Seasonal variations of nasal resistance in allergic rhinitis and environmental pollen counts. III: Efficacy of antiallergic agents.

We previously reported that, in a patient with Japanese cedar pollinosis, preseasonal increase in total nasal airway resistance (NAR) possibly caused from circannual endogenous rhythm was suppressed by preseasonal systemic administration of antiallergic agent. In this study, in order to determine the influence of preseasonal use of a topical antiallergic agent and seasonal administration of a peroral antiallergic agent, we made measurement of nasal airway resistance and pollen counts under the previous respective conditions in separated years. Preseasonal high mean values of total NAR for a couple of weeks was not suppressed by the preseasonal topical administration of ketotifen nasal spray in 1992 which was a low pollen season (974/cm2). However, the patient did not complained of severe episodes of hay fever at all throughout the season. Preseasonal increases in mean values of total NAR were observed in 1993 which was a high Japanese cedar pollen season (4,875/cm2) with NAR the same as untreated seasons. This may have been because the patient was not treated with preseasonal use of peroral azelastine. On extremely high Japanese cedar pollen count days further increased NAR occurred in the patient. The patient experienced relatively severe hay fever episodes, but used no other anti-histamine nor topical steroid during the season.

A novel form of self tolerance dictated in the thymus of transgenic mice with autoreactive TCR alpha and beta chain genes.

Transgenic (TG) mice with TCR alpha and beta chain genes from a CD4-dependent auto-I-Ak reactive T cell clone were generated. H-2k TG mice had a large number of thymic and splenic CD4 T cells expressing the autoreactive TCR without manifestation of autoimmunity. The cells were not anergic, as they could respond to autologous antigen presenting cells and anti-TCR antibodies in vitro to proliferate and to produce interleukins. Various degrees of down-regulation of CD2 and CD44 was observed in TG mice, indicating the presence of a defective co-stimulatory process in TG T cells. These features indicate that the self tolerance in autoreactive TCR TG mice is due not to clonal deletion and anergy but to a novel mechanism where T cells cannot sufficiently respond to normally existing self ligand in vivo. That such an in vivo unresponsiveness of autoreactive T cells is dictated in the thymus during CD4 T cell differentiation atypical form of positive selection of autoreactive T cells was suggested by the abnormal surface expression of CD69 and HSA.

Increased insulin sensitivity in patients with aldosterone producing adenoma.

OBJECTIVE: Primary aldosteronism is a recognized endocrine cause of glucose intolerance. A blunted insulin response to glucose in patients with primary aldosteronism is well known, but insulin sensitivity has not been thoroughly determined. We investigated insulin sensitivity and insulin secretion in patients with aldosterone producing adenoma. DESIGN: Glucose tolerance and insulin responses to glucose were evaluated using a standard 75 g oral glucose tolerance test in patients with aldosterone producing adenoma before and after surgery, and in control subjects. Parameters for insulin resistance (HOMA-R) and pancreatic beta-cell function (HOMA-beta F) were derived from a homeostasis model assessment. PATIENTS: Fifteen patients with aldosterone producing adenoma and 41 control subjects with normal glucose tolerance matched for age and body mass index, were studied. Eight patients were re-evaluated after surgical removal of the adenoma. In 5 patients 125I-insulin binding to erythrocytes was also measured. MEASUREMENTS: Plasma glucose was measured by a glucose oxidase method. Plasma insulin, aldosterone and renin activity were measured by radioimmunoassay. RESULTS: Both fasting plasma glucose and insulin were lower in the patients than in the controls. Although the areas under the curve for plasma glucose during the oral glucose tolerance test did not differ, that for plasma insulin was less in the patients with hyperaldosteronism. Insulin sensitivity was increased in the patients, as evidenced by decreased HOMA-R compared with controls, while HOMA-beta F was comparable between the groups. 125I-insulin binding to erythrocytes was higher in 5 patients than in 19 normal controls. After surgical removal of adenoma, neither fasting plasma glucose nor the glucose area under the curve during the oral glucose tolerance test changed. However, fasting plasma insulin and the insulin area under the curve increased significantly. HOMA-R increased and HOMA-beta F remained unaltered. CONCLUSION: Insulin sensitivity was increased in untreated patients with aldosterone producing adenoma. Enhanced insulin receptor binding may contribute to this increased insulin sensitivity.

