Modulation of Fc-mediated phagocytosis by cyclic AMP and insulin in a macrophage-like cell line.

Studies on genetically selected mutants in phagocytosis of E[IgG] indicated that the defect in some mutants could be corrected by addition of 8 Br-cAMP, and suggested that cyclic AMP may be involved in the mechanism of phagocytosis through the Fc receptor. In order to elucidate the role of cyclic AMP in phagocytosis in the parental, nonmutant macrophage-like cell line, J774.2, it was necessary to employ restrictive conditions which rendered phagocytosis suboptimal. When 4774.2 cells were cultured in nontissue culture Petri dishes phagocytosis was markedly reduced. Addition of 8 Br-cAMP or inducers of intracellular cyclic AMP such as isoproterenol restored the phagocytic ability of these cells. Similarly, treatment of the parental J774.2 cells with insulin reduced the level of phagocytosis, and once again this suppression could be corrected by addition of 8 Br-cAMP. In no case did AMP mimic the effects of 8 Br-cAMP. The effect of cyclic AMP action in this system was not instantaneous, but rather reached optimal levels at 5 to 10 hr, suggesting that cyclic AMP is not the immediate signal for phagocytosis. The genetic analysis of macrophage variants may provide a useful model for studies on the mechanisms of phagocytosis, and also the effects of insulin and cyclic AMP on an easily measurable biologic function in a specialized cell type.

Miochol-induced transient cataract.

Acute transient lens opacity occurred in a patient with ruptured cornea following injection of Miochol into the anterior chamber. It is postulated that the lens changes were due to osmotic effect of the hypertonic Miochol solution.

Congenital bronchiolitis obliterans.

A case is described of intrauterine pneumonia with "Congenital Bronchiolitis Obliterans" in a premature infant who died 35 hours after delivery. The patient presented from birth with increasingly severe respiratory insufficiency. Post-mortem histological examination of the lung revealed obliterative bronchiolitis, a rare disease which the patient most probably developed in utero. Only two other similar cases of congenital obliterative bronchiolitis have been reported in the literature, both with obscure etiology. A detailed pathological description of pulmonary changes is given and the literature is reviewed.

Ulnar nerve palsy following fracture of the shaft of the humerus.

We present an unusual case of ulnar nerve palsy associated with humeral shaft fracture. Initial examination showed swelling and tenderness. No mention of neurological deficit was found in the patient's file. Radiography showed a displaced midshaft fracture. Closed reduction and intramedullary nailing were performed. Ulnar nerve palsy was noted by the attending physician afterward, and, after four months of nonoperative treatment without any obvious improvement, the patient was referred to our center. During surgery, the nerve was found to be transected and an end-to-end repair was performed. Five months after surgery, signs of motor recovery were present. This case demonstrates that ulnar nerve injury can occur with a closed fracture of the humeral shaft.

Orbital lipogranuloma after sinus surgery.

PURPOSE: To report the treatment and histopathological findings in two cases who developed eyelid swelling, proptosis and diplopia due to orbital and lid lipogranuloma after endoscopic surgery of the maxillary and ethmoidal sinuses. METHODS: To relieve the proptosis and diplopia, debulking surgery was done on the eyelids and orbit. The tissue removed was sent for histopathological examination. RESULTS: The two patients improved after surgery. The eyelid swelling, proptosis and diplopia subsided and ocular movements became normal. Histopathologic examination disclosed an extensive lipogranuloma. CONCLUSIONS: Extensive orbital and eyelid lipogranuloma causing proptosis and diplopia is a rare complication of endoscopic sinus surgery, and can be relieved by surgical debulking.

Expression of the lck tyrosine kinase gene in human colon carcinoma and other non-lymphoid human tumor cell lines.

The tyrosine kinase p56lck is felt to be expressed solely in cells and tissues of lymphoid lineage. We have observed in the particulate fractions of colon carcinoma cells the presence of a 56 kilodalton (kD) alkali resistant phosphoprotein containing phosphotyrosine. Further analysis revealed that the partial proteolytic cleavage pattern of this protein is analogous to that of the lck gene product found in murine and human T-cells. We have confirmed by cDNA cloning that lck RNA is also expressed in these cells proportionally to the amount of p56lck found in membrane preparations. Additional analysis demonstrated that lck RNA is frequently expressed in certain non-lymphoid tumor cell lines such as cell lines derived from colon carcinomas and carcinomas of the lung. Interestingly, higher levels of lck expression were found in human tumor cell lines derived from metastatic tumor sites when compared to those derived from primary tumors. These observations raise the possibility that the lck gene product may be involved in progression of some non-lymphoid human tumors.
