1,2,3,4-Tetrahydroisoquinoline (TIQ) was quantitated by use of gas chromatography-mass spectrometry in brains and livers of marmosets which showed parkinsonism after daily subcutaneous injection of TIQ. TIQ showed greatly increased levels in the brains and livers of the TIQ-treated marmosets, with no detectable metabolites of TIQ. TIQ was present as an endogenous amine in the brains and livers of saline-treated marmosets at very low concentrations. It thus seems that TIQ can pass easily through the blood-brain barrier but cannot be metabolized in the brain or the liver. It is possible that TIQ accumulated in the brain may produce parkinsonism.

Multicentricity of hepatocellular carcinoma (HCC) is considered, especially in patients with liver cirrhosis. This paper describes an operative case of a male patient with probable multicentric development of HCC in precirrhotic fibrosis. The main tumors inside the capsule were completely necrotic due to transarterial embolization (TAE). Histologic examination disclosed discrete tiny nodules of HCC that were not detected grossly. They showed highly differentiated trabecular arrangements: Edmondson I. A scirrhous type was noted in the center of the tumors. At the borders of the tumors the carcinoma cells exhibited replacing growth patterns and it was thought that they developed multicentrically. Partial resection must be performed as extensively as possible in a case such as the present one.
The present immunocytochemical study has demonstrated the presence of a dense fiber plexus containing calcitonin gene-related peptide (CGRP) in the vestibular end-organs in the rat. These fibers appear to originate from the central nervous system.
3-carboxy-4-methyl-5-propyl-2-furanpropionic acid, 3-carboxy-4-methyl-5-pentyl-2-furanpropionic acid, 3-carboxy-4-methyl-5-ethyl-2-furanpropionic acid and 3-carboxy-5-propyl-2-furanpropionic acid were detected in uremic serum using gas chromatography-mass spectrometry. Mass chromatography revealed that the serum concentrations of the furancarboxylic acids especially 3-carboxy-4-methyl-5-propyl-2-furanpropionic acid, were increased in the chronic hemodialysis patients and that the acids could not be removed by conventional hemodialysis due to their strong binding to plasma protein. 3-Carboxy-4-methyl-5-propyl-2-furanpropionic acid was also quantitated in uremic serum by high-performance liquid chromatography. Serum level of the acid in uremic patients showed significant but weak correlation with serum level of urea and duration on hemodialysis. Equilibrium dialysis demonstrated that 3-carboxy-4-methyl-5-propyl-2-furanpropionic acid and 3-carboxy-4-methyl-5-pentyl-2-furanpropionic acid inhibited the bindingof salicylate and 5,5-diphenylhydantoin to albumin. In conclusion, the furancarboxylic acids especially 3-carboxy-4-methyl-5-propyl-2-furanpropionic acid were accumulated in uremic serum as inhibitors of drug binding.
A 14-year-old boy had persistent haematuria along with complete C7 deficiency. No significant changes in glomeruli and tubules were found in a renal biopsy specimen by light microscopy and immunofluorescence gave negative results for immune deposits. Electron microscopic examination demonstrated an attenuation of the glomerular capillary basement membrane without lamination and a diagnosis of thin basement membrane disease was made. It would be difficult to conclude that patients with C7 deficiency were predisposed to develop glomerulonephritis caused by immunologic aberrations. A family study failed to provide evidence of an association of C7 deficiency and thin basement membrane disease.
Eleven gliomas were serially cultivated and examined for DNA distribution by flow cytometry and simultaneously for morphological features by light microscopy at the various passage levels until passage 50 at most. Seven gliomas (four low-grade gliomas, three anaplastic gliomas) showed a similar DNA distribution pattern with a main diploid and small tetraploid peaks at various passages. In this group, only one culture formed a permanent cell line, whereas six cultures showed a limited growth ranging from 6 to 24 passages. In contrast, the other four gliomas (each an anaplastic glioma) showed a marked change of DNA distribution through passages and finally a single DNA aneuploid population prospered. Each of these four gliomas yielded established cell lines. Thus, it is suggested that the change of DNA ploidy and prosperity of DNA aneuploid populations in flow cytometry might be used as early and reliable indices for the later establishment of glioma-derived permanent cell lines. Since the changes of DNA distribution are frequently associated with the morphological changes, as seen in the latter group, careful tracing of morphological features is valuable in determining of the fate of cultures, especially in the absence of a flow cytometer. The correlation between the potential to become established cell lines and histology of the original gliomas is also discussed.

A patient with a variant form of acute promyelocytic leukemia (M3 variant) associated with an increased number of basophils was found to present a reciprocal translocation, t(9;14)(q34;q22) in addition to t(15;17)(q22;q12). Although several cases of acute nonlymphocytic leukemia with increased bone marrow and peripheral blood basophils have been reported, this is the first case in which both the t(9;14) and basophilia were observed in a patient with M3. Our findings support the hypothesis that 9q34 may be associated with the chromosomal location of genes regulating the production and maturation of basophils.
