Lipid peroxidation in malignant tumors of human kidneys.

The intensity of ascorbate-dependent free radical oxidation of endogenous lipids in malignant tumor tissue of human kidneys was studied. The LPO induction period was markedly increased in renal-cell carcinoma and malignant mesenchymal tumors compared to normal renal cortex. The content of alpha-tocopherol in cancer tissue lipids was considerably (7-fold) increased compared to that in normal renal cortex, which explains high antioxidant activity of these carcinomas. A less pronounced increase in the content of alpha -tocopherol in lipids of malignant mesenchymal tumors compared to the cortex (2-fold) can also contribute to their LPO resistance.

[Accidental involution of the thymus after splenectomy (an experimental study)].

Using immunological, general clinical and luminescent histochemical methods experimental study of immunological status and bioamine-containing thymus structures was performed on third, seventh and fifteenth postsplenectomy days in 90 outbred albino male rats. Splenectomy leads to significant changes in blood immunological parameters and morphofunctional state of thymus bioamine-containing structures. Significant increase of biogenic amines content in all thymus structures, leukocyte number, complement activity neutrophilic granulocytes phagocytic activity, were registered in 3 days. Further growth of bioamine content, decrease of cortical and medullary substances diameter, significant number growth of juvenile forms of mast cells were observed in 7 days. Essential decline in erythrocyte number, hemoglobin and IgG and IgA content, leukocytosis with leukemoid reaction development were defined in blood formula. On 15th postsplenectomy day maximal biogenic amine concentration, especially histamine was reached and further reduction in cortex and medulla as well as decline of all the immunoglobulins and decrease of neutrophilic granulocytes phagocytosis were determined. Thus, splenectomy and subsequent development of accidental involution leads to secondary immunodeficiency and severe anemia.

[Marker proteins in cerebrospinal fluid of patients with grave craniocerebral injury].

The permeability of the blood-brain barrier for the cerebrospinal fluid marker proteins has been assessed in patients with grave craniocerebral injuries. The content of albumin and alpha 2-macroglobulin in the survivors decreased with time and by day 7 after the injury was virtually normal. In patients who died the content of these proteins was reliably increased starting from day 1. By day 7 the content of albumin in the cerebrospinal fluid approached the norm in this group, whereas the content of alpha 2-macroglobulin remained increased. The level of IgG surpassed the normal value on day 1 only in the patients who died, and later it did not differ from the norm in both groups.

[Kidney involvement and arterial hypertension in middle-aged and elderly subjects].

The examination covered 125 elderly and senile subjects (106 females and 19 males) which made up 90.6% of those living in the boarding house. Their mean age was 81.3 years. Arterial hypertension was detected in 61.6% of the examinees. 18.5% of the examinees had hyperuricemia. Concentration function of the kidneys was affected in all the examinees. Half of them exhibited a moderate rise in the level of blood creatinine. The authors discuss the relationship between high prevalence of arterial hypertension, its tendency to crises and involutional processes in the kidneys in the onset and development of renoprival condition.

Liposoluble vitamins E and A in human renal cortex and renal cell carcinomas.

Vitamin E was quantified in renal cell carcinomas (RCC) and in 'intact' renal cortex, obtained from 31 patients subjected either to unilateral nephrectomy or to partial resection of the only kidney. Histologically, 14 tumors consisted predominantly of clear cells (group 1) and 17 of other cell types (group 2). In both groups, a significant increase in vitamin E concentration, as compared to the 'intact' cortex, was observed: 167.8 +/- 27.9 and 68.2 +/- 15.2 micrograms/g wet tissue weight (mean +/- SEM) for groups 1 and 2, respectively, versus 10.1 +/- 0.53 micrograms/g wet tissue weight for the cortex. Although the total lipid content was also increased in tumors (especially in group 1), the vitamin E concentration in tumor tissue, calculated per milligram of total lipids, proved to be much higher in both groups than in 'intact' cortex. A significant positive correlation was observed between vitamin E and total lipid content in group 1 and 2 carcinomas. It was also found that vitamin E accumulation in RCC is unlikely to be attributed to an enhanced lipid deposit in the tumor cells. Thus, in 8 tumors of group 2 the vitamin E levels were markedly enhanced although these tumors did not differ from the cortex in total lipid concentrations. Vitamin A content determined in 17 carcinomas, when calculated per milligram of total lipids, was the same as in 'intact' cortex.

