In situ spatial organization of Potato virus A coat protein subunits as assessed by tritium bombardment.

Potato virus A (PVA) particles were bombarded with thermally activated tritium atoms, and the intramolecular distribution of the label in the amino acids of the coat protein was determined to assess their in situ steric accessibility. This method revealed that the N-terminal 15 amino acids of the PVA coat protein and a region comprising amino acids 27 to 50 are the most accessible at the particle surface to labeling with tritium atoms. A model of the spatial arrangement of the PVA coat protein polypeptide chain within the virus particle was derived from the experimental data obtained by tritium bombardment combined with predictions of secondary-structure elements and the principles of packing alpha-helices and beta-structures in proteins. The model predicts three regions of tertiary structure: (i) the surface-exposed N-terminal region, comprising an unstructured N terminus of 8 amino acids and two beta-strands, (ii) a C-terminal region including two alpha-helices, as well as three beta-strands that form a two-layer structure called an abCd unit, and (iii) a central region comprising a bundle of four alpha-helices in a fold similar to that found in tobacco mosaic virus coat protein. This is the first model of the three-dimensional structure of a potyvirus coat protein.

The in situ spatial arrangement of the influenza A virus matrix protein M1 assessed by tritium bombardment.

Intact influenza A virions were bombarded with thermally activated tritium atoms, and the intramolecular distribution of the label in the matrix protein M1 was analyzed to determine the in situ accessibility of its tryptic fragments. These data were combined with the previously reported x-ray crystal structure of the M1 fragment 2-158 [Sha, B. & Luo, M. (1997) Nat. Struct. Biol. 4, 239-244] and the predicted topology of the C domain (159-252) to propose a model of M1 arrangement in the virus particle.

Covalent chromatography of influenza virus membrane M1 protein on activated thiopropyl Sepharose-6B.

The M1 protein of influenza virus is a highly hydrophobic polypeptide that is resistant to enzyme cleavage during incubation in water solutions. We show here that the M1 protein that is immobilized on an insoluble activated support (thiopropyl Sepharose-6B) by means of a thiol-disulfide exchange reaction acquires sensitivity to trypsin. After tryptic digestion noncysteine-containing peptides of M1 were removed by washing the support, while cysteine-containing ones were detached from the support by reduction. As a result, 24 unique tryptic peptides of M1 protein were clearly separated by reversed-phase high-performance liquid chromatography. The described method opens a new way to the investigation of functional properties of distinct domains of viral thiol proteins.

Scale of Quality of Life of Care-Givers (SQLC).

There is growing interest today in the design and application of tools for measuring quality of life. However, the rating scales applied do not reveal the consequences that Parkinson's disease has for patients' relatives. The need for permanent care and help to be given to the chronically disabled person leads to a decrease in the life quality of caregivers. The contribution describes an inventory for evaluating the quality of life of parkinsonian patients' caregivers and analyses some factors determining their social disadaptation.

[Biochemical criteria for assessing the severity of the status of wounded patients with gunshot damages to the chest and abdomen].

The biochemical assessment of severity was made in 34 patients with thoracic and abdominal gunshot wounds (12 uncomplicated; 11 with purulent, and 11 non-purulent complications). The levels of hormones, biogenic amines, lipid peroxidation (LP) indices, and antioxidant system (AOS) in serum were studied in 1, 3, 7, 14 days after admission. It was documented that the integral indices: cortisol/insulin and LP/AOS ratios were the most informative biochemical criteria for the assessment of severity of the injured patients. A high informative significance of these indices in the assessment of severity of homeostatic disturbances is pathogenetically substantiated due to stress-realizing (cortisol and free-radical LP components) and stress-limiting (insulin and AOS components) factors.

[Use of neurotransplantation in the treatment of Parkinson's disease].

The outcomes of transplantation of the human embryonal mid-brain into the striatum of patients with Parkinson's disease are presented. For this, a procedure for obtaining, isolating, preparing the human embryonic midbrain, as well as a method for stereotactic administration of embryonal nerve tissue suspension having high dopaminergic neuron levels into the patients' striatum are specified. Patients underwent neurological examinations by the international protocol Core Assessment Program for Intracerebral Transplantation including monthly video recording within 3 months before and 3-24 months after surgery. Electrophysiological, neuropsychological, and immunological examinations were made at the same intervals. Most patients had positive dynamics: diminution of the severity of main disease symptoms, reduction in the efficiency of a single dose of DOPA-containing drugs, decrease in the magnitude of side effects of drug therapy.

[A method for the self-assessment of social adaptation in parkinsonism patients].

