The herpes simplex virus immediate-early protein ICP0 affects transcription from the viral genome and infected-cell survival in the absence of ICP4 and ICP27.

ICP4, ICP0, and ICP27 are the immediate-early (IE) regulatory proteins of herpes simplex virus that have the greatest effect on viral gene expression and growth. Comparative analysis of viral mutants defective in various subsets of these IE genes should help elucidate how these proteins affect cellular and viral processes. This study focuses on the mutant d97, which is defective for the genes encoding ICP4, ICP0, and ICP27 and expresses the bacterial beta-galactosidase (beta-gal) gene from the ICP0 promoter. Together with the d92 virus (ICP4- ICP27-) and the ICP0-complementing cell line L7, d97 provided a unique opportunity to evaluate ICP0 function in the absence of the regulatory activities specified by ICP4 and ICP27. The pattern of protein synthesis in d97-infected cells was unique relative to other IE gene mutants in that it was similar to that seen in the absence of prior viral protein synthesis, possibly approximating the effect of cellular factors and virion components alone. Inactivation of ICP0 in the absence of ICP4 produced a significant decrease in the levels of the early mRNAs ICP6 and thymidine kinase (tk). There was also a marginal reduction in the levels of the IE ICP22 mRNA, and this was most notable at low multiplicity of infection (MOI). In d97-infected L7 cells, the levels of the viral mRNAs were mostly restored to those observed in infections with d92. Nuclear runoff transcription analysis demonstrated that the presence of ICP0 resulted in an increase in the transcription rates of the analyzed genes. The transcription rates of the early genes were dramatically reduced in the absence of ICP0. At low MOI, the transcription rates of ICP6 and tk were comparable to the rate of transcription of a cellular gene. Relevant to the potential use of d97 as a transfer vector, it was also determined that the absence of ICP0 reduced the cellular toxicity of the virus compared to that of d92. The beta-gal transgene expressed from an IE promoter was detected for up to 14 days postinfection; however, the level of beta-gal expression declined dramatically after 1 day postinfection. In the presence of ICP0, the level of expression of beta-gal was increased; however the infected monolayer was destroyed by 3 days postinfection. Therefore, deletion of ICP0 in the absence of ICP4 and ICP27 reduces toxicity and lowers the level of expression of genes from the viral genome.

[Interactions between selenium and cadmium on lipid peroxidation of encephalon in mice].

70 Kunming male mice were randomly divided into five groups. Three groups were injected intraperitoneally with selenium (Se) on the dosages of 0.00, 0.05 and 0.10 mg/kg body weight and other two groups were treated with 0.23 mg/kg cadmium (Cd) and 0.10 mg/kg Se plus 0.23 mg/kg Cd, for 50 days. The concentrations of glutathione (GSH) and malonic dialdehyde (MDA) of cerebellum, pons, corpora quadrigemina, thalamus and pallium were detected respectively. The results showed that the GSH concentrations in these tissues decreased in all exposed groups. Cd increased the MDA level significantly in pons, corpora quadrigemina and thalamus, and Se could antagonize the harmful effect induced by Cd. But Se itself could also decrease the GSH level of encephalon.

[Retinal detachment--severe complication after posterior scleral reinforcement operation].

OBJECTIVE: To study the severe complication of retinal detachment resulted from posterior scleral reinforcement operation. METHOD: The specialty of this type of retinal detachment through the clinical observation of 22 eyes of 20 cases was analyzed. RESULTS: The posterior scleral reinforcement surgery cannot achieve good effects on high myopia and has many complications especially retinal detachment. This type of retinal detachment is more severe than others. The retinal reattachment was obtained in 59.1% of cases in the immediate postoperative period. CONCLUSIONS: The posterior scleral reinforcement is an immature operative method, and it should not be widely applied.

[Studies on all-spectrum analysis for X-ray diffraction of Chinese herbal medicine calculus bovis].