[Vitamin E content in cortical and medullary layers of mammalian kidney].

Vitamin E was quantified in the cortex and medulla of human, rat, canine and rabbit kidneys. For the all species investigated the vitamin concentrations, calculated both per g wet tissue weight and per mg tissue protein, was shown to be 1.4-2.2 times higher in medullary layer than in cortical one. The least vitamin E levels were observed in rabbit kidneys of which the cortical and medullary concentrations were 4.4 +/- 0.3 mkg (M +/- SEM) and 6.3 +/- 0.56 mkg per g wet tissue weight, respectively. In human renal cortex and medulla the vitamin concentrations were found to be 10.6 +/- 0.66 mkg and 16.2 +/- 1.1 mkg per g wet tissue weight, respectively. Close values were observed for other species studied as well. The increased renal medullary vitamin E concentrations are likely related to the higher phospholipid metabolism and enhanced prostaglandin synthesis in this tissue.

[Content of vitamins E and A in tumor tissue in kidney cancer].

Vitamin E was quantified in tumor tissue obtained from 17 patients with renal cell carcinoma (RCC). The tumors consisted of two groups: (I) represented primarily by clear cells (10 cases) and (II) without predominant appearance of those cells (7 cases). Vitamin E concentration in tumor tissue was compared with that in intact tissue sites of the kidney. The vitamin content in intact renal medulla was shown to be 1.5 times higher than in intact cortex. Vitamin E level in group I tumors was shown to be on average 15 and 10 times as much as in intact renal cortex and medulla, respectively. The vitamin concentration in group II tumors, as compared with the above intact renal tissues, was 7- and 5-fold higher, respectively. In spite of the high tumor total lipids levels, the vitamin E concentration, calculated per mg total lipids, was still higher in tumor tissue than in the normal renal tissues. This finding was especially demonstrable for the group I tumors. In the benign renal tumor angiomyolipoma vitamin E concentration, calculated on a wet weight basis, was higher than in intact renal tissue, but no differences were observed when a per total lipids calculation was performed. Vitamin A concentration determined in the tumor samples obtained from 6 patients was the same as in intact cortex.

[Immunohistochemical characteristics of serum nephrotoxic nephritis in rats: effect of methylprednisolone and potassium fenozan].

Late heterologous and autologous stages of nephrotoxic serum nephritis were studied against the background of MP-induced immunological suppression and PP effect on arachidonic acid metabolism. MP prevented mononuclear infiltration of the renal glomerulus and the development of severe morphological lesions in the autologous stage, while cell culture and mononuclear leukocyte number drastically decreased. PP did not significantly influence the autologous stage of the nephritis though resulted in marked proliferation in both renal glomerulus and mesangial cell culture during the heterologous stage. In spite of more pronounced proliferative activity of mesangial cells interleukin-I level was lower than in animals with nephritis untreated with PP. The enhancement of the proliferative reaction is obviously related, apart from interleukin-I, to the effect of other growth factors.

[Immunohistochemical features of nephrotoxic serum nephritis in the rat: the role of interleukin-1 in the development of heterologous and autologous stages].

Late heterologous and autologous stages of serum nephritis together with glomerular cells in culture are studied biochemically, morphologically and immunohistochemically for the assessment of the role of mononuclear leucocyte mediator system. The heterologous stage of the nephrotoxic nephritis is associated with the increase of Interleukin-I and TxA2 production. Role of Interleukin-I in the autologous stage is less pronounced in spite of proliferative activity of cultured mesangial cells. The effect of suppressive factors prevails in the glomerulus.

[Lipid peroxidation in the kidneys of rats with nephritis induced by nephrotoxic serum and proteinuria induced by albumin overload].