1. The changes in FMRFamide (Phe-Met-Arg-Phe-NH2) immunoreactivity in response to incubation in dopamine, serotonin, met-enkephalin, oxytocin, arg-vasopressin and FMRFamide were examined in the central nervous system of the snail, Achatina fulica. 2. When the central nervous system was cultured in medium which contained dopamine and in medium which contained serotonin, the number of immunoreactive neurons increased in the anterior part of the cerebral ganglion and decreased in the sub-esophageal ganglion. 3. When arg-vasopressin was added to the culture medium, the number of immunoreactive neurons increased in the pedal ganglion and decreased in the other sub-esophageal ganglion. 4. By contrast, when the central nervous system was cultured in medium which contained oxytocin, the number of immunoreactive neurons did not increase, but rather decreased, in each ganglion. 5. No changes in immunoreactivity were detected in the central nervous system when it was cultured in medium which contained FMRFamide. 6. It appears, from these results, that the production and release of FMRFamide from different neurons are differentially affected by the physiologically active substances tested.
1. Coexistence of FMRFamide, met-enkephalin and serotonin immunoreactivities was examined in Achatina fulica and Aplysia kurodai. 2. Coexistence of FMRFamide and serotonin was found in some neurons of the visceral, right parietal and pedal ganglia of Achatina fulica, and in the pedal ganglion of Aplysia kurodai. 3. In Achatina fulica, coexistence of FMRFamide and met-enkephalin was found in a neuron of the left parietal ganglion and that of met-enkephalin and serotonin was found in a giant neuron of the right parietal ganglion. 4. Based on these results, the biological significance of coexistence was discussed.
1. A phylogenetic study of oxytocin (OXT)-like immunoreactive cells was performed by the PAP method in the central nervous system of invertebrates. 2. The immunoreactivity was detected in the nerve cells of Hydra magnipapillata of the Coelenterata; Neanthes japonica and Pheretima communissima of the Annelida; Oncidium verrucosum, Limax marginatus and Meretrix lamarckii of the Mollusca; and Baratha brassica of the Arthropoda. 3. No immunoreactive cells were found in Bipalium sp. of the Platyhelminthes; Pomacea canaliculata, Aplysia kurodai, Bradybaena similaris and Achatina fulica of the Mollusca; and Gnorimosphaeroma rayi, Procambarus clarkii, Hemigrapsus sanguineus, Helice tridens and Gryllus bimaculatus of the Arthropoda; Asterina pectinifera of the Echinodermata; and Halocynthia roretzi of the Protochordata. 4. These results demonstrate that an OXT-immunoreactive substance is widely present not only in vertebrates but also in invertebrates. 5. OXT seems to have been introduced into these invertebrates at an early stage of their phylogenetic history.
1. A phylogenetic study of arg-vasotocin (AVT)/arg-vasopressin (AVP)-like immunoreactive cells was performed by the PAP method in the central nervous system of invertebrates. 2. The immunoreactivity was detected in the nerve cells of Hydra magnipapillata of the Coelenterata; Neanthes japonica and Pheretima communissima of the Annelida; Pomacea canaliculata, Aplysia kurodai, Oncidium verrucosum, Bradybaena similaris, Achatina fulica, Limax marginatus and Meretrix lamarckii of the Mollusca; Gnorimosphaeroma rayi, Hemigrapsus sanguineus, Gryllus bimaculatus and Baratha brassicae of the Arthropoda; Asterina pectinifera of the Echinodermata; and Halocynthia roretzi of the Protochordata. 3. No immunoreactivity was detected in Bipalium sp. of the Platyhelminthes, or in Procambarus clarkii and Helice tridens of the Arthropoda. 4. From these results, it appears that AVT/AVP is a phylogenetically ancient peptide which is present in a wide variety of invertebrates. 5. The actions of AVT/AVP and its presence in invertebrates are discussed.
Rats eat kaolin after treatment with poisons or rotation. Thus, eating of nonnutritive substances such as kaolin, so-called pica, is an illness-response behavior of rats analogous to vomiting in humans. For use of rotation-induced pica as a behavioral index of motion sickness in rats we examined what kind of rotation was effective for inducing pica in rats and whether the vestibular apparatus was necessary for its induction. Rats ate much kaolin after double rotation with continuously changing centrifugal and angular accelerations, but little after single rotation with no accelerative changes. However, even double rotation failed to induce pica in bilaterally labyrinthectomized rats. Thus, rotation-induced pica in rats was induced in the same way as motion sickness in humans, suggesting that it resulted from motion sickness in rats. We conclude that pica can be used as a behavioral index of motion sickness in rats.
Pica, the eating of nonnutritive substances such as kaolin, can be induced by rotation in rats. We used this rotation-induced pica as a behavioral index of motion sickness in rats and examined whether diphenhydramine, methamphetamine and scopolamine, which are anti-motion sickness drugs for humans, are effective for reducing motion sickness in rats. Intraperitoneal injection of diphenhydramine or methamphetamine suppressed the rotation-induced kaolin intake of rats. Intraperitoneal injection of scopolamine had no effect on the rotation-induced kaolin intake, but its transdermal administration reduced this kaolin intake. These findings show that human anti-motion sickness drugs also prevent motion sickness in rats. Since the pharmacological mechanisms for preventing motion sickness in rats and humans are similar, we conclude that rats are a suitable animal model for use in studies on putative anti-motion sickness drugs.