The rating scale of self-estimation of social adaptation which permits to determine the patient's adaptation in three life spheres (professional-working, family and self-service) was described. 103 patients with different forms of parkinsonism were examined by means of this scale. High effectiveness of this scale is demonstrated for estimation of the influence of such parameters as the disease gravity, its clinical form, mnemic-intellectual disorders expression, age and the duration of disease on social adaptation. Such effect determines the validity of the scale for the prognosis of the disease development.

[The efficacy of the Umex treatment of parkinsonism patients in the initial stage of the disease].

18 patients with early parkinsonism presenting in different clinical forms have undergone 3-month treatment with umex (10 mg/day). The response was evaluated in accordance with unified scale assessing the condition, motor tests, personal and reactive anxiety. Umex brought a significant improvement of motor activity, prolonged the effect of a single dose of DOPA-containing medication, reduced anxiety. Side effects were insignificant.

A model for the study of the mechanism of a low pH-induced interaction of the virus fusion proteins and cell membranes.

A model is proposed for the study of molecular mechanisms of a low pH-induced interaction of fusion proteins of enveloped viruses and cell membranes. The model consists of large monolamellar liposomes containing ionophore nigericin in their membranes and ectodomains of fusion protein in their inner space. The process of interaction of the protein with the lipid bilayer is triggered by acidification of the liposomal constituents to the pH of fusion with the help of nigericin by adding citric acid to the outer medium. To visualize the protein structural reorganization, the tritium planigraphy was used. Comparison of the values of specific labelling of the proteins and distribution of radioactivity in individual amino acids in control (at neutral pH) and experimental liposome samples (at the pH of fusion) permits to realise the character of protein-membrane interaction. We have obtained the first results in the study of interaction of the bromelain-released soluble ectodomain of the HAXX molecule (BHA)--with the lipid membrane. The observed increase in the protein specific activity and selective increase in the specific activity of hydrophobic amino acids Ile, Phe and Tyr in experimental liposome samples as compared with the controls did not contradict to the conventional concept, that a hydrophobic N-terminus of HA2 subunit of hemagglutinin is responsible for its interaction with lipid membranes.

[Changes in hemostasis indices in patients with gastrointestinal hemorrhages depending on treatment methods].

The dynamics of changes in some values of the hemostatic system was studied on days 1, 3, and 7 after gastrointestinal hemorrhage (GIH) of ulcerous etiology during enteral (administration of an electrolyte-monomer solution through a tube) and parenteral (traditional administration of infusion solutions) treatment. In operated on patients with enteral correction marked hyperfibrinogenemia was revealed, as well as diminished blood fibrinolytic activity, prolonged thrombin time, and a positive ethanol test, which indicates the predominance of hypercoagulation changes in the blood. Patients who were not subjected to operation but were given enteral correction had marked thrombocytopenia, reduced tolerance of plasma to heparin in all test periods tendency towards reduction of the fibrinogen content during the whole week after the hemorrhage in normal values of fibrinolytic activity, positive ethanol and protamine sulfate tests, which is evidence of hypocoagulation shifts in the hemostatic system. Reduction of the 4th thrombocytic factor, a tendency towards diminution of plasma tolerance to heparin, normal fibrinogen level, reduced blood fibrinolytic activity, and negative ethanol and protamine sulfate tests were revealed in patients who did not undergo operation and were given the traditional parenteral treatment, which reflects the compensatory character of the blood coagulation system response to rapidly developing blood loss. Disorders of the hemostatic system were more marked in patients with GIH who were not operated on and were given enteral correction, who require additional therapy.

[Clinico-rheographic study of the cerebrovascular effect of alpha-adrenergic blockers in vascular diseases of the brain].

The effect of the alpha-adrenoblockers, nicergoline (N) and dihydroergotoxin (DHET), on the cerebral vessels and the systemic hemodynamics was studied in 152 patients with acute and chronic vascular diseases of the brain. It was established that the hypotensive action of alpha-blockers was the greater the higher was the initial arterial hypertension. REG conducted during an hour after the intravenous administration of N and DHET revealed an increase in the pulse blood filling and an improvement of the arterial tone. Changes in vascular resistance were heterogeneous. Both drugs induced the venous outflow but there were no signs of venous hypotension. An improvement in the systemic and cerebral hemodynamics correlated with clinical improvement.

[Role of changes in blood serotonin and histamine concentrations in the pathogenesis of pneumonia in burns].

Sixty patients with associated burn disease and pneumonia were examined for the time course of the content of serotonin and histamine in the blood. In patients with pneumonia, the concentration of biogenic amines in the blood was substantially higher than in healthy subjects and in patients with similar burns as regards the severity without pneumonia. High concentration of biogenic amines in the blood was discovered to lead to functional and morphological lesions in the lungs favouring pneumonia development. High content of serotonin and histamine in the blood ascertained immediately after burns allows for the possibility of pneumonia development.
