of (+/-)-4-amino-5-chloro-2-ethoxy- N-[[4-(4-fluorobenzyl)-2-morpholinyl]methyl]benzamide (mosapride) [(+/-)-1], a new and selective gastroprokinetic agent, were prepared from optically active [4-(4-fluorobenzyl)-2-morpholinyl]methylamines (S)-(-)-4 and (R)-(+)-4, respectively. The requisite (S)-(-)-4 and (R)-(+)-4 were prepared by optical resolution of [4-(4-fluorobenzyl)-2-morpholinyl]methyl p-toluenesulfonate [(+/-)-5] using (-)- and (+)-N-(p-toluenesulfonyl)glutamic acids, followed by amination of the tosyloxy groups of (R)-(-)-5 and (S)-(+)-5, respectively. The absolute configurations of (R)-(-)-5 and (S)-(+)-5 were determined on the basis of an asymmetric synthesis of (R)-(-)-5 from (S)-(+)-benzyl glycidyl ether [(S)-(+)-11]. Mosapride and its enantiomers, (S)-(-)-1 and (R)-(+)-1, were essentially equipotent in serotonin 5-HT4 receptor agonistic activity on the electrically evoked contractions in isolated guinea pig ileum.

[Comparison of formulas for calculating average skin temperature and their characteristics].

In order to obtain data of skin temperatures experiments were carried out using three healthy young Japanese males. The subjects were exposed to each of the four environments with dry bulb temperatures of 15 degrees C, 19 degrees C, 25 degrees C and 33 degrees C. At each of these air temperatures, relative humidity and air movement were set at 50% and 0.15m/s respectively. The subjects wore only athletic shorts, seated on the meshed chair. Each subject was measured with thermisters continuously for one hour under these conditions to obtain twenty-nine regional skin temperature. The above experiments were made with one subject at a time in the test chamber. The data of skin temperatures observed were substituted into twenty-eight different weighting formulas for comparison. The present analysis revealed that the calculation from the 12-point and the 7-point skin area formulas by Hardy-DuBois showed approximate mean values of the twenty eight. Moreover, the values calculated from the formula by Nadel et al, which was weighted by skin area and thermal sensitivity, are similar to the values calculated by the formula of Mochida, which was weighted by skin area, heat transfer coefficients and thermal sensitivity. Furthermore, the authors verified that the area-mean weighting factor was derived from the Teichner's definition in which a limiting value of arithmetical mean of skin temperatures gave a value of average skin temperature.

[Validity of the values of maximal oxygen intake for women recommended in the 'Quantity of exercise for developing health' by Ministry of Health and Welfare].

The purpose of this study was to examine the value of maximal oxygen intake per kilogram body weight (VO2max/wt) recommended for women between 20 and 29 years of age by the Ministry of Public Welfare. The subjects were 199 female college students, who were not in the habit of drinking and smoking. Results obtained were as follows: After controlling the effects of age and percent body fat (%fat), VO2max/wt showed a significant correlations with %fat, high-density lipoprotein cholesterol and systolic blood pressure. The values of VO2max/wt corresponding to borderline values of these CHD risk factors ranged from 27.6 to 29.5ml/kg.min. The highest value of VO2max/wt was very close to the value recommended for women older than 60 years of age by the Ministry of Public Welfare. The subjects were then divided into 4 groups; at above and below the value of VO2max/wt (35ml/kg.min) recommended for women between 20 and 29 years of age, and at above and below the value of VO2max/wt (31ml/kg.min) recommended for women older than 60 years of age by the Ministry of Public Welfare. There were no significant differences among 4 groups in occurrence of abnormal values in CHD risk factors. The results of this study support the validity of the value of VO2max/wt (31ml/kg.min) recommended for women older than 60 years of age by the Ministry of Public Welfare. However, it seems that it is not necessary to recommend the value of VO2max/wt (35ml/kg.min) for women between 20 and 29 years of age, who were not in the habit of drinking and smoking.

Six cases of heterochronous gastrointestinal cancer preceded by breast cancer.