Differentiation between wild and vaccine-derived strains of poliovirus by stringent microplate hybridization of PCR products.

Procedures for differentiation between wild and vaccine-derived strains of poliovirus are required, particularly in countries where wild and vaccine-related strains coexist. For this differentiation, we tested the method of Inouye and Hondo (S. Inouye and R. Hondo, Arch. Virol. 129:311-316, 1993) for discrimination of closely related viruses by using stringent microplate hybridization of PCR products. We used a pair of primers with enterovirus common sequences (between these primers there is a variable region for capsid proteins) for PCR using templates from wild and vaccine-derived poliovirus strains which were isolated in tissue culture and serotyped by neutralization assay. We also used the same primers for preparation of probes, which were labelled by incorporation of biotin-dUTP in the PCR, with the three original Sabin vaccine virus strains used as templates. The amplified DNAs from the isolates were immobilized on microplate wells and were then hybridized with the labelled probes. We found that, under the usual hybridization conditions, the Sabin vaccine virus strain probes hybridized with both wild and vaccine-derived viruses, but under stringent conditions, they reacted only with vaccine-derived viruses of the same serotype, clearly differentiating these from wild-type viruses.

Molecular evolution of the major capsid protein VP1 of enterovirus 70.

Nucleotide sequences of the genome RNA encoding capsid protein VP1 (918 nucleotides) of 18 enterovirus 70 (EV70) isolates collected from various parts of the world in 1971 to 1981 were determined, and nucleotide substitutions among them were studied. The genetic distances between isolates were calculated by the pairwise comparison of nucleotide difference. Regression analysis of the genetic distances against time of isolation of the strains showed that the synonymous substitution rate was very high at 21.53 x 10(-3) substitution per nucleotide per year, while the nonsynonymous rate was extremely low at 0.32 x 10(-3) substitution per nucleotide per year. The rate estimated by the average value of synonymous and nonsynonymous substitutions (W.-H. Li, C.-C. Wu, and C.-C. Luo, Mol. Biol. Evol. 2:150-174, 1985) was 5.00 x 10(-3) substitution per nucleotide per year. Taking the average value of synonymous and nonsynonymous substitutions as genetic distances between isolates, the phylogenetic tree was inferred by the unweighted pairwise grouping method of arithmetic average and by the neighbor-joining method. The tree indicated that the virus had evolved from one focal place, and the time of emergence was estimated to be August 1967 +/- 15 months, 2 years before first recognition of the pandemic of acute hemorrhagic conjunctivitis. By superimposing every nucleotide substitution on the branches of the phylogenetic tree, we analyzed nucleotide substitution patterns of EV70 genome RNA. In synonymous substitutions, the proportion of transitions, i.e., C<==>U and G<==>A, was found to be extremely frequent in comparison with that reported on other viruses or pseudogenes. In addition, parallel substitutions (independent substitutions at the same nucleotide position on different branches, i.e., different isolates, of the tree) were frequently found in both synonymous and nonsynonymous substitutions. These frequent parallel substitutions and the low nonsynonymous substitution rate despite the very high synonymous substitution rate described above imply a strong restriction on nonsynonymous substitution sites of VP1, probably due to the requirement for maintaining the rigid icosahedral conformation of the virus.

A novel nonreceptor tyrosine kinase, Srm: cloning and targeted disruption.

We have isolated a novel nonreceptor tyrosine kinase, Srm, that maps to the distal end of chromosome 2. It has SH2, SH2', and SH3 domains and a tyrosine residue for autophosphorylation in the kinase domain but lacks an N-terminal glycine for myristylation and a C-terminal tyrosine which, when phosphorylated, suppresses kinase activity. These are structural features of the recently identified Tec family of nonreceptor tyrosine kinases. The Srm N-terminal unique domain, however, lacks the structural characteristics of the Tec family kinases, and the sequence similarity is highest to Src in the SH region. The expression of two transcripts is rather ubiquitous and changes according to tissue and developmental stage. Mutant mice were generated by gene targeting in embryonic stem cells but displayed no apparent phenotype as in mutant mice expressing Src family kinases. These results suggest that Srm constitutes a new family of nonreceptor tyrosine kinases that may be redundant in function.

