The immune adherence phenomenon in experimental candidiasis.

The immune adherence reaction was shown in experimental candidiasis. This phenomenon has not been previously described for candidiasis, to the best of our knowledge. Forty-eight sera of guinea pigs inoculated with a single or several inoculations of live Candida albicans were tested at different time intervals after inoculation for anti C. albicans antibodies detectable by the immune adherence reaction. From the 14th day after inoculation, a positive immune adherence reaction was observed with titer and strength increasing as time passed after inoculation and as the number of inoculations increased. All the sera were tested parallely in agglutination and indirect mast cell degranulation tests. The significance of the data obtained is discussed.

Mast cell sensitizing antibodies in experimental candidiasis.

Inoculation into foot-pads of guinea-pigs of a living suspension of Candida albicans together with Complete Freund's Adjuvant, provoked a hypersensitive state, characterized by: 1) positive skin tests; 2) direct degranulation of peritoneal mast cells when exposed in vitro to the antigen; and 3) homologous "mast cell sensitizing antibodies" detected by the "indirect mast cell degranulation test". The mast cell sensitizing antibodies were heat labile, and their activity could not be restored by complement.

Mast cell sensitizing antibody (MCSAb) response in experimental candidiasis: chromatographic studies.

A single dose of the yeast-like fungus Candida albicans (CA), mixed with Complete Freund's Adjuvant (CFA), and inoculated into guinea-pig foot-pads, provoked the productions of homologous mast cell sensitizing antibodies (MCSAb) present in the psiI-globulin fraction only, and of agglutinins localized in the same fraction. Heterologous MCSAb and skin-sensitizing antibodies detectable by passive cutaneous anaphylaxis (PCA) were constantly absent. These facts are in contradiction to results of previous experiments performed with other antigens; which after similar inoculation elicited production of heterologous MCSAb attaching themselves on rat mast cells (MC) and present in the psi2 fast moving globulin, homologous MCSAb localized in the psiI-globulin and in the psi2 fast moving globulin, and PCA antibodies. The immunogenicity of CA is discussed.

Antigenic relationships between Candida albicans and various yeasts as reflected by immunoglobulin-class specificity.

A group of guinea pigs was inoculated into the foot pads with a single dose of Candida albicans in complete Freund's adjuvant, while another group was similarly inoculated once in the foot pads but also several times intramuscularly, with Candida alone. All guinea pigs were bled at different intervals after immunization and sera were separated chromatographically into IgG and IgM fractions. In order to study the antigenic relationships as reflected by immunoglobulin-class specificity, IgG and IgM fractions and whole sera obtained from guinea pigs differently immunized, were tested for the presence of agglutinins against C. albicans, six other species of Candida, and species of the ascosporogenous genera Saccharomyces, Kluyveromyces and Schizosaccharomyces. The results show that (1) only IgG fractions of the different sera prepared contained the specific anti-C. albicans antibodies; (2) IgG and IgM fractions of the sera obtained from a single inoculation did not reveal a specific pattern expressing antigenic relationships of the yeast studied, and (3) the IgM fractions of the sera obtained from several inoculations had a more homogenous pattern of reactivity, since mainly these contained the agglutinins against the ascosporogenous yeast species.

Mast cell sensitizing antibodies in myasthenia gravis.

Mast Cells Sensitizing Antibodies (MCSAb) were detected in patients with Myasthenia Gravis (MGr), by the Indirect Mast Cell Degranulation (IMCD). In most of the examined sera a correlation could be established between MCSAb and fluorescent antibodies to muscle and to thymus. The MCSAb were specific for muscle extract. They were resistant to prolonged heating and their reactivity in the IMCD test was not complement depending. Presence of MCSAb in sera of patients with MGr evidences the auto-allergic features of this disease.

Dementia and antiphospholipid antibodies.

Antiphospholipid antibodies (aPLAb) may cause both focal ischemic and diffuse brain damage and may be associated with dementia. We have examined the relationship of aPLAb to dementia in the elderly. Blood samples were obtained from 87 consecutive patients with dementia (74 +/- 11 years old) and 69 controls (78 +/- 9 years old), residents of an old age home who were not overtly demented. Levels of aPLAb were measured by a standardized ELISA, utilizing cardiolipin as antigen, and we considered levels above 20 IgG antiphospholipid units (GPLU) as significantly elevated. We found that 5 of the 87 demented patients (6%), but none of the 69 controls, had significantly elevated aPLAb levels (p = 0.03, one-tailed Fisher's exact test). All the patients with high aPLAb levels were diagnosed clinically as having dementia of the Alzheimer type, except for 1 who had mixed dementia, and none had features of an immune-mediated disease. Thus, a small but significant number of patients with dementia have high levels of aPLAb. The role of the aPLAb in these patients, with apparently diffuse brain disease, is currently unknown.

Felty's syndrome without concomitant arthritis: a variant.

A young female patient who had documented seropositive rheumatoid arthritis and was treated consecutively with aspirin, diclofenac, and gold salts was admitted years later for severe neutropenia. On examination she had, in addition, fever, positive rheumatoid factor, reversible swan-neck deformity of the fingers but otherwise normal joint findings. The patient responded to prednisone therapy. This case would appear to be a most unusual variant of Felty's syndrome.

Lack of exposure to HTLV1 among Ethiopian immigrants of operation Solomon (1991) arriving to the Jerusalem area.

HTLV1 antibodies were tested in a group of 740 Ethiopian immigrants who arrived in Israel in Operation Solomon, 1991. HTLV1 antibodies were not found in a single individual. This survey and previous ones conducted on 314 immigrants of the 1984-85 Operation Moses suggest that HTLV1 infection is not present among Ethiopian immigrants residing in Israel.