Among 432 women with primary breast cancer, six (1.4%) were diagnosed as having gastrointestinal cancer more than six months after operation for the breast cancer. This paper presents these six cases. The patients ranged from 56 to 78 years of age at the time of breast cancer surgery, and the interval after surgery until diagnosis of the second cancer ranged from 7 months to 5 years 1 month. The second cancer was gastric cancer in 3, esophageal cancer in 2, and hepatic cancer in 1. All of the 6 patients had received postoperative adjuvant chemotherapy for breast cancer. The most frequent histological type of breast cancer was solid-tubular carcinoma (3 patients). Three patients died, due to the second cancer, 4 days, 2 months, and 6 months, respectively, after diagnosis of the second cancer, and the other patients are alive 2, 3, and 4 years after diagnosis. The Japanese literature regarding multiple cancer among breast cancer patients is reviewed. It is concluded that care should be taken to examine breast cancer patients with gastrointestinal symptoms, which are likely to be dismissed as a side effect of postoperative chemotherapy.

[Effect of combination therapy with disodium cromoglycate (DSCG) and Ozagrel on non-atopic asthmatics].

Ozagrel (OKY-046), a selective inhibitor of thromboxane biosynthesis has been reported to reduce airway hyperresponsiveness in asthmatics. DSCG has been widely used for atopic, but not for non-atopic asthmatics. We evaluated the additive effect of combination therapy with DSCG and Ozagrel on nonatopic asthmatics. Fourteen asthmatics were divided into two groups. The patients in the O-D group were initially treated with Ozagrel (400 mg/day) for 4 weeks, and then DSCG (8 mg/day) was added for another 4 weeks. The patients in D-O group were initially treated with DSCG (8 mg/day), and then Ozagrel (400 mg/day) was added for 4 more weeks. TXB2, PGF2 alpha, PGE2, 6-keto-PGF1 alpha in sputum and 11-dehydro TXB2 in urine were measured before and after 4th and 8th weeks of therapy. Ozagrel decreased the metabolites of thromboxane and PGF2 alpha in sputum and urine. PGE2 in sputum tended to be increased by Ozagrel. DSCG did not affect mediators in sputum. Combination therapy with Ozagrel and DSCG showed the additive effect to single use on attack score and PF in non-atopic asthmatics.

[Clinical study of renal cell carcinoma invading adjacent organs].

We studied 15 patients with renal cell carcinoma invading adjacent organs (stage T4) between January 1980 and December 1991. Such invasion was four times more frequent in males than in females. The patients were between 41 and 78 years old, with a mean age of 63.9 years. The tumor was on the right side in 4 cases, and on the left side in 11 cases. Six patients (40%) presented with flank pain. The pancreas was the organ involved most frequently. Eleven patients had regional lymph node involvement or distant metastasis. Most patients had an increased erythrocyte sedimentation rate (ESR), elevated alpha-2 globulin levels, and positivity for c-reactive protein (CRP). In 6 patients, nephrectomy was extended to the abdominal or retroperitoneal structures that seemed to be invaded by tumor. Patients with T2 or T3 tumor had a significantly longer overall survival than patients with a T4 tumor. However, there was no significant difference in survival between T2/T3 tumors and T4 tumors in nephrectomized patients. Two patients who survived longer than 3 years showed no abnormalities of ESR, alpha-2 globulin and CRP. They also had no nodal or distant metastases, and had a good initial performance status. These findings suggest that extended local resection can improve the survival and quality of life for selected patients with T4 tumors.

Neurotransmitter release from lysophosphatidic acid stimulated PC12 cells: involvement of lysophosphatidic acid receptors.

The significance of phospholipase D in metabolic regulation is being recognized. The products of phospholipase D action are phosphatidic acid and lysophosphatidic acid (LPA). Whether these lipid products exert their effect through plasma membrane receptor-mediated mechanism is not yet clear. In the present study we have shown the presence of specific receptors for LPA in plasma membranes of PC12 pheochromocytoma cells. LPA evoked a transient rise in [Ca2+]i concomitant with formation of IP3. LPA also stimulated dopamine release from PC12 cells. These results indicate that LPA stimulates catecholamine release via specific LPA receptors presumably by activating the IP3 mediated Ca2+ release.

Effects of strong magnetic fields on cell growth and radiation response of human T-lymphocytes in culture.