Effects of vestibular stimulation on acetylcholine release from rat hippocampus: an in vivo microdialysis study.

1. The effects of electrical stimulation to the round window of the inner ear and caloric vestibular stimulation on the in vivo release of acetylcholine (ACh) from rat hippocampus were investigated, using brain microdialysis coupled with high performance liquid chromatography-electrochemical detection. 2. Hippocampal ACh release was increased to 152% of the basal release by 1-Hz, 200-ms, and 500-microA electrical stimulation to the round window. This effect was dependent on current intensity. 3. Microinjection of 6,7-dinitroquinoxaline-2,3-dione (DNQX) (10 nmol/0.2 microliter), an antagonist of the non-N-methyl-D-aspartate (non-NMDA) type of glutamate receptor, into the medial vestibular nucleus blocked the increase of hippocampal ACh release by electrical stimulation to the round window. In contrast, microinjection of DNQX (10 nmol/0.2 microliter) into the ventral cochlear nucleus did not inhibit the increase of hippocampal ACh release by electrical stimulation to the round window. 4. Electrical stimulation of the round window increased histamine release from the medial septum, which is the origin of the hippocampal cholinergic terminals. However, the depletion of neuronal histamine by alpha-fluoromethylhistidine (100 mg/kg ip) did not suppress the vestibular-evoked release of hippocampal ACh. 5. Although caloric stimulation with water at 37 degrees C did not affect the hippocampal ACh release, the release was increased to 131% of the basal release by caloric stimulation with hot water at 45 degrees C and was also increased to 160% of the basal release by caloric stimulation with ice water, suggesting that the changes in activities of vestibular afferents increased the hippocampal ACh release.(ABSTRACT TRUNCATED AT 400 WORDS)

A case of oncocytoma in the eyelid.

Oncocytoma rarely arises from the eyelid and only 2 cases have been reported previously. We present a case of oncocytoma in the upper eyelid around the lacrimal punctum. Anatomical location and histological findings suggested that oncocytes of this tumor originated from the epithelium of Moll's gland or the lacrimal canaliculus.

Electrophysiological studies on light damage in the mouse retina after sodium iodate injection.

Acute experimental retinal degeneration was induced in 3-month-old mice with sodium iodate (NaIO3) injection to investigate the effect of bright light with electroretinography (ERG). Eight C 57 black male mice were anesthetized. The ERG was recorded before and 24 h after the injection of NaIO3. Next, only one eye of each mouse was exposed to a bright light of 3.0 x 10(4) lx white light for 30 min. Ten hours later, ERGs of both the light-exposed and the unexposed eye were recorded. The amplitudes and peak latencies of the a wave and b wave were measured. The ERG was recorded as both eyes were exposed to stimulating flashes that were given in an increasing order of 0.6 log unit steps from the dimmest flash to produce a detectable ERG. The ERG from the light-exposed eye showed a distinctly elevated threshold (approx 2.0 log), while the contralateral unexposed eye did not. The peak latencies of both waves were significantly prolonged by exposure to light. In conclusion, light exposure affected ERG thresholds in mice with experimentally induced acute retinal degeneration.

Immunohistological studies of an oncocytoma.

An oncocytoma of the upper eyelid that occurred in an 82-year-old male was studied immunohistologically according to the avidin-biotin-peroxidase complex method (secretory component, carbohydrate antigen 19-9, carcinoembryonic antigen, anticytokeratin AE1/AE3, S-100 protein, myoglobin, actin, vimentin). The staining properties of columnar-type oncocytes were the same as those of epithelial cells of excretory ducts of Moll's gland, but those of polygonal-type oncocytes were similar to those of myoepithelial cells of Moll's gland. Our results suggested that excretory duct epithelial cells of Moll's gland are the origin of oncocytoma of the eyelid.

Sequencing and expression of sheep angiotensinogen cDNA.