Lipid peroxidation (LPO) stimulated by ascorbate was studied in renal cortex of 20 rats with nephrotoxic nephritis (NTN) and of 9 rats with proteinuria induced by a 3-day course of i. p. injections of the human serum albumin. At the early stages of NTN (0.5 h. and 3 h.) LPO activities were of the same values as in control rats. A small decrease in renal cortex LPO was found on the 4-th day of NTN when nephrotic syndrome has been developed. A significant reduction in LPO activity was observed on the 16-th day of NTN characterized by a more pronounced nephrotic syndrome. LPO activity in renal cortex of the rats with albumin overload proteinuria was also reduced. An inhibitory effect of proteinuria on LPO activity in kidney is discussed.

[Interleukin-1 production by mononuclear leukocytes and mesangial cells in experimental nephrotoxic nephritis].

We investigated the intensiveness of the cell proliferation, number of mononuclear leucocytes (MNL) and production of interleukin-1 (IL-1) in glomerular cell cultures of rats with nephrotoxic serum (NTS) nephritis and control (C) rats. Five days after intravenous injection of nephrotoxic serum from rabbits the glomeruli were isolated, treated with collagenase and cultured over a period of 20 days. In cultures the number and intensity of mesangial cell proliferation in the NTS group were significantly higher than in the C group. Thus, the intensive mesangial cell proliferation in experimental nephrotoxic serum nephritis is related to infiltration of glomerulus by MNL of bone marrow origin with increased production of IL-1.

[Urinary enzymes induced in rats by intensive physical loading and the effect of the antioxidant potassium fenozan].

The rats subjected to repeated heavy exercises (swimming with the load equal to 10% of body weight) had an increased activity of gamma-glutamyl transpeptidase in the urine. Both the percentage of the rats with hyperenzymuria and the level of enzyme were the highest after the fifth swimming. Water-soluble antioxidant (potassium fenozan) administered intraperitoneally 30 minutes before the swimming abolished the increase of enzymuria and maintained the enzyme activity to the normal level.

[Current possibilities of correction of hormonal imbalance in uremia].

The authors' and literature data on hormonal imbalance in uremia are analysed. The pathogenesis, clinical and laboratory signs and present-day methods of correction of renal osteodystrophy, anemia, arterial hypertension, derangements of carbohydrate and lipid metabolism, and hypogonadism developing as complications of chronic renal insufficiency (CRI), are considered. Increased potentialities of pharmacological correction of the above endocrine disturbances in CRI have been shown.

[Phosphorus and calcium metabolism and its hormonal regulation in chronic kidney failure].

In order to elucidate the blood serum calcium, inorganic phosphorus, parathyroid hormone (PTH) and calcitonin (CT) content in the course of the development of chronic renal failure (CRF), 80 patients with chronile glomerulonephritis were examined. Of these, in 24 glomerular filtration (GF) was normal, whereas 56 had CRF of varying degree. Thirty-three out of the 80 patients had the nephrotic syndrome (NS). The parameters enumerated were also determined in 45 patients with CRF treated by hemodialysis. In patients without the NS, hypocalcemia was discovered only at the final predialysis stage of CRF, while in the presence of the NS, it was detectable in normal GF and was aggravated as CRF progressed. The PTH level ascended if GF was lower than 40 ml/min, not correlating with calcium concentration in the serum. A direct correlation was disclosed between the PTH level and that of inorganic phosphorus. The role of the latter in stimulation of PTH and CT secretion in CRF is discussed. In patients treated by hemodialysis, the PTH level significantly rose with treatment time increase. There was also a tendency toward elevation of CT concentration. The activity of the osseous fraction of blood serum alkaline phosphatase in patients treated by hemodialysis was increased in 61-84% of cases depending on the treatment period.

[Masugi's nephritis: preparation of an active nephrotoxic serum].

An active nephrotoxic serum (NTS) was prepared by immunization of rabbits with isolated renal glomeruli of rats and Freund's complete adjuvant. The glomeruli were isolated from the renal cortex by the sieving procedure. Intravenous injection of the NTS to rats induced the development of marked proteinuria and the nephrotic syndrome. Histological and electron microscopic studies of renal tissue performed by days 5 and 15 since the first injection of the NTS revealed extracapillary proliferative glomerulonephritis. IgG depositions on the basal membranes of the glomerular capillaries were linear on day 5 and granular on day 15 since the first injection of the NTS.