Investigation on famouse Chinese herbal medicine-Niu huang (calculus bovis) was carried out by all-spectrum X-ray diffraction analysis. Diffraction spectrums, as well as the specific symboling peaks of calculus bovis, artificial bezoar, bile ductstone, human gallstone and hog gallstone, were recognized. The error distribution curves of d-delta d for specific symboling peaks was also obtained by calculation under diffrent testing conditions, by which we identified successfully three samples provided by a pharmaceutical factory. This article shows that all-spectrum X-ray diffraction analysis can be used to identify Chinese traditional crude drug, and provides for morphological and microscopical study.

[Adenosquamous carcinoma of the lung. A radiologic and pathologic study].

OBJECTIVE: To study if there are characteristic radiologic features of adenosquamous carcinoma (ASC) of the lung and correlations of histo-pathologic components with radiologic findings and prognosis. METHODS: Fifty one pathologicall confirmed ASC cases (52 foci) were reviewed. All had chest radiographs, CT scans were available in 23 cases and MRI in 6 cases. These cases were subdivided into three groups according to the amount of adenocarcinoma component on microscopic manifestations: less than 40%, 40%-60%, and more than 60%. RESULTS: (1) Radiologic findings: In three cases, the tumor was of central type and in 48 cases (94.1%) of peripheral type. The size of tumor varied from 2 to 15 cm in diameter (mean, 5.7 cm). All the peripheral tumors were lobulated. In 19 cases (39.6%) the tumors were well demarcated, but poorly defined in 18 cases (19 foci) (37.5%) and spiculated in 14 cases (29.2%). Eccentric cavities with thickened wall were seen in 8 cases (15.7%) and pleural tag in 12 cases (25%). (2) Correlations between the amount of adenocarcinoma component and radiologic features: There were no statistically significant differences among the three groups with regard to tumor location, margin, and pleural tag. CONCLUSION: The radiologic findings of ASC are not specific; the most common finding is a peripheral mass. The amount of adenocarcinoma component is not significantly related with the radiologic features and prognosis.

[Studies on X-ray diffraction pattern of traditional Chinese medicinal materials].

Traditional Chinese medicine(TCM) as prescribed by Chinese traditional physicians is usually available as "decoction slices" which calls for a ready method for assessment of quantity or even genuineness. Three groups of TCM drugs (radix Rubiae, bulbus Fritillariae and rhizoma Dioscoreae) were selected by using powder X-ray diffraction analysis with complete absorption profile. Topological regularity and characteristic peaks of the absorption profile were obtained as fingerprint in X-ray diffraction diagrams of the pertinent medicinal materials. We have discovered the similarities of 3 radix Rubiae samples gathered from different localities and their differences with Rubia tinctorum, the similarities and differences between 7 species belonging to Fritillaria Genus, the similarities of 2 rhizoma Dioscoreae samples from different localities and their differences with 2 counterfeits. The result points to a promising prospect in achieving an objective assessment of genuineness at TCM drugs.

[The structure correction of villosolside].

The crystal structure of villosolside was determined by X-ray diffraction, which led to some stereochemical amendment of the structure proposed in the article by Xu CJ et al. in Acta Pharm Sin 1985, 20:652.

[A further study on the correlation between molar response value and molecular connectivity indices].

It has been shown that there exists very good correlation between molar response values and corresponding molecular connectivity indices for alkane, benzene series and alkanol on both TCD and FID and for ketone on TCD. The results of multivariate linear regression are as follows. [Empirical expressions for the chromatographic peak width at inflection points and base].

