Immunohistochemical localization of myoepithelial cells and basement membrane in normal, benign and malignant human breast lesions.

Distributions of actin and type IV collagen were investigated immunohistochemically as markers for myoepithelial cells and basement membranes. Carnoy's and Methacarn-fixed, paraffin-embedded tissues from 103 human breast lesions from 103 patients were examined; 65 with carcinomas, 27 with mastopathies, 9 with fibroadenomas and 2 with phyllodes tumours. Fifty-five samples of the normal mammary gland tissue adjacent to tumours were also included for comparison. In normal breast and benign breast diseases, type IV collagen was identified around the mammary glandular cells and actin-positive cells were demonstrated to attach to basement membranes. In noninvasive carcinomas, type IV collagen was found as a continuous lining around a cell nest, while actin-positive cells were usually absent in ductal but quite numerous in lobular carcinomas. In invasive carcinomas, type IV collagen was fragmented or absent and actin-positive cells were very uncommon around the fragmentary basement membranes. These results suggest that the different distributions of myoepithelial cells and basement membrane material is useful in the differential diagnosis of surgical pathology of the breast.

Morphological and biological characteristics of mammary tumours induced by the direct application of DMBA powder to rat mammary glands.

Mammary tumours were induced by the direct dusting of 1 mg, 7,12-dimethylbenz(a)anthracene (DMBA) powder onto the mammary gland of both 30-day-old female and male Sprague-Dawley rats, and the tumours were examined histologically. Mammary tumours developed in 43/43 (100%) of the females 11 to 20 weeks after DMBA dusting and 16/23 (70%) of the males 18 to 28 weeks after dusting, while non-mammary spindle cell sarcomas occurred in 5/23 (22%) of the males 15 to 24 weeks after dusting. A variety of benign and malignant mammary tumours of epithelial and/or mesenchymal origin were induced, which are comparable to human mammary tumours. Different histological patterns were observed in different areas of the same tumours. Ovariectomy revealed hormone (ovary)-dependency in 10/17 (59%) of the tumours, revealing regressing epithelial and proliferating mesenchymal tumour elements on histological examination.

Immunoelectron microscopic localization of mammary tumor virus (MTV) antigens in normal and cancerous mammary glands of mice.

Peroxidase-labeled Fab' fragments of rabbit antisera against gp52 (major envelope protein) and A-particles of mammary tumor virus (MTV) were prepared and used for investigation by immunoelectron microscopy of the replication cycle of MTV-specific envelope and core antigens in normal and malignant mammary gland cells of female mice. The specificity of the antisera was proven by absorption tests and lack of reactivity to MTV-free mammary tissues. Periodate-lysine-paraformaldehyde (PLP) fixation sufficiently preserved the antigenicity of gp52, while Zamboni's fixative was useful to preserve the core antigen. Saponin pretreatment was necessary to reveal the intracellular antigen of A particles but had no influence on gp52. In addition to its presence at the envelope of D particles, gp52 was clearly associated with the biomembrane system, including the nuclear membrane, endoplasmic reticulum, Golgi apparatus and plasma membrane independent of the presence of virus particles. In mammary tumors, a significant level of gp52 antigen was often expressed on the entire cell surface membrane. In contrast, it was localized only to the apical plasma membrane in normal mammary gland cells. A particle antigens were confined to the intracytoplasmic A particles, usually visible as clusters, and to the inner part of B particles. These ultrastructural findings support the available biochemical data on the morphogenesis of MTV particles.

Systemic amyloidosis with a mesenteric mass which had the appearance of proteinaceous lymphadenopathy.

An autopsy case of systemic amyloidosis in a 46-year-old Japanese female who had M component in sera but no manifestations of myeloma is described. Autopsy revealed massive milky ascites and a large mesenteric mass, which showed histologically perivascular concentric deposition of eosinophilic materials with mixture of residual follicles and scattered plasma cells. These morphological findings closely resembled those seen in proteinaceous lymphadenopathy, and the material in the mesenteric mass proved to be an amyloid A lambda protein. Subsequent immunohistochemical observations strongly suggested that this mass lesion was a "burnt-out" plasmacytic dyscrasia.

Immunohistochemical expression of mammary tumor virus antigens in mammary glands of virgin mice, in relation to Mtv genes.

MTV antigens in the resting mammary glands of GRS/A, SHN, C3H, and Balb/c virgin mice were detected by immunoperoxidase techniques using antiserum against MTV whole virion, gp 52 or p 25 to differentiate the expression between endogenous and exogenous MTV. Balb/c mice were crossed to infect MTV into each reciprocal hybrid or fosternursed inbred. Immunohistochemical stainings of gp 52 in the formol-fixed sections were almost the same as those of the whole MTV virion, and the results on various cases were as follows: In the mice with endogenous GR-MTV, positivity was first observed at the age of 14 days, while the first expression of exogenous GR-MTV was delayed to the age of 140 days. The mice with endogenous and/or exogenous SHN-MTV showed the first antigen appearance at the age of 65 days, and those with exogenous C3H-MTV did at the age of 80 days. The virgins with only endogenous C3H-MTV came to express the antigen after the age of 200 days. Staining of p 25 in Carnoy-fixed sections of MTV-positive mammary glands was found in the supranuclear cytoplasm and apical surface of the glandular cells and the lumen, all of which are the site of A and B particles. By means of preembedding method for gp 52, the reaction products were ultrastructurally detected not only on the MTV-budding apical surface, together with the intraluminal B particles, but also on the MTV-free apical cell membrane of the glandular cells in the mammary gland of the GR virgins.