Experiments were undertaken in order to verify whether or not a strong magnetic field would have any biological effects on the cell growth, viability and radiation response of mammalian cells. Magnetic field exposures were conducted using a superconducting magnet with freshly-isolated human peripheral blood T-lymphocytes maintained at their normal growing temperature of 37 degrees C. The static magnetic fields with intensities up to 6.3-tesla (T) exerted little influence on the cell growth and viability of actively-growing T-lymphocytes under normal cell-culture conditions. On the other hand, the T cells exposed to the magnetic fields (4 T-6.3 T) during PHA stimulation were inhibited in their cell growth when compared to controls. The effects of the magnetic fields with intensities up to 2 T on cell growth properties, however, were minimal in this system. Also, the radiosensitivity of T-lymphocytes previously exposed to the strong magnetic fields was more sensitive than that of control cells. These results suggest that exposure to a static magnetic field of 4 T or stronger might lead to physiological and growth abnormalities at the cellular level.

Purification, characterization and molecular cloning of an acidic amino acid-specific proteinase from Streptomyces fradiae ATCC 14544.

We have isolated a novel acidic amino-acid-specific proteinase from Streptomyces fradiae ATCC 14544, using benzyloxycarbonyl-L-Phe-L-Leu-L-Glu-p-nitroanilide (Z-Phe-Leu-Glu-pNA) as a substrate. A proteinase, which we propose to call SFase, was purified from the culture filtrate by salting out, repeated S-Sepharose chromatography, and affinity chromatography (CH-Sepharose-Phe-Leu-D-Glu-OMe). The purified enzyme showed a single band having an apparent molecular weight of 19,000 on sodium dodecyl sulfate polyacrylamide gel electrophoresis. When synthetic peptides were used as substrates, SFase showed high specificity for Z-Phe-Leu-Glu-pNA. Comparison with nitroanilides of glutamic acid and aspartic acid as substrates revealed that the reactivity was about 10-fold higher for a glutamyl bond than an aspartyl bond. SFase selectively hydrolyzed the -Glu-Ala-bond of two glutamyl bonds in the oxidized insulin B-chain within the initial reaction time until the starting material was completely digested. Diisopropylfluorophosphate and benzyloxycarbonyl-Phe-Leu-Glu chloromethylketone completely inhibited SFase, while metalloproteinase inhibitors, such as EDTA and o-phenanthrolin, did not inhibit the enzyme. The findings indicate that SFase can be classified as a serine proteinase, and is highly specific for a glutamyl bond in comparison with an aspartyl bond. To elucidate the complete primary structure and precursor of SFase, its gene was cloned from genomic DNA of the producing strain, and the nucleotide sequence was determined. Consideration of the N- and C-terminal amino-acid sequences of the mature protein of SFase indicates that it consists of 187 amino acids, which follows a prepropeptide of 170 residues. In comparison with the acidic amino-acid-specific proteinase from Streptomyces griseus (Svendsen, I., Jensen, M.R. and Breddam, K. (1991) FEBS Lett. 292, 165-167), SFase had 82% homology in the amino acid sequence. The processing site for maturation of SFase was a unique sequence (-Glu-Val-), so that the propeptide could be released by cleavage of the peptide bond between Glu and Val.

Cyclic flow variations in a conscious dog model of coronary artery stenosis and endothelial injury correlate with acute ischemic heart disease syndromes in humans.

OBJECTIVES: The purpose of this study was to test the hypothesis that episodes of cyclic flow variations (CFVs) in conscious dogs with coronary stenoses and endothelial injury correlate with acute ischemic heart disease syndromes in humans. BACKGROUND: Although the canine model with CFVs has proved to be a useful model of coronary thrombosis, whether CFVs progress to these syndromes has not been clearly described. METHODS: Cyclic flow variations were produced by an external constrictor placed at the site of the left anterior descending coronary artery with injured endothelium. Blood flow in this artery and 24-h Holter electrocardiogram (ECG) were recorded during the 1st 5 postoperative days. RESULTS: Of 41 dogs that underwent the initial operative procedure successfully, 29 developed an episode of CFVs. In five dogs in which CFVs persisted throughout the monitoring period, the left anterior descending coronary artery flow decreased until day 3 and thereafter increased through day 5. Transient coronary occlusion during CFVs induced ST segment changes that returned to baseline after reflow. In 12 dogs, CFVs progressed to persistent coronary occlusion, and histologic examination revealed thrombus formation at the stenotic site and evidence of myocardial infarction. Four of these 12 dogs died suddenly of ventricular arrhythmias during persistent coronary occlusion; another 5 dogs died of reperfusion arrhythmias during CFVs with no evidence of myocardial infarction. CONCLUSIONS: Conscious dogs with CFVs closely correlated with clinical acute ischemic heart disease syndromes, suggesting them to be a useful model for investigating the complex mechanisms of cellular interactions in the pathogenesis of these syndromes.