From EMG function and linear regression, the projective distances on abscissa between inflection points in the left side (TL) or in the right side (TR) and the apex of a chromatographic peak which the asymmetry at 0.1 height fraction is within 1.09-3.00 can be expressed as: TL = -0.7898 + 0.8219 W(0.1,L), TR = 0.03536 + 0.4582 W(0.1,L), where W(0.1,L) is the projective distance on abscissa between the point at 1/10 height fraction in the left side and the apex. The correlation coefficients are all over 0.998 and the relative error limits are within -0.65%-0.40% and -0.39%-0.67%, respectively. The relationship between the peak width at base Wb and the peak width at half height W(0.5) or the peak width at 1/10 height fraction W(0.1) can be expressed as: Wb = 0.04827 + 1.6907 W(0.5), Wb = 1.9066 + 0.5300 W(0.1). The correlation coefficients are all over 0.999 and the relative error limits are within -0.54%-0.69% and -1.05%-1.40%, respectively.

Reliability of a modified tissue factor-dependent factor V assay for activated protein C resistant factor Va using a calcium-containing thromboplastin.

The original tissue factor-dependent factor V assay for activated protein C resistant factor Va (Blood 1995; 85: 1704-1711) has been modified to use a calcium containing thromboplastin and to express results as an observed to expected ratio (Obs/Exp.). The latter permits establishing a normal range independent of variations due to differences in reagents. Comparing Obs/Exp ratios with DNA analysis in 72 persons revealed that an Obs/Exp ratio of 0.6 distinguished without overlap normals from heterozygotes for FV R506Q. Three homozygotes had a ratio of < 0.1. Application of this Obs/Exp cut-off ratio of 0.6 to a total of 226 plasma samples tested to date discriminated without overlap between normals and heterozygotes. We conclude that this assay-readily adaptable to any dedicated coagulation laboratory and capable of yielding reliable results in all clinical circumstances in which testing is indicated-can distinguish between normals and heterozygotes for the FV R506Q mutation without the need for confirmatory DNA analysis.

Secondary prevention of cardiac events following myocardial infarction: effects of atenolol and enalapril. Beijing Collaborative Study Group.

OBJECTIVE: To investigate the actions of beta-blocker (atenolol) and ACE inhibitor (enalapril) for the secondary prevention of the main cardiac complications after acute myocardial infarction (AMI). METHODS: 1106 cases of AMI from 7 hospitals in the Beijing area were collected and were divided randomly into three groups: control (group C), atenolol (group A), and enalapril (group E). Drugs for investigation were administered 2-4 weeks after the onset of AMI, and the subjects were followed up for a median period of 19 months. All patients were given aspirin 50 mg/day. The end points of observation were cardiac events and non-cardiac events. Cardiac events included sudden cardiac death (SCD), heart failure death, total cardiac deaths, and myocardial re-infarction. RESULTS: The clinical conditions of the three groups were compatible. Sixty-six cardiac events (6.0%) occurred. Comparing with group C, the rate of SCD decreased significantly by 68% in group A after atenolol treatment for 28 months. Both atenolol and enalapril significantly increased left ventricular ejection fraction (LVEF), whereas in group C the LVEF did not change during the follow-up period. There was obvious decreasing tendency of the survival curve in group C, compared with the other two groups. Totally drugs decrease one cardiac death 0/00/month. But the rate of myocardial re-infarction was the same in the three groups. No serious side effects on blood pressure or heart rate were observed. CONCLUSIONS: Both atenolol and enalapril (domestic products) are beneficial to the secondary prevention of SCD and heart failure death after AMI, but not to re-infarction. Both drugs should be continued for a prolonged period to be effective. Drugs given 2-4 weeks after acute stage are also effective, with no serious side effects.

Detection of minimal residual disease in leukemia using polymerase chain reaction and restriction analysis.