[An operative case of spermatic cord rhabdomyosarcoma--with a review of similar cases in Japan].

A 17-year-old Japanese boy was admitted with the complaint of painless, progressive enlargement of the left scrotal contents during the preceding two months. The tumor was firm and nontender without transillumination. Orchitectomy with high ligatation of the spermatic cord was performed. The tumor, weighing 13.1 g, was gray, solid, located at the distal part of the spermatic cord, and separated from the testis and epididymis. The pathological diagnosis showed left spermatic cord rhabdomyosarcoma of the combined embryonal and alveolar type. All of the retroperitoneal lymph nodes dissected were negative for metastasis. The patient was treated with actinomycin D and vincristin postoperatively, and is well three years after the operation. Ninety-two cases of intrascrotal rhabdomyosarcoma reported in Japan are reviewed.

Fibroma of the vulva.

We describe a patient with fibroma of the vulva. The tumor had areas of marked hypointensity consistent with fibrosis on T1 and T2 weighted magnetic resonance (MR) images. The presence of abundant fibrous tissues on MR images enabled us to make a preoperative diagnosis of fibroma.

Immunohistochemical study of cytokeratins in hidradenitis suppurativa (acne inversa).

In 14 cases of hidradenitis suppurativa, cytokeratin (CK) expression was studied immunohistochemically, using six antikeratin antibodies against CK1, CK10, CK14, CK16, CK17 and CK19, respectively. The draining sinus tract epithelium of hidradenitis suppurativa lesions was divided into three components: infundibular-like keratinized epithelium (type A), non-infundibular keratinized epithelium (type B), and non-keratinized epithelium (type C). In type A samples, CK17 (which is found in normal infundibulum) was not detected, suggesting fragility of this epithelial type. Keratin expression in types B and C epithelia was similar to that observed in the outer root sheath in normal hair follicles. Our results suggest that the draining sinus epithelium may possess characteristics of fragility, undifferentiation and hyperproliferation, as shown with CK expression.

Correlation between microvessel density and tumor cell proliferation with clinical factors in breast carcinomas.

Fifty-nine methacarn-fixed, paraffin-embedded human breast carcinomas were immunostained by QB-END/10 (an antibody to CD34 antigen) to observe microvessels and by PC10 (an antibody to proliferating cell nuclear antigen; PCNA) to determine tumor cell proliferation; 9 normal human breast tissue specimens were also immunostained by QB-END/10. The number of microvessels in the periphery of the breast carcinoma was significantly greater than both that in the center of the breast carcinoma and that in the normal breast. There was also a significant relationship between the number of microvessels in the periphery of breast carcinomas and the histological tumor size and lymph node status. However, there was no significant relationship between the tumor cell proliferation activity (PCNA positive cell ratio) and any clinical or histopathological variables. The number of microvessels and the tumor cell proliferation activity showed a weak negative correlation.

Immunohistochemical study of mammary and extramammary Paget's disease.

Immunophenotypes of mammary (MPD) and extramammary Paget's disease (EPD) are still not well understood. Thirty-four formalin-fixed paraffin-embedded tissue sections from 33 patients with 6 MPD and 28 EPD were studied immunohistochemically with the use of polyclonal c-erbB-2 and pS2 antisera, and monoclonal nm23, B6.2, GCDFP-15, and p53 antibodies. Cases of MPD expressed a high incidence of c-erbB-2 and nm23 compared with those of EPD (100% vs. 29%; p < 0.01, and 83% vs. 29%; p < 0.05, respectively). Although high expression of B6.2 (> 83%) and moderate expression of GCDFP-15 (33-39%), pS2 (33-46%) and p53 (39-50%) were seen, the positivity was not significantly different between MPD and EPD. These findings indicate that MPD and EPD share immunohistochemical features but partially differ in their patterns of antigen expression.

Transcatheter arterial embolization with cisplatin: apoptosis in VX2 tumour uterus transplants.

Effects of chemoembolization with cisplatin on gynecological malignancies were investigated using rabbit uterine tumour. 4 weeks after inoculation, the 35 rabbits were divided into 3 groups. In the experimental groups, the tumour tissue specimens were stained with hematoxylin and eosin, ApopTag stain and proliferating cell nuclear antigen (PCNA) dye. In two days the Pt level of tumour tissue in the transcatheter arterial chemoembolization (TACE) group was 1.97 times more than that in the intra-arterial (IA) infusion group (p < 0.001). Apoptotic index was 1.03% in the IA group versus 5.9% in the TACE (p < 0.001). The PCNA index was 86.6% in the IA group, compared with 8.6% in the TACE group (p < 0.001). Experiments reported here have revealed enhanced effects of the simultaneous action of nutrient restriction and increase concentration of drug caused by the vascular occlusion on cell death.
