Molecular size difference of urinary heparan sulfates from normal individuals and genetic mucopolysaccharidoses.

Heparan sulfates were isolated from the urine of normal individuals and patients with genetic mucopolysaccharidoses after exhaustive digestion with chondroitinase ABC. Electrophoresis of these preparations on cellulose acetate membrane revealed one spot corresponding in mobility to reference heparan sulphate in barium acetate buffer, while electrophoresis in 0.1 M HCl resulted in two distinct spots for each case; one corresponded in migration rate to reference heparan sulfate, and the other was faster in mobility than reference heparan sulfate but slightly retarded when compared with reference heparin. On thin-layer gel filtration on Sephadex G-200 (superfine) heparan sulfate from normal urine was polydispersed in character and its molecular size was larger than those of other preparations. Heparan sulfates from Hunter's and Sanfilippo's urine were monodispersed and small in molecular size. The molecular size of heparan sulfate from Sanfilippo's urine was the smallest of all. Heparin sulfate from Hurler's urine appeared to be composed of two populations; one corresponded in molecular size to heparan sulfate from normal urine, and the other corresponded to that of Hunter's urine.

Carbonic anhydrase isozymes, hemoglobin-F and glutathione levels in lead-exposed workers.

Red cell carbonic anhydrase B and C isozymes and hemoglobin F were studied by the single radial immunodiffusion technique in lead workers exposed to low concentrations of lead. The lead contents, delta-aminolevulinic acid dehydrase and glutathione levels were simultaneously determined. Carbonic anhydrase B, glutathione and delta-aminolevulinic acid dehydrase activity in lead-exposed workers showed relatively low values compared to those of normal subjects not exposed to occupational lead exposure in the past or present. Subjects with blood lead contents of 30 mug per 100 ml or higher showed a significant increase in hemoglobin-F and a significant decrease in carbonic anhydrase B and glutathione levels. These findings indicate that the syntheses of these red cell components are affected by lead, and hence in addition to delta-aminolevulinic acid dehydrase activity, these components were also found to be of clinical value for the diagnosis of subclinical lead poisoning.

Human erythrocyte carbonic anhydrase B and C in chronic obstructive lung disease.

The levels of carbonic anhydrase B and C isozymes in human red cells were determined using a quantitative immunological technique in patients with chronic obstructive lung disease. A significant increase in the level of carbonic anhydrase B was observed in these patients, while the level of carbonic anhydrase C did not change substantially. Positive correlations were found between the level of carbonic anhydrase B and arterial CO2 tension and plasma HCO3 concentration. A negative correlation was observed between the levels of carbonic anhydrase B and blood pH. These findings suggest that the synthesis or degradation of carbonic anhydrase B isozyme is affected by arterial CO2 tension or plasma HC03 concentration. The clinical significance was also discussed in relation to these isozyme levels in red cell.