OBJECTIVE: To investigate the relationship between the clinical outcome of childhood acute lymphoblastic leukemia and the status of minimal residual disease after termination of therapy. METHODS: A total of 18 children with acute lymphoblastic leukemia were studied. Eleven cases were treated with chemotherapy, the other 7 with autologous peripheral blood stem cell transplantation (ABSCT) or autologous bone marrow transplantation (ABMT). The outcome of the 18 patients was obtained by clinical follow-up. Minimal residual disease was detected by means of the polymerase chain reaction (PCR) and restriction analysis. RESULTS: Minimal residual disease was detected in 9 patients after completion of therapy. Three out of the 9 patients relapsed within 3 to 6 months. Among the other 9 patients with no detectable minimal residual disease, no one relapsed. CONCLUSIONS: PCR detection of minimal residual disease after termination of therapy can identify patients at risk for relapse in future. Minimal residual disease that cannot be detected after termination of therapy may be one of the important factors for long-term disease-free survival.

Electrically Conductive Crust in Southern Tibet from INDEPTH Magnetotelluric Surveying

The crust north of the Himalaya is generally electrically conductive below depths of 10 to 20 km. This conductive zone approaches the surface beneath the Kangmar dome (dipping north) and extends beneath the Zangbo suture. A profile crossing the northern Yadong-Gulu rift shows that the high conductivity region extends outside the rift, and its top within the rift coincides with a bright spot horizon imaged on the INDEPTH CMP (common midpoint) profiles. The high conductivity of the middle crust is atypical of stable continental regions and suggests that there is a regionally interconnected fluid phase in the crust of the region.

Clinical variability of Fanconi anemia (type C) results from expression of an amino terminal truncated Fanconi anemia complementation group C polypeptide with partial activity.

Fanconi anemia (FA) is an autosomal recessive disease characterized by congenital anomalies, aplastic anemia, and cancer susceptibility. Mutations within the FA complementation group C (FAC) gene account for approximately 14% of diagnosed FA cases. Two mutations, one in exon 1 (delG322) and one in exon 4 (IVS4 + 4 A to T), account for 90% of known FAC mutations. The delG322 mutation results in a mild FA phenotype, while the IVS4 + 4 A to T mutation results in severe FA phenotype. To determine the molecular basis for this clinical variability, we analyzed patient-derived cell lines for the expression of characteristic mutant FAC polypeptides. All cell lines with the delG322 mutation expressed a 50-kD FAC polypeptides, FRP-50 (FAC-related protein), shown to be an amino terminal truncated isoform of FAC reinitiated at methionine 55. All cell lines with the IVS4 + 4 A to T mutation lacked FRP-50. Overexpression of a cDNA encoding FRP-50 in an FA(C) cell line resulted in partial correction of mitomycin C sensitivity. In conclusion, expression of an amino terminal truncated FAC protein accounts, at least in part, for the clinical heterogeneity among FA(C) patients.

A scanning electron microscopy and computer image processing morphometric study of the pharmacological regulation of patency of the peritoneal stomata.

The experiment on mice was carried out by injecting intraperitoneally Chinese materia medica for treating hepatocirrhosis with ascites. Observations and a quantitative analysis were carried out on the pharmacological regulation of the peritoneal stomata by using a scanning electron microscope (SEM) and a computer image processing system attached to the SEM. There was a significant increase in both the diameter (P < 0.05) and distribution density (P < 0.01) of the peritoneal stomata in the red sage root and alismatis rhizome groups, whereas the effect of poria and poria peel was not significant compared with the control group (P > 0.05). Our findings confirm the effect of red sage root and alismatis rhizome on the regulation of the peritoneal stomata, which can enhance the absorption of ascitic fluid, taking into consideration the absorbent function of these stomata. They indicate that the patency of peritoneal stomata can vary in response to the effect of some Chinese materia. They also suggest that the ascites is drained mainly by means of enhancing the patency of the stomata and lymphatic absorption of the stomata during the process of treatment by traditional Chinese medicine.

Prolonged gene expression and cell survival after infection by a herpes simplex virus mutant defective in the immediate-early genes encoding ICP4, ICP27, and ICP22.

