Establishment and characterization of a carcinoembryonic antigen producing cell line derived from human pancreatic exocrine cancer.

A human pancreatic carcinoma cell line derived from well differentiated tubular adenocarcinoma of the pancreas head has been established and maintained for nearly 4 years. This established cell line produces and releases carcinoembryonic antigen into the culture medium. The cell line grows as a monolayer in RPMI-1640 medium containing 10 percent fetal calf serum. Xenotransplantation in athymic nude mice after subcutaneous injection of EDTA-trypsin treated cells did not succeed.

Factors affecting the CEA secretion of human adenocarcinoma cell lines into the spent medium.

CEA producing cell lines were established from human gastric cancer (HGC-Y1), pancreatic cancer (HPC-Y9) and lung cancer (HLC-Y1). The culture medium was used RPMI-1640 supplemented with 10% fetal bovine serum. The secretion of carcinoembryonic antigen (CEA) into the spent medium from these cultured cell lines was modified by several factors, such as theophylline, cyclic AMP (cAMP), dibutyryl cyclic AMP (dbcAMP), Bromodeoxyuridine (BrdUrd), dimethyl sulfoxide (DMSO), Prostaglandin E2 (PGE2) and human interferon (INF). CEA secretion was enhanced by theophylline, cAMP, PGE2 and INF. Theophylline had an optimal dose to maximally enhance CEA secretion. cAMP and INF apparently enhanced CEA secretion dose dependently. PGE2 appeared to enhance CEA secretion, although cell growth was markedly suppressed dose dependently, dbcAMP, DMSO and BrdUrd did not affect CEA secretion. Here, the kinetics of CEA secretion was discussed.

Anti-tumor activity of endotoxin depends on activation of serum complement fragments.

The anti-tumor activity of endotoxin (LPS), derived from E. coli O127, on the subcutaneous tumor MH134 inoculated into C3H/He mice in vivo was examined. On repeated examination, LPS administration of 50-100 micrograms/head induced a 50% complete cure rate. However, pretreatment with K76COONa inhibited the anti-tumor activity of LPS, and no case of cure could be found. These results showed that the main anti-tumor activity of LPS administration consisted of complementary activation, mainly from the C5 step. The fact that the intraperitoneal administration of human complement activated serum with agar apparently inhibited the tumor growth in vivo certified these phenomena.

"Convulsoid responses" suggesting development of autonomous epileptogenicity.

To test a convenient and reliable neuronal index that suggests the development of autonomous epileptogenicity, we conducted in acute experiments on rabbits a comparative study of the changes in direct cortical responses (DCRs) in primary and mirror sites during electrically induced primary (PRID) and projected seizure discharges (PROD). It has been assumed that the primary site shows autonomous epileptogenicity during the PRID, whereas the mirror site during the PROD does not. We observed that in the primary site, the dendritic potentials (field EPSPs) were remarkably suppressed in amplitude or disappeared during the PRID and that there was a loss of the after-positivity (field IPSPs); these DCRs recovered gradually after termination of the PRID. Instead of DCRs, "convulsoid responses" (a general term for responses similar to the individual spontaneous waves which occur in all types of seizures discharges) were usually elicited in the primary site during PRID. In the mirror site during PROD, the DCRs showed three different behaviors: they were either unaffected, increased, or suppressed. Convulsoid responses were never elicited. In a few cases, independent seizure discharges were induced in the mirror site. During the independent seizure discharges, the DCRs in the mirror site disappeared and convulsoid responses similar to the individual independent waves were usually elicited, as in the primary site during PRID. We conclude that the convulsoid response was the most reliable indicator of the development of autonomous epileptogenicity, and that suppression or disappearance of DCRs was the supplementary sign of that development. These neuronal indexes may be useful for identification of secondary epileptogenesis.

Prostaglandin D2 inhibits the proliferation of human neuroblastoma cells.

The cytotoxic action of prostaglandin (PG) D2, E1 and F2 alpha was examined on human neuroblastoma cells (NB-1 cell line), and PGD2 was found to be the most effective. PGE1, thought to be the most effective among all PGs in the therapy of neuroblastoma, was much less effective than PGD2. PGF2 alpha did not show any inhibitory effect on the proliferation of NB-1 cells. When PGD2 was added, the cytoplasma became microscopically larger, then the cells gradually died off. PGD2 also exerted a dose-dependent inhibition of DNA and RNA syntheses. These results strongly suggest an antineoplastic activity of PGD2 for human neuroblastoma.

Pseudothrombocytopenia with cold-type platelet agglutinins.

Pseudothrombocytopenia is an in vitro phenomenon of platelet agglutination and clumping in the presence of EDTA on the electronical measurement of platelet count. Agglutinins reacting with platelet antigens belong to one heavy and one light chain of immunoglobulins with or without complement by immunofluorescence. The platelets are markedly agglutinated in EDTA, but scarcely in the presence of other anticoagulants. Agglutinations seem to occur more readily at cold temperature rather than at 37 degrees C.

The effect of maternal antigenic stimulation upon the active immune responsiveness of their offspring.

Pregnant mice were stimulated by sheep erythrocytes (SRBC) and the active immune responses of their offspring were investigated. The offspring whose mothers were stimulated with SRBC did not develop either IgM or IgG plaque-forming cell (PFC) target cells. From the dose response of pregnant mice for inducing suppression, enough doses (10(8)-10(10) cells of SRBC) for inducing primary anti-SRBC PFC could establish suppression in the young. Both intravenous and intraperitoneal administration of SRBC induced almost complete suppression of the specific PFC response (98.6-95.2%), but only partial suppression (57.8%) was induced by subcutaneous injection. For suppression to take place, female mice had to be injected with SRBC from 2 days before fertilization to day 16 of gestation. Suppression of the PFC response was not obtained when SRBC were given 3 days before fertilization or just 24 hr before delivery. This suppressive effect on the PFC response persisted until the 15th week after birth. In these newborn mice, detectable amounts of specific anti-SRBC antibodies were found. After exchanging mothers and newborns, the stimulated newborns fostered by normal mothers were still unresponsive following antigenic stimulation, even though a specific antibody from the mother was not detected. However, normal newborns nursed by stimulated mothers could respond to SRBC injection, no matter how the specific antibodies were transferred. Possible mechanisms of immunosuppression of the PFC response by antibody transmitted by the mother to her offspring are discussed.