A sheep liver cDNA library was screened with a BstEII fragment of human angiotensinogen cDNA to obtain sheep angiotensinogen cDNA. The cDNA sequence showed that preangiotensinogen consisted of 452 amino acid residues (49 kDa) of angiotensinogen and 24 residues of a signal peptide. Recombinant sheep angiotensinogen (56 kDa) was expressed in COS-7 cells.

The molecular cloning and sequence of an open reading frame encoding for non-structural proteins of feline calicivirus F4 strain isolated in Japan.

The nucleotide sequence of the 5'-end of feline calicivirus (FCV) Japanese F4 strain genome was determined. This region had 5311 bases and contained a large open reading frame (ORF1) encoding the non-structural proteins. The nucleotide sequence of the ORF1 region was highly conserved as compared with that of FCV F9 strain. When the deduced amino acid sequence of the ORF1 was compared with those of FCV F9 and CFI strains, the sequence was also highly conserved (88.9% and 88.8%, respectively). Functional motifs of the non-structural proteins were common to these strains. There were 2C polypeptide-, 3C cysteine protease- and 3D RNA-dependent RNA polymerase-like regions. The N-terminal region of 2C-like region continued upstream from the region identified by Neill [Virus Res. 17: 145-160]. Furthermore, the presence of 2B-like region was suggested in the upper stream of the 2C-like region, although the function of the region is unknown. When Kyte and Dolittle hydrophobicity profiles of the predicted amino acid sequences of the ORF1s of FCV F4 and F9 were computed and compared, both the profiles had striking similarities. In the region between residues 950-1000, there was a high rate of basic amino acid residues, suggesting that the polypeptide in this region of FCV may have a nucleic acid-binding function.

Strial dysfunction in a melanocyte deficient mutant rat (Ws/Ws rat).

A homozygous mutant rat at the white spotting (Ws) locus showing a deficiency of melanocytes has recently been found (4, 5). The function and morphology of the inner ear of the Ws/Ws rat were examined by auditory brainstem response (ABR), endocochlear DC potential (EP), and electron microscopy. The mean ABR threshold of the Ws/Ws rat was significantly higher than that of the control +/+ rat. Most Ws/Ws rats showed no or very little EP. In electron microscopy, the stria vascularis of the Ws/Ws rat proved to be very thin and flat with poor interdigitation of marginal cells, and absence of intermediate cells. The organ of Corti appeared to be intact in both the +/+ rat and the Ws/Ws rat. These electrophysiological and morphological findings suggest that the Ws/Ws rat suffered from severe hearing loss caused by strial dysfunction.

[Chlordane accumulations in adipose tissue of mice chronically exposed to chlordanes in indoor air].

Chlordane concentrations were analyzed in the adipose tissue of mice after prolonged inhalation of low levels of chlordanes in indoor air. After inhalation of 4.22-11.36 micrograms/m3 chlordanes (total of 5 compounds) in air for 1-6 months, high levels of chlordanes (4.19-11.63 ppm, total of 5 compounds and 2 metabolites) were found in the adipose tissue. Transnanochlor, which accounted for only 14% of the total chlordane in the inhaled air, showed a high percentage (50%) in the adipose tissue, followed by 2 metabolites, i.e., oxychlordane (25%) and heptachlorepoxide (22%). Little transchlordane, cis-chlordane, or heptachlor, which accounted for high percentages of the total chlordane inhaled, was detected in the adipose tissue. A low level of cis-nonachlor was observed in the adipose tissue, and its percentage was similar to that in the air (2%). The level of chlordane exposure and the adipose tissue chlordane concentration was closely correlated (r = 0.9077, p < 0.01), showed a dose-effect relationship. These findings suggest that chlordanes such as trans-nonachlor, oxychlordane, and heptachlorepoxide may accumulate in the adipose tissue of people living in an environment containing even low levels of chlordanes and that there is a risk from indoor air contamination by chlordanes.

Histological findings of capsular delamination of the lens.

Histological findings in a 75-year-old man with idiopathic capsular delamination were reported. The anterior capsules were obtained during extracapsular cataract extraction, and examined by light and electron microscope. The epithelial side of the anterior capsules protruded towards the anterior chamber at the edge of the delaminated areas. This finding has not been described previously, and the defect in this fragile region was thought to be the cause of capsular delamination.