Very early in infection, herpes simplex virus (HSV) expresses four immediate-early (IE) regulatory proteins, ICP4, ICP0, ICP22, and ICP27. The systematic inactivation of sets of the IE proteins in cis, and the subsequent phenotypic analysis of the resulting mutants, should provide insights into how these proteins function in the HSV life cycle and also into the specific macromolecular events that are altered or perturbed in cells infected with virus strains blocked very early in infection. This approach may also provide a rational basis to assess the efficacy and safety of HSV mutants for use in gene transfer experiments. In this study, we generated and examined the phenotype of an HSV mutant simultaneously mutated in the ICP4, ICP27, and ICP22 genes of HSV. Unlike mutants deficient in ICP4 (d120), ICP4 and ICP27 (d92), and ICP4 and ICP22 (d96), mutants defective in ICP4, ICP27, and ICP22 (d95) were visually much less toxic to Vero and human embryonic lung cells. Cells infected with d95 at a multiplicity of infection of 10 PFU per cell retained a relatively normal morphology and expressed genes from the viral and cellular genomes for at least 3 days postinfection. The other mutant backgrounds were too toxic to allow examination of gene expression past 1 day postinfection. However, when cell survival was measured by the capacity of the infected cells to form colonies, d95 inhibited colony formation similarly to d92. This apparent paradox was reconciled by the observation that host cell DNA synthesis was inhibited in cells infected with d120, d92, d96, and d95. In addition, all of the mutants exhibited pronounced and distinctive alterations in nuclear morphology, as determined by electron microscopy. The appearance of d95-infected cells deviated from that of uninfected cells in that large circular structures formed in the nucleus. d95-infected cells abundantly expressed ICP0, which accumulated in fine punctate structures in the nucleus at early times postinfection and coalesced or grew to the large circular objects that were revealed by electron microscopy. Therefore, while the abundant accumulation of ICPO in the absence of ICP4, ICP22, and ICP27 may allow for prolonged gene expression, cell survival is impaired, in part, as a result of the inhibition of cellular DNA synthesis.

[Effect of dexamethasone on the human interleukin-2 receptor alpha chain gene expression].

Glucocorticoids have wide effects on the immune system, the mechanisms of these effects have not been clearly defined. The interaction of interleukin-2 (IL-2) with its receptor (IL-2R) is a critical control point in the T-cell mediated immune response. The effect of dexamethasone, a synthetic glucocorticoid hormone, on IL-2R is unclear in comparison with its inhibition on the accumulation of IL-2 mRNA. Our study shows that dexamethasone increases the basal and PHA-induced mRNA levels of IL-2R alpha gene in Jurkat cells, but it does not affect the expression of reporter gene constructs containing 5' flanking sequences (-482bp/ +16bp or -350bp/+16bp) of the IL-2R alpha gene in PHA activated Jurkat cells. These results indicate that dexamethasone plays a positive regulatory role in the IL-2R alpha gene expression, but the effect is not mediated by the previously identified proximal regulatory elements located between -482bp/+1bp. Meanwhile, our results also provide evidence that the activation mechanism of dexamethasone is different from that of PHA, and that dexamethasone and PHA show a synergistic effect in the induction of IL-2R alpha gene expression.

[Relationship between heart rate variability and clinical features and prognosis of acute myocardial infarction].

Heart rate variability (HRV) was studied prospectively in 118 consecutive patients with acute myocardial infarction (AMI). HRV in AMI patients was significantly lower than that in normal controls, both at the 19th hour and the end of the 3rd week after the onset of AMI. Moreover, HRV at the end of 3rd week HRV in patients with cardiac death was also lower than that in the survivals. Measurements of ultra-low frequency power, total frequency power, very low frequency power, and the average of the standard deviation of every 5-minute period were independent predictors of cardiac death. All these 4 measurements have highly significant correlation (P < 0.01) and are better predictors of prognosis than other clinical variables. It is revealed that HRV at the end of 3rd week after the onset of AMI has much greater prognostic value than that at the initial period.