Localization and invasion by a soft tissue tumor induced by pellets made of hydroxypropylcellulose mixed with 20-methylcholanthrene in the femur of rats.

BACKGROUND: Pellets made of a carcinogenic substance, methylcholanthrene, admixed with a high molecular weight substance, hydroxypropylcellulose, were implanted into the thigh of 90 rats (three groups, 30 rats each). METHODS: In Group 1, the pellets were placed at the window made in the cortex of the femur; in Group 2, into the space between the dorsal femoral cortex and periosteum; and in Group 3, into the pouch created outside of the dorsal periosteum. RESULTS: Twenty weeks after implantation, rhabdomyosarcoma developed in 28 rats in Group 1, 27 in Group 2, and 21 in Group 3. Tumor invasions into the marrow of the femur were seen in 14 rats in Group 1 and 5 in Group 2. In Group 3, however, tumor invasion was not observed. There were significant differences in the invasion rate of tumor between Groups 1 and 2 (P < 0.01) and between Groups 2 and 3 (P < 0.05). At implant sites, Group 2 showed thinning out of the periosteum, whereas in Group 3 thickening of both cortex and periosteum was found. CONCLUSION: These experimental results are interpreted to mean that the periosteum provides an important barrier around the bone against tumor invasion into the bone.

Trypanosoma cruzi: antibody production and T cell response induced by stage-specific surface glycoproteins purified from metacyclic trypomastigotes.

The main surface glycoproteins of metacyclic trypomastigotes of Trypanosoma cruzi, gp90, gp82, and gp35/50, were purified and the immune response elicited by these antigens was analyzed. Balb/c mice immunized with antibody-affinity-purified gp82, plus alum as adjuvant, produced antibodies that recognized both the gp82 and the heterologous gp90 and gp35/50. On the other hand, antisera to gp90 reacted only with the homologous antigen, either by immunoprecipitation or by immunoblotting. Neither sera reacted with unrelated proteins in ELISA. Both antisera lysed 90-100% metacyclic forms in a complement-mediated reaction, a property associated with protection. However, in contrast to gp90, previously shown to induce protective immunity against acute T. cruzi infection, gp82 was not immunoprotective. Lymph node (LN) cells of mice primed with gp82 or gp90, which display 40% amino acid sequence identity at the carboxy terminal domain, were strongly stimulated in vitro by either one of these antigens. Proliferation, inhibitable by anti-CD4 but not by anti-CD8 antibodies, was T. cruzi-specific, no activation being observed with irrelevant antigens. LN cells of mice immunized with unrelated proteins did not proliferate in vitro in the presence of gp82 or gp90. The 35/50-kDa glycoconjugate, which was phenol-extracted, did not elicit any detectable antibody or T cell response.

A case of inflammatory malignant fibrous histiocytoma of the colon.

Primary malignant fibrous histiocytoma seldom occurs in the alimentary tract. This report documents a case in which a 50-year-old man presented with abdominal fullness and anorexia, together with inflammatory reactions such as fever, leukocytosis, an elevated erythrocyte sedimentation rate and positive CRP. Radiologic and endoscopic studies revealed a large mass in the ascending colon. The histology of the resected tumor revealed inflammatory malignant fibrous histiocytoma, a histologic subtype which is rare among not only soft tissue fibrous but also as gastrointestinal histiocytomas. The inflammatory reactions disappeared following resection of the tumor. Although we suspected that the tumor produced granulocyte colony-stimulating factor because of the slightly elevated serum level and the characteristics of the tumor histology, an immunohistochemical study failed to show the G-CSF production by the tumor cells.
